Expression and purification of rhFGF4 in Escherichia coli and its protective effect on human umbilical vein endothelial cells damaged by high glucose

Objective: To prepare and identify recombinant human fibroblast growth factor 4 (rhFGF4) protein and observe the protective effect of rhFGF4 on human umbilical vein endothelial cells (HUVEC) model with high glucose damage. Methods: The recombinant plasmid pET3d-hFGF4 was constructed, and the rhFGF4 protein was efficiently expressed by E.coli expression system. Most soluble objective proteins are expressed in the form of inclusion, including the cleaning, degeneration and restoration and purification of heparin column.The MTT method was used to detect the biological activity of rhFGF4 protein in promoting cell proliferation,the cell damage model was established and the phosphorylation expression of NF-κB, JNK and p38 was detected by Western blot technology. Results: The recombinant plasmid pET3D-HFGF4 was successfully constructed.SDS-PAGE electrophoresis and Western blot identification results showed that rhFGF4 protein was successfully expressed in E.coli. It was also confirmed that rhFGF4 protein repaired HUVEC cells damaged by high glucose and inhibited their apoptosis by preventing the phosphorylation of NF-κB, JNK and p38, and reduced their MDA and ROS content. Conclusion: This study has successfully obtained high purity and biologically active rhFGF4 protein, which provides a useful tool for further research on the role of this protein in vascular diseases caused by high glucose and related mechanisms.

The relationship of 12 candidate single nucleotide polymorphisms with susceptibility and prognosis of acute lymphoblastic leukemia in Chinese children

Objective: To investigate the relationship of 12 candidate single nucleotide polymorphisms(SNP) loci with the susceptibility and prognosis of Chinese children with acute lymphoblastic leukemia (ALL).Methods: Review the literature and select 12 candidate SNP loci, and then detect the genotype of each SNP by multiplex PCR targeted capture next-generation sequencing technology. A case-control study on 556 children with ALL and 191 healthy volunteers was conducted using chi-square test and logistic regression analysis to verify the relationship between SNP loci and the susceptibility of Chinese children with ALL. Kaplan-Meier survival analysis and Cox regression were used to analyze the relationship between SNP loci and the prognosis of ALL in 365 ALL cases. Results: Logistic regression analysis showed that the genotype of the SNP locus was significantly correlated with the increased risk of ALL: the rs10821936 risk allele C (OR=1.687, 95%CI=1.318-2.135, P<0.001) and rs10994982 risk allele A (OR=1.594, 95%CI=1.259-2.018, P<0.001) of the ARID5B gene;rs11978267 risk allele G (OR=2.133, 95%CI=1.495-3.043, P<0.001) and rs4132601 risk allele G (OR=2.108,95%CI=1.466-3.031, P<0.001) of the IKZF1 gene and the rs7554607 risk allele C (OR=1.481, 95%CI=1.161-1.890, P=0.002) of the ZNF230 gene. Multivariate Cox regression analysis showed that the following were an poor prognostic factor of event-free survival rate: TC genotype at rs10873876 locus of SIAT7C gene (HR=2.111,95%CI=1.260-3.539, P=0.005) and GG genotype at rs11155133 locus of LOC102723724 gene (HR=4.787,95%CI=1.652-13.870, P=0.004). At the same time, they were also poor prognostic factors of recurrence rate (the former HR=2.082, 95%CI=1.242-3.492, P=0.005, the latter HR=4.067, 95%CI=1.380-11.988, P=0.011).Conclusion: The rs10821936 and rs10994982 of ARID5B gene, rs7554607 of ZNF230 gene, rs11978267 and rs4132601 of IKZF1 gene are the potential susceptibility loci of ALL in Chinese children. Polymorphism loci of rs10873876 of SIAT7C and rs11155133 of LOC102723724 can be prognostic factors for childhood ALL.

