775 Relationship between SECTM1 expression and clinicopathological features of colorectal cancer and itseffect on cell function

Objective: To investigate the expression between the expression of secreted and transmembrane1 (SECTM1) in colorectal cancer (CRC) and clinicopathological features as well as its influence on cell function.Methods: The expression of SECTM1 in CRC and matched adjacent tissue samples of 370 CRC patients from from the Second Affiliated Hospital of Wenzhou Medical University between 2011 and 2017 was evaluated by immunohistochemical method. The HCT116 cell line and DLD-1 cell line transfected with a SECTM1-overexpression lentivirus or SECTM1-siRNA, respectively, were further used to examine the effect of SECTM1 on cell proliferation, apoptosis, cell cycling, and invasion. Results: Immunohistochemical staining showed thatSECTM1 was over-expressed in CRC tissue samples compared with paired adjacent tissue samples. SECTM1 was associated with well-or-poorly differentiated adenocarcinoma classification (P<0.001) and lymph node metastasis (P=0.001), though SECTM1 expression was independent of age (P=0.259), sex (P=0.768), tumour location (P=0.299), tumour size (P=0.458), UICC staging (P=0.105) and CEA level (P=0.202). Furthermore, in vitro experiments showed that in HCT116 cell line, the up-regulation of SECTM1 expression promoted cell proliferation, reduced
apoptosis, accelerated the process of cell cycle and increased cell invasion (P<0.05). In theDLD-1 cell line, its down-regulation led to the opposite result (P<0.05).
Conclusion: SECTM1, a new potential carcinogen, is highly expressed in CRC tissue, which can promote cell proliferation, reduce apoptosis, accelerate cell cycle
process, and increase cell invasion.

784 Transcriptome characteristics analysis and Hub genes prediction of respiratory syncytial virus subtypeA bronchiolitis based on RNA sequencing

Objective: To screen potential key genes (Hub genes) and pathways involved in the pathogenesisof bronchiolitis in children caused by respiratory syncytial virus (RSV) subtype A using RNA sequencing data analysis, aiming to provide scientific clues for intervention and treatment of RSV bronchiolitis. Methods: Twentythree patients with RSV bronchiolitis (case group) and ten healthy children (healthy control) were enrolled.RNA was extracted from peripheral blood leukocytes for high-throughput RNA sequencing. The differentially expressed genes (DEGs) between case and control were obtained by three R packages including DESeq2, edgeR and limma. All DEGs were selected for GO and KEGG enrichment analysis by “clusterProfiler” package and Metascape tool, respectively. Protein-protein interaction (PPI) analysis for all DEGs was constructed using STRING database, and finally Hub genes were identified in Cytoscape software. Results: We screened a total of 286 DEGs between case and control, including 166 upregulated genes and 120 downregulated genes. GOenrichment analysis showed that these DEGs were mainly enriched in several biological processes including hydrogen peroxide catabolic process, cellular response to biotic stimulus and vitamin D receptor signaling pathway. KEGG pathway analysis showed that these DEGs were mainly involved in several pathways related to immune response, such as cytokine signaling in immune system, adaptive immune system and neutrophildegranulation. Using the MCODE plug-in of Cytoscape, we determined four core modules from entire PPI network, mainly associated with viral protein interaction with cytokine and cytokine receptor, and RAF/MAP kinase cascade. Finally, we screened a total of 11 Hub genes related to RSV subtype A bronchiolitis, including RRM2, BUB1B, BUB1, MCM10, CDC45, MKI67, ASPM, NCAPG, NUSAP1, ESPL1 and CDT1. Conclusion:These immune characteristics and hub genes may be correlated with the pathogenesis of RSV bronchiolitis in children, and the specific pathogenic mechanism deserves further investigation.

