345 Construction of a microbial reporter strain for detecting bioavailable copper ions in drinking water and wastewater

Objective: To construct a microbial reporter strain that can detect bioavailable copper ions in drinking water and wastewater. Methods: Firstly, the gene sequence encoding enhanced green fluorescent protein was fused with the promoter region of copper ion “efflux pump” gene copA by recombinant PCR. The obtained PCR product was subcloned to pET28a plasmid to generate the copper-responsive vector CopAp::gfpmut2pET28a. Next, the genes responsible for the copper resistance in wild-type E. coli cell (MC4100) were deleted using the Red recombination method, before the CopAp::gfpmut2-pET28a vector was transformed into the engineered E. coli cell to accomplish the construction of the copper-sensing E. coli strain. Finally, the optimal conditions for copper detection were established and the copper-detecting strain was applied to detect copper ions in water. Results: Compared with wild-type strain, the sensitivity of the E. coli ΔcusA ΔcopA ΔcueO mutant to copper was enhanced by 64-fold, thus higher fluorescence signal to copper ions was successfully achieved. Furthermore, the fluorescence intensity of the biosensor also had dose-dependent relation with copper concentration. In optimized assay conditions, the linear detection range of Cu2+ was 0.05-2.5 μmol/L (0.0032-0.16 mg/L). In comparison with atomic absorption spectrometry, the recovery rate of 0.05 mg/L and 1 mg/L copper ions in drinking water was 87.90% and 104.37% respectively. Conclusion: According to the national standard, the linear detection range of the bioreporter strain constructed in this study can fully meet the demand for effective copper detection in drinking water and wastewater, which may provide this whole-cell bioreporter strain with technical  feasibility for practical application.

356 The mitochondrial tRNAPhe 593T>C mutation may influence the phenotypic expression of the LHONassociated m.14484T>C mutation

Objective: To investigate the role of mitochondrial tRNAPhe 593T>C in the development of Leber’s hereditary optic neuropathy (LHON)-associated with the MT-ND6 14484T>C mutation. Methods: A cohort of 742 Han Chinese subjects with LHON and 376 control subjects underwent sequence analysis of the complete mitochondrial DNA. 4 Chinese families with LHON were evaluated for clinical, genetic and molecular biological characteristics. Results: The penetrance of vision loss in these pedigrees (WZ133, WZ1224, WZ1225 and WZ1260) was 42.9%, 12.5%, 12.5% and 5.6%, respectively. The age-at-onset of vision impairment in the four families varied from 9 to 23 years. The average age-of-onset was 16. Sequence analysis of mitochondrial genome indicated that WZ133 carried m.14484T>C and m.593T>C, while the other three families only carried m.593T>C. Analysis of the secondary structure of mitochondrial tRNA showed that this mutation resulted in a secondary structure change and higher free energy of tRNAPhe. The alterations reduced the stability of tRNAPhe structure and led to mitochondrial dysfunction. Conclusion: The tRNAPhe 593T>C mutation may have a potential modifier role in the phenotypic manifestation of the primary LHON-associated m.14484T>C mutation.

364 NMR-based metabolomics analysis of brain region-specific metabolic changes in diabetic rats

Objective: To investigate the metabolic changes in different brain regions of diabetic rats. Methods: Hippocampus, cortex, hypothalamus and midbrain were dissected from STZ-induced diabetic rats and the age-matched controls. 1H NMR-based metabolomics was performed to detect metabolic profiles in different brain regions, and metabolic characteristics were analyzed by using metabolic pattern recognition and pathway analysis. Results: Diabetic rats had a significant metabolic phenotype in cortex, striatum, hypothalamus and midbrain as compared with age-matched controls. Quantitative results of metabolites showed that diabetic rats had brain region-specific metabolic alterations, compared with controls. Lactate and taurine were distinctly increased in cortex (P<0.05), while the levels of N-acetyl-aspartate and choline were markedly decreased (P<0.05); Lactate and taurine were distinctly increased in striatum (P<0.05), while the levels of N-acetyl-aspartate and GABA were markedly decreased (P<0.05); Taurine and myo-inositol were distinctly increased in hypothalamus (P<0.05), while the levels of N-acetyl-aspartate and Succinate were markedly decreased (P<0.05); Lactate and myo-inositol were distinctly increased in midbrain (P<0.05), while the levels of N-acetyl-aspartate and GABA were markedly decreased (P<0.05). Conclusion: Diabetic rats exhibit region-specific changes in brain metabolism, mainly involving energy metabolism, neurotransmitter metabolism and choline metabolism.