Protective effect of chalcone derivative L2H24 on cardiomyocytes injured by H2O2

Objective: To investigate the anti-oxidative protective effect of chalcone derivative L2H24 on H2O2-induced H9c2 cardiomyocytes injury and its related molecular mechanism. Methods: The survival rate of cardiomyocytes was detected by MTT experiment; cell proliferation ability was detected by colony cloning experiment; cell pyroptosis was observed under the inverted microscope; MDA level of lipid peroxidation in cells was detected by thiobarbituric acid color reaction; the expression level of HO-1, Nrf2, Caspase-1 p48, Caspase-1 p20 and IL-1β in the cells was measured by Western blotting analysis. Results: L2H24 not only increased the survival rate of H9c2 cells damaged by H2O2 (P<0.05), promote the formation of cell colonies and reduce the level of cell lipid peroxidation (P<0.05), but also reduced the expression level of Caspase-1 p48, Caspase-1 p20 and IL-1β in cells (P<0.05) and improved the pyrolysis. Furthermore, L2H24 promoted the translocation of Nrf2 into the nucleus and up-regulated the expression level of its downstream antioxidant protein HO-1, which protected cardiomyocytes from H2O2-induced oxidative damage. Conclusion: Chalcone derivative L2H24 plays a protective role against oxidative damage, and its mechanism may be related to the activation of Nrf2/HO-1 signaling pathway.

The antioxidant activity of chalcones analogues in scavenging DPPH free radical and protecting against H2O2-induced PC12 cell injury

Objective: To investigate the antioxidant activity of the synthesized chalcone analogues in scavenging DPPH free radical and protecting against H2O2-induced PC12 cell injury. Methods: PC12 cells were divided as control group (DMSO group), injury group (H2O2 group), chalcone analogue+H2O2 group and positive control group (quercetin+H2O2 group). Sixteen chalcone analogues were designed and synthesized. DPPH assay was used to detect free radical scavenging activity; MTT assay was used to detect cell activity; MDA kit was used to detect lipid peroxidation; Hoechst staining was used to detect cell apoptosis; Western blot was used to detect cleaved-caspase-3, Bcl-2 and Bax protein expression. Results: Antioxidant 13 with excellent activity and low toxicity was obtained. Compared with DMSO group, the cell survival rate and Bcl-2 protein expression were decreased significantly, but cleaved-caspase-3 and Bax protein expression, MDA content were increased significantly in H2O2 group (all P<0.05); Compared with H2O2 group, cell survival rate was increased, Bcl-2 protein expression was up-regulated, cleaved-caspase-3 and Bax expression was down-regulated, MDA level were decreased in a dose-dependent manner by group 13 treatment (all P<0.05). Conclusion: The novel chalcone analogue 13 can inhibit H2O2-induced PC12 cells apoptosis, which has good antioxidant activity.

The protective effect of estradiol on cardiac function in ischemia reperfusion injury via MIF-AMPK signal pathway

Objective: To investigate the protective effect of estradiol on myocardial ischemia-reperfusion injury in mice and its possible mechanism. Methods: B-ultrasonography and myocardial TTC staining were used to study the effect of estradiol on the acute stage of myocardial ischemia-reperfusion model in mice, and protein western blot was used to detect the effect of estradiol on macrophage migration inhibitory factor (MIF) and downstream AMPK and Acetyl CoA carboxylase (ACC) phosphorylation. Results: The left ventricular ejection fraction and short-axis shortening rate of mice after myocardial ischemia-reperfusion injury under estradiol were improved, compared with other groups (P<0.05). MIF level and AMPK and ACC phosphorylation were increased in ischemia-reperfusion myocardium after estradiol treatment (P<0.05). Conclusion: Estradiol may play a protective role in the acute stage of myocardial ischemia-reperfusion injury in mice through the MIF-AMPK signaling pathway.