792 Intervention effect of executive function training on school-age children with attention deficit hyperactivity disorder

Objective: To explore the effectiveness of executive function training on school-age children with attention deficit hyperactivity disorder (ADHD), so as to provide a new perspective for the treatment of children with ADHD. Methods: A total of 56 school-age children diagnosed as ADHD in Wenzhou People’s Hospital between September 2019 and October 2020 were randomized into executive function training group (n=27) and parent training group (n=29) for 12 weeks of respective executive function training and parent training, and then intervention effect was compared between the two groups. Multidimensional attention test, SNAP-IV Rating Scale, and Conner’s Child Behavior Scale were used for evaluation before and after each intervention and 1 year after the interventions as well. Results: After 12 weeks of interventions, there was statistical difference in scores of attention test, SNAP-IV Rating Scale, and Conner’s Child Behavior Scale in the executive function training group, as
compared with those before intervention and those in parent training group after intervention (P<0.05). The 1-year follow-up after intervention showed that the executive function training group failed to maintain completely stable curative effect, and there was significant difference in all scores, as compared with those before intervention and in the parent training group 1 year after intervention (P<0.05). However, the scores of attention test decreased as compared with those 12 weeks after intervention, and the scores of attention deficit in SNAPIV rating scale and the scores of learning problem and hyperactivity index in Conner’s Child Behavior Scale increased as compared with those 12 weeks after intervention, which showed statistical difference (P<0.05). Incontrast, there was no significant difference in the scores of
hyperactivity/impulsivity, oppositional defiance, conduct problem, psychosomatic disorder, impulsivity-hyperactivity and anxiety in the scales, as compared with
those 12 weeks after intervention (P>0.05). Conclusion: Executive function training can effectively improve thecore symptoms and behavioral problems in ADHD children, and long-term outcomes as well.

800 The effect of miRNA-195 inhibitor on the biological function of high-glucose cultured vascular endothelialcells and its mechanism

Objective: To explore the effect of miRNA-195 inhibitor on the biological function of vascularendothelial cells cultured by high glucose and its mechanism. Methods: Human umbilical vein endothelial cell line (HUVECs) ECV304 was culutred and treated with high glucose, miRNA-195 mimic, miRNA-195 inhibitor and Sirt1 agonist resveratrol. CCK-8 kit was used to detect proliferation activity, the levels of miRNA-195 and Sirt1 mRNA detected by RT-PCR, the protein expression levels of Sirt1, p-FoxO1, FoxO1, Caveolin-1, vascular endothelial growth factor (VEGF) detected by Western blot. The scratch test was used to detect the migration ability of cells,
transwell chamber test was used to detect the invasion ability of cells. Matrigel glue was used to detect the ability of vascular endothelial cells to form tubes. Results: The proliferation, migration and tube formation of vascular endothelial cells were enhanced under high glucose conditions (P<0.05); while miRNA- 195 inhibitor
significantly inhibited the proliferation of vascular endothelial cells and reduced the ability of tubeformation (P<0.05). Conclusion: MiRNA-195 inhibitor may affect the biological function of endothelial cellsthrough miRNA-195/Sirt1/FoxO1/Caveolin-1/VEGF pathway under high glucose conditions.

808 The biological function of fucosyltransferase 2 in prostate cancer cell

Objective: To investigate the effect of fucosyltransferase II (FUT2) on the biological function of prostate cancer cells and its potential mechanism. Methods: The
expression of FUT2 in prostate cancer tissueswas detected by immunohistochemistry. FUT2 knockdown prostate cancer cell line (PC-3) and empty control cell line were constructed by liposome transfection technology. The proliferation of PC-3 cells was detected by MTT and plate cloning formation assay. The migration and invasion of PC-3 cells was detected by transwell assay.Western blot was used to detect the expression levels of FUT2, β-catenin, Cyclin D1 and ICAM-1. Results:
Immunohistochemical analysis showed that FUT2 expression was significantly increased in prostate cancer tissues (P<0.05). Knockdown FUT2 inhibited the proliferation, migration and invasion of prostate cancer cells; meanwhile, Cyclin D1 expression was decreased and ICAM-1 expression was increased (P<0.05). Conclusion:FUT2 can promote the proliferation, migration and invasion of prostate cancer cells.