The association between CT measured fatty liver and the severity of hyperlipidemia acute pancreatitis

Objective: To explore the association between CT measured fatty liver and the severity of hyperlipidemic acute pancreatitis (HLAP). Methods: Clinical data from 223 HLAP patients were retrospectively collected. According to the scoring by the modified CT severity index (MCTSI), there were 62 patients with mild HLAP and 161 with moderate to severe HLAP. Based on the liver/spleen CT value ratio, the patients were divided as no-to-mild fatty liver group and moderate-to-severe fatty liver group. Univariate and multivariate logistic regression analyses were used to analyze the independent risk factors of the severity of HLAP. Results: Of the HLAP patients, 184 (82.5%) presented fatty liver. Compared with patients with no or mild fatty liver, patients with moderate-to-severe fatty liver had a higher probability of hospitalization by more than 2 weeks (55.7% vs.39.3%, P<0.05). Compared with the mild HLAP group, the average triglyceride (TG) level of patients in the moderate-to-severe HLAP group was significantly higher (P<0.01), while the liver/spleen CT value ratio was significantly lower (P<0.01). There were no significant differences in body mass index, waist circumference,muscle area, subcutaneous fat area, visceral fat area, visceral fat area/total fat area between the two groups (all P>0.05). Univariate Logistic regression analyses showed that a co-existence of moderate-to-severe fatty liver and a high TG level were risk factors for moderate-to-severe HLAP (both P<0.05). After adjustment by multivariate Logistic regression analyses, the presentation of moderate-to-severe fatty liver was still an independent risk factor for moderate-to-severe HLAP (P<0.05). In addition, after treatment, 67.0% of the moderate-to-severe fatty liver presented in the HLAP patients were normalized to no or mild fatty liver. Conclusion: The severity of fatty liver assessed by CT was associated with the severity of HLAP patients and the length of hospitalization.

The effect of delayed cord clamping on umbilical artery blood gas and jaundice in term infants of diabetic mothers

Objective: To investigate the effect of delayed cord clamping on umbilical artery blood gas and jaundice in term infants of diabetic mothers. Methods: Totally 161 full-term pregnant women with gestational diabetes mellitus who delivered in the Second Affiliated Hospital of Wenzhou Medical University from January 2020 to August 2020 were enrolled in this study. They were randomly allocated into the early cord clamping group (<15 seconds, ECC) and the delayed cord clamping group (≥30 seconds, DCC). According to the inclusion and exclusion criteria, 132 cases remained, with 66 cases in the ECC group and 66 cases in the DCC group. The results of umbilical artery blood gas analysis and neonatal transcutaneous bilirubin levels at 1-4 days after birth were compared between the two groups. Results: Arterial cord blood base excess (BE) in the DCC group was significantly lower than that in the ECC group, and lactate was significantly higher [BE: -3.72(-5.58, -2.41) vs.-3.06 (-4.79, -1.62)mmol/L, lactate: 3.0 (2.4, 4.3) vs. 2.1 (1.6, 4.0)mmol/L, P<0.05]. There was no significant difference in pH and bicarbonate (HCO3-) between the two groups [pH: 7.29 (7.24, 7.33) vs. 7.31 (7.25, 7.33),HCO3-: 24.0 (22.5, 25.0) vs. 24.3 (24.3, 25.5)mmol/L, P>0.05]. There were no significant differences in the partial pressure of carbon dioxide (PCO2) and partial pressure of oxygen (PO2) between the two groups (P>0.05). Also, DCC did not improve hemoglobin and hematocrit levels (P>0.05). There was no difference in transcutaneous bilirubin levels at 1-4 days after birth between the two groups (P>0.05), but and the incidence of hyperbilirubinemia in the DCC group was markedly higher than that in the ECC group (17.0% vs. 6.1%) (P<0.05). Conclusion:Delayed cord clamping enhanced anaerobic metabolism, increased metabolic acid in term infants of diabetic mothers, and increased the incidence of neonatal hyperbilirubinemia but did not affect pH value.

Comparative analysis of multislice spiral CT characteristics between pulmonary disease due to mycobacterium avium-intracellularis and multi-drug resistant tuberculosis

Objective: To investigate the similarities and differences of imaging features of multislice spiral CT (MSCT) between pulmonary disease due to mycobacterium avium-intracellularis and multi-drug resistant tuberculosis. Methods: The difference of MSCT images between 41 cases of MAC lung disease (MAC group) identified by bacteria and 41 cases of MDR-TB (MDR-TB group) matched by 1:1 was analyzed retrospectively.Statistical analysis was performed. Results: The detection rate of thin-walled cavity in the MAC group was 69.2%,higher than that in the MDR-TB group; average wall thickness in MAC group was 2.5 (1.8, 3.3)mm, while it was 3.9 (2.7, 5.0)mm in MDR-TB group. The differences were all statistically significant (P<0.001). The detection rate of lobular of outer wall of cavity or burr sign, satellite focus of cavity and drainage bronchus in MAC group was 53.8%, 49.2% and 27.7%, respectively, lower than those in the MDR-TB group (79.6%, 93.9%, 67.3%,respectively) with significant differences (P<0.05). The detection rate of bronchiectasis in the MAC group,bronchiectasis involvement range ≥3 lung lobes, right lung middle lobe, left lung tongue lobe, and left lower lobe bronchiectasis detection rate, columnar bronchiectasis, bronchiectasis with lobular central nodules wasrespectively 73.2%, 43.9%, 63.4%, 48.8%, 22.0%, 53.3%, 56.1%, higher than the MDR-TB group (46.3%, 9.8%,12.2%, 2.4%, 4.9%, 21.1%, 24.4% respectively). The differences were statistically significant (P<0.05). The proportion of cystic bronchiectasis in the MAC group (13.3%) was lower than MDR-TB group (47.4%) with statistical difference (P<0.05). The detection rate of nodules/spherical masses in the MAC group > 1 cm was 22.0%, and compared with 68.3% in the MDR-TB group, there was statistical difference (P<0.001). Conclusion:The MSCT manifestations of MAC lung disease and MDR-TB lung disease have their own specificity. The type of cavities, the type of bronchiectasis, dominant location and distribution, and the size of nodules are of differential significance, which are helpful for clinical diagnosis.

Synthesis of Schiff base of adenosine 3’, 5’-cyclicmonophosphoric acid with xanthone and its binding to LC3B

Objective: To design the potential proteolysis targeting chimeras (PROTAC) drug S1 synthesized from adenosine 3’, 5’-cyclicmonophosphoric acid and xanthone. Methods: The binding kinetics of S1 and LC3B was characterized by UV-Vis spectroscopy. The resulted data were fitted to first order reaction kinetics.The binding constants and thermodynamic constants of the S1-LC3B complex were determined by fluorescence spectroscopy, and the molecular binding mechanism was deduced. Results: S1 bound to LC3B fast within 160 s.The binding kinetics obeyed the first order reaction kinetics equation and specific transport rule. Thermodynamic
study found that the Gibbs free energy change was negative, indicating a spontaneous binding process. And negative enthalpy change suggested a static attraction between S1 and LC3B. Such course was attributed to the interaction of C=N at S1 with LC3B, resulting in fluorescence quenching. Conclusion: The Schiff base S1 can
spontaneously bind to LC3B.

The effect of prior cesarean section on outcomes of in vitro fertilization-embryo transfer

Objective: To investigate the effect of prior cesarean delivery (CD) on the clinical outcomes of in vitro fertilization-embryo transfer (IVF-ET). Methods: Clinical data of women who underwent IVF-ET in the assisted reproduction technology center of the Second Affiliated Hospital & Yuying Children’s Hospital of Wenzhou Medical University from January 2019 to December 2019 were retrospectively analyzed. According to their delivery history, patients with a history of live birth by CD were assigned to the CD group (n=292), and those with only vaginal delivery (VD) were categorized into the VD group (n=311). The embryo implantation rate and treatment outcomes were compared between the two groups. Results: The embryo implantation rate of VD group was significantly higher than VD group (P<0.05). However, there was no statistical difference in miscarriage rate, twinning rate and incidence of ectopic pregnancy between the two groups, neither in indexes of premature, stillbirth, birth weight, birth defects, placenta previa and postpartum hemorrhage. Conclusion: A prior CD does affect the embryo implantation rate, but it is not associated with increased risk of miscarriage and other adverse treatment outcomes in IVF patients.