813 Application value of dual-energy CT virtual non-contrast scan technique in the evaluation of adrenal tumors

Objective: To explore the application of dual-energy CT virtual non-contrast (VNC) technology in the diagnosis of adrenal tumors. Methods: Fifty-six patients with adrenal tumors confirmed by postoperative histopathology in the Fifth Affiliated Hospital of Wenzhou Medical University from January 2020 to September 2021 were retrospectively collected. All patients underwent true non-contrast (TNC) and dual-energy CT enhanced three-phase (arterial, venous, and delayed) scans before surgery, and then obtained VNC images of different phases by post-processing software (VNCA, VNCV, and VNCD). The image quality of TNC and VNC was evaluated both subjectively (displaying the number of tumor morphological features and image quality score) and bjectively (measurement of CT value, noise, SNR, and CNR of the lesion), with the radiation was recorded. The one-way analysis of variance method was used to compare the differences in the objective evaluation of TNC and VNC at different phases. The Kruskal-Wallis test was used to compare the differences in the subjective evaluation between TNC and VNC at different phases. The independent sample t-test was used to compare the difference in radiation dose values between VNC mode and conventional mode. Results: There were no significant differences in tumor morphological features and image quality scores between TNC and VNC at different stages (all P>0.05). The CT values of lesions and normal adrenal parenchyma on TNC images were significantly lower than those onVNCV and VNCD images (all P<0.05), but showed no statistical difference compared with VNCA images (P=0.091, \0.085). Compared with TNC, the SNR and CNR of VNCA, VNCV, and VNCD images were significantly higher (all P<0.05). The lesion noise of VNCA, VNCV, and VNCD images was significantly lower than that of TNC (all P<0.05). In addition, there was no statistical significance in CT value, SD value, SNR, and CNR of lesions in the three groups of VNC images (all P>0.05). In terms of radiation dose value, the effective radiation dose (ED) of the VNC mode was reduced by 23.2% compared with the conventional mode. Conclusion: The VNC images obtained by applying dual-energy CT arterial phase scan data can significantly reduce the radiation dose received by patients with adrenal tumors, which meets the diagnostic requirement.

818 Application of contrast-enhanced ultrasound LI-RADS in the diagnosis of hepatocellular carcinoma in patients with occult hepatitis HBV infection

Objective: To investigate the diagnostic performance of contrast-enhanced ultrasound (CEUS) LI-RADS algorithm in the diagnosis of hepatocellular carcinoma (HCC) in patients with occult hepatitis B infection (OBI). Methods: A retrospective analysis was performed on 334 newly diagnosed hepatic focal lesions in OBI patients in Lishui Central Hospital from January 2018 to December 2021. Each nodule was assessed according to CEUS LI-RADS, and subgroup analysis was performed in patients with or without alpha-fetoprotein (AFP) >25 μg/L. The diagnostic performance of CEUS LI-RADS system for liver cancer was verified by sensitivity, specificity, precision, positive predictive value (PPV) and negative predictive value (NPV). Results: There were 120 HCC cases (36.0%), of which 4 (3.3%) were classified as LR-4, 7 (58.3%) as LR-5 and 46(38.3%) as LR-M. The sensitivity, specificity, precision, PPV and NPV of LR-5 class for predicting liver cancer was 58.3%, 88.8%, 77.8%, 74.5% and
79.2%, respectively. The sensitivity, specificity, precision, PPV and NPV of the LI-RADS diagnostic algorithm in OBI patients with AFP>25 μg/L was 63.3%, 80.0%, 65.7%, 95.0%and 26.7%, respectively. Given its high PPV, 13.0% (40/308) of OBI patients were highly suspected of HCC by CEUS. Conclusion: In OBI patients withFP>25 μg/L, using CEUS LR-5 as the diagnostic criteriion can help to confirm the diagnosis of HCC.