The clinical effect of intraperitoneal chemotherapy utilizing carboplatin and paclitaxel on high-grade serous ovarian cancer

Objective: To investigate the curative effect of carboplatin/paclitaxel intraperitoneal chemotherapy on high-grade serous ovarian cancer (HGSOC). Methods: According to the diagnostic result given by senior pathologists from our hospital, 110 cases of the most typical high-grade serous ovarian cancer were selected from numerous gynecological tumor slices of the First Affiliated Hospital of Wenzhou Medical University from January 2015 to July 2019. Considering their clinical data, these patients were assigned to four groups. Group A included 34 patients treated by paclitaxel combined with carboplatin intravenous chemotherapy;group B was 30 patients by paclitaxel combined carboplatin intraperitoneal chemotherapy; group C included 28 patients by paclitaxel combined with cisplatin intraperitoneal chemotherapy and group D was intravenous group,including 18 patients treated with traditional chemotherapy drugs (cyclophosphamide, adriamycin, etc.). The effectiveness and safety of carboplatin /paclitaxel intraperitoneal chemotherapy in high-grade serous ovarian cancer (HGSOC) were evaluated by comparing the four groups in the survival rate and side effects. Results: The 5-year survival rate of group A, group B, and group C was significantly higher than that of group D, with statistical difference between each group (P<0.05). The 3-year survival rate of group B and C was also higher than that of group D, with statistical difference between the groups (P<0.05).However, the 3-year and 5-year survival rates between groups A, B, and C showed no statistical difference (P>0.05).The incidence of grade III, IV bone marrow suppression was the highest in group D without significant difference among the four groups (P>0.05). The incidence of grade III, IV gastrointestinal reactions and abdominal pain was significantly different (P<0.05), but with no difference either between group A and group B or between group C and group D (P>0.05). And there was no significant difference in the incidence of rash among the four groups (P>0.05). Conclusion: Compared with cisplatin, carboplatin combined with paclitaxel intraperitoneal chemotherapy can prolong the survival time of patients with HGSOC, improve the treatment effectiveness and have fewer side effects, which can provide reference for future clinical chemotherapy.

Investigation among medical staff in emergency critical care department on the status quo of knowledge,attitude, behavior regarding hyperoxemia and its influencing factors

Objective: To investigate and study the current situation of oxygen therapy cognition of medical staff in emergency critical care unit in Jiangsu and Zhejiang regions, so as to find out relevant problems and put forward corresponding measures. Methods: A questionnaire was made to investigate the knowledge, attitude and behavior status regarding oxygen therapy among medical staff in the emergency critical care unit in some tertiary hospitals in Jiangsu and Zhejiang provinces. The method of convenient sampling was adopted to carry out the secret test, and the literature about oxygen therapy was reviewed, and the survey results were counted and analyzed. Results: The average scores of the knowledge, attitude and behavior regarding hyperoxemia was (27.90±3.91), (23.99±2.59) and (38.15±7.21), respectively. The equation of influencing factors of hyperoxia cognition formed by the medical staff in the in the emergency critical care unit was as follows: y=105.121-7.067x1 5.155x2+2.892x3. The results showed that the regression equation could account for 14.6% of the total cognitive variation of hyperoxia in emergency critical care medical staff. Conclusion: The medical staff in the emergency department of tertiary hospitals have a good knowledge and attitude about hyperoxemia, but need to
improve their behavior in Jiangsu and Zhejiang regions. Therefore, it is urgent to establish a standardized clinical system of oxygen treatment.