823 The characteristics of gene mutations associated with cross-resistance to aminoglycosides and peptides in MDR-TB

Objective: To analyze the association between the cross-resistance of streptomycin (SM),kanamycin (KAN) and capreomycin (CAP) and mutations of genes in muti-drug resistant tuberculosis (MDRTB)isolates. Methods: 125 clinical isolates of MDR-TB were revived, drug susceptibility testing to SM, KANand CAP was performed using proportion method, and PCR and sequencing were conducted for the four selected genes (rrs, tlyA, eis promoter, rpsL). Results: Of 125 MDR-TB strains, 7.2% (9/125) were resistant to SM+ KAN, 3.2% (4/125) resistant to SM+CAP, 2% (2/125) resistant to KAN+CAP, and 4.8% (6/125) were all resistant to SM+KAN+CAP. Sequencing showed that the 33A-G mutation of tlyA gene was found in all 125 MDR-TB strains. Four strains of eis promoter mutation were found, all of which were
found in KAN resistant-relatedstrains. A total of 14 mutations, with 8 types of mutation, were found in the rrs gene mutation, of which the 1401A-G type mutation was found in 5 strains, and the mutation in 530 loop region was found in 3 strains. The sensitivity to SM+KAN resistance detection by rrs gene was 33.33%, the sensitivity to SM+CAP resistance detection was 40%, the sensitivity to KAN+CAP resistance detection was 50%, and the sensitivity to SM+ KAN+CAP resistance detection was 50%. Conclusion: Cross-resistance was found between SM, KAN and CAP.The 530 loop region mutations and 1401A-G mutation in rrs of MTB were associated with the cross-resistance toSM, KAN and CAP. The detection of SM, KAN and CAP cross-resistance by rrs mutation is of significance for reference.

829 Curative effect of disposable electronic ureteroscope in the treatment of lower calyceal stones

Objective: To investigate the efficacy of a home-made disposable digital flexible ureteroscope compared with fiberoptic flexible ureteroscope in the treatment of lower calyceal stones ≤2 cm. Methods: A total of 80 patients with stones ≤2 cm in the lower calyx of the kidney from Wuxing District People’s Hospital in Huzhou from April 2020 to June 2021 were retrospectively analyzed. They were divided into disposable digital flexible ureteroscope group (DDFU group, 38 patients) and fiberoptic flexible ureteroscope group (FFU group, 42 patients). The preoperative, intraoperative, and postoperative data as well as the 3-month postoperative followup data were compared between the 2 groups, and the performance indexes of the 2 ureteroscopes were also compared. Results: There were no differences between the 2 groups in intraoperative stone seeking rate, duration of hospital stay, postoperative fever (P>0.05), but statistical differences were observed in operative time,occurrence of postoperative hemorrhage/hematoma, and stone clearance (P<0.05). Conclusion: The operating performance of disposable digital flexible scopes is better than that of fiberoptic flexible ureteroscopes. In the treatment of lower calyceal stones, the disposable digital flexible ureteroscope is safer and more effective with shorter procedure time, less bleeding and higher stone clearance rate.

833 The infection status of streptococcus agalactiae and antimicrobial resistance in 43034 pregnant women in the third trimester of pregnancy

Objective: To analyze the infection status and antibiotic resistant of Group B streptococcus (GBS) in the third trimester of pregnancy women in Wenzhou, and to compare the infection status of pregnant women between different age groups. Methods: Data were collected from 43 034 pregnant women in third trimester of pregnancy (36-42 gestational week) treated in the Obstetrics Department of the Second Affiliated Hospital of Wenzhou Medical University from January 2016 to December 2020. The statistical analysis was conducted on GBS culture and antibiotic sensitivity test of their urogenital tract, accompanied with the comparison in the detection of GBS in the third trimester of pregnancy of different ages. Results: Of 43 034 pregnant women,2 928 cases of GBS lactis were detected from the urogenital tract samples with a detection rate of 6.80%. The detection rate displayed an upward trend from 2016 to 2019, as was the highest in 2019, showing statistical difference (P<0.05); the detection rate had a slight decrease in 2020, but exhibited no significant difference. The detection rate was the highest in the pregnant women over 40 years old (7.92%), with statistical significance (P<0.05). In the drug sensitivity test of pregnant women with GBS infection, the sensitivity rate to vancomycin was found to be 100.00%, and the highest antibiotic resistance was erythromycin (74.11%), followed by clindamycin (72.81%), levofloxacin (37.06%), with significant difference among the three drugs (P<0.05).Conclusion: The detection rate of GBS in the third trimester of pregnancy women in Wenzhou was relatively high, reaching about 6.80%, highest in the pregnant women over 40 years. For pregnant women with GBS infection, penicillin is still the preferred prophylactic drug during labor. For women with penicillin allergy, antibiotics should be reasonably used according to the results of drug sensitivity test.

837 Application of disaster vulnerability analysis based on Kaiser model in obstetric nursing

Objective: To evaluate obstetric nursing risk events by using disaster vulnerability analysis,identify the weak links in obstetric nursing management and take preventive measures to improve the risk prevention ability of nursing staff and ensure nursing safety. Methods: The disaster vulnerability analysis based on Kaiser model was used to investigate the obstetric risk index for value calculation and the matrix analysis was made to identify potential high-risk events. Results: Among 29 indicators, the top 5 risk values included inadequate implementation of delivery safety checklist (48.07%), weak legal awareness of nursing staff (41.71%), low compliance and accuracy of hand hygiene (38.52%), inadequate prevention of postpartum hemorrhage (37.82%), and poor five-minute cesarean delivery process (37.15%); The matrix analysis of the risk assessment indicators showed 6 items in the high risk zone, 21 items in the medium risk zone, and 2 items in the low risk zone. Conclusion: The disaster vulnerability analysis can identify the obstetric high-risk events, improve the emergency response of nursing staff through emergency drills, and lead nurses to take corresponding preventive measures to avoid the occurrence of adverse events.

850 The innovative approach to the construction of special discipline zone in Chinese universities and its practical reference

The construction of special discipline zone is a new way for Chinese universities to explore the discipline development and promote the construction of “double first-class” in recent years. Although discipline special zones and economic special zones belong to different sectors, they are similar in developmental thinking and constructional essence. The successful experience in the construction of special economic zones is of important value and feasibility in guiding the construction of discipline special zones from two aspects: institutional model and practical model. Based on transition economics theory, this paper attempts to take the construction of special economic zones as a comparison and discuss the construction path of discipline special zones in terms of object selection and path development.

856 Influencing factors of professional master’s degree student-tutor relationship from the perspective of formal organization theory

Objective: To explore the influencing factors of relationship between professional master’s degree postgraduate and supervisor. Methods: Based on the formal organization theory, a questionnaire was designed to investigate the influencing factors of the student-supervisor relationship, and an anonymous survey by random sampling was conducted among 413 professional master’s degree postgraduates in Wenzhou,Hangzhou and other universities. A structural equation model was constructed to analyze the relationship among four latent variables, i.e. common goals, willingness to cooperate, communication and the degree of harmony in the student-supervisor relationship through 22 observation variables. Results: The respondents’ average scores of the dimensions in harmony, common goals, willingness to collaborate, and communication were all slightly higher than the median value of 3, indicating there was room for improvement. Among them, whether the university or the discipline is “double first-class” has no statistical significance in each dimension of the mentor relationship (P>0.05). However, the indicators of the common goal, collaboration willingness and communication in the student-supervisor relationship are higher in the fresh graduate group than in the non-fresh graduate group, showing statistical difference (P<0.05). The path analysis results of the structural equation model showed that common goals (β=0.29, P<0.01), willingness to cooperate (β=0.38, P<0.01) and communication (β=0.32,P<0.01) all have a significant impact on the harmony degree of the student-supervisor relationship. By comparing the standardized path coefficients of the model, it is found that the willingness to cooperate (β=0.38, P<0.01) has the most significant impact on the harmony degree of the student-supervisor relationship, followed by communication (β=0.32, P<0.01), common goals (β=0.29, P<0.01); the coefficient of observation variable shows that the student’s own willingness to cooperate, the academic exchanges of the mentors, and the support of the mentors have more obvious effect on the student-supervisor relationship. Conclusion: Both students and supervisors should clarify common goals and form a high level of team consensus, enhance their willingness to collaborate and create a new type of cooperative community, increase the communication frequency and build a multidimensional interactive platform.