371 M2 phenotype in THP-1 macrophages induced by supernatants of cervical cancer cell culture

Objective: To study the effect of supernatant in cervical cancer cell culture on inducing stable M2 phenotype in THP-1 macrophages. Methods: When cervical cancer cell line HeLa cells were cultured on day 3, cell culture supernatant was collected to detect tumor-related cytokines (IL-4, IL-6 and IL-10) and growth factors (angiopoietin-2, hepatocyte growth factor, vascular endothelial growth factor). Macrophages were cultured to logarithmic growth stage and then divided into blank control group (Mθ group), M2 cell induction group (M2 group), and Hela cell culture supernatant treatment group (Mθ+sHeLa group). The M2 group was treated with 20 ng/mL IL-4 and 20 ng/mL IL-10 to polarize M2 cells, and Mθ+sHeLa group was treated with 50% HeLa cell culture supernatant for 3 d, subsequently. Flow cytometry was used to detect the changes of HLA-DR, CD163 and CD206 on the cell surface. Meanwhile, the supernatant of cell culture was collected and the contents of related cytokines, growth factors and TGF-β3 were detected. Finally, the changes of the JAK-STAT6 signaling pathway in macrophages were detected by flow cytometry and Western blot. Results: The content of IL-6, HGF, and VEGF in Hela cell culture supernatant was high. The content of CD163 and CD206 on the surface of macrophages increased after treatment with HeLa cell culture supernatant. Compared with the Mθ group, the levels of related cytokines (IL-4, IL-6 and IL-10), growth factors (ANG-2, HGF, VEGF) and TGF-β3 increased in supernatant of M2 and Mθ+sHeLa group. After removal of the supernatant of HeLa cell culture for 48 h, the content of related cytokines, growth factors and TGF-β3 were still increased in the Mθ+sHeLa group compared with the Mθ group. Finally, flow cytometry and Western blot results showed that the level of pSTAT6 was increased in the Mθ+sHeLa group compared with the Mθ group. Conclusion: Hela cell culture supernatant can promote tumor growth and angiogenesis by activating the polarization of THP-1 macrophages into the M2 via JAK-STAT6 signal pathway.

377 The role of 1, 25-dihydroxyvitamin D3 in palmitic acid induced insulin resistance in human umbilical vein endothelial cells

 Objective: To investigate the effect of 1, 25-dihydroxyvitamin D3 [1, 25(OH)2D3] on the insulin resistance of human umbilical vein endothelial cells (HUVECs) induced by palmitic acid (PA) and its possible mechanism. Methods: HUVECs were cultured in DMEM medium containing (0.3, 0.4, 0.5, 0.6, 0.7, 0.8, 0.9 mmol/L) PA to establish insulin resistance endothelial cell model. 0, 0.01, 0.1, 1, 10, 100 nmol/L 1, 25(OH)2D3 intervention, nitrate/nitrite fluorescence kit to determine the content of NO, Western blot to determine the expression of pAkt and penos protein. The expression of VDR and ET-1 was detected by qPCR. Results: after HUVECs were cultured with 0.6 mmol/L PA for 18 hours, the glucose concentration in the culture medium was significantly lower than that in the control group, the cell viability was reduced, and the endothelial cell model of insulin resistance was established. The NO content of HUVECs in PA group was lower than that in control group (P<0.05). After the intervention of 1 nmol/L 1, 25(OH)2D3 on insulin resistant endothelial cells, the protein expression of peNOS and pAkt increased significantly (P<0.05), and the mRNA expression of ET1 decreased significantly (P<0.05). Conclusion: 1, 25(OH)2D3 can increase the expression of peNOS and pAkt, increase the production of NO and decrease the expression of ET-1, which may have a protective effect on insulin resistant vascular endothelial cells.

382 Effect of febuxostat on patients with CKD3-4 diabetic nephropathy complicated with hyperuricemia

Objective: To evaluate the effect of febuxostat on patients with CKD3 and CKD4 stage diabetic nephropathy (DN) with hyperuricemia (HUA) through prospective randomized controlled clinical studies. Methods: Diabetic nephropathy patients [glomerular leachate (eGFR) 15-59 mL/(min·1.73 m-2)] combined HUA, who were treated in the Second Affiliated Hospital of Wenzhou Medical University from January 2017 to June 2019, were recruited as subjects. According to the eGFR value, patients were divided into CKD3 group [eGFR is 30-59 mL/(min·1.73 m-2)] and CKD4 [eGFR is 15-29 mL/(min·1.73 m-2)]. CKD3 group was further divided randomly as treatment group 1 and control group 1. CKD4 group was further divided randomly as treatment group 2 and control group 2. Blood glucose, blood pressure, lipids controlling and renin angiotensin inhibitors (RASI) treatment were performed on all groups in the meantime. Febuxostat was given to patients in treatment group 1 and treatment group 2. Serum uric acid (UA), serum creatinine (SCr), serum cystayl C (Cys-C), eGFR, 24 hours of urine protein quantification, albumin creatinine Ratio (ACR), neutrophi gelatinase-associated lipocalin (NGAL) and renin-angiotensin- aldosterone system (RASS) were observed after 24 weeks. Results: A total of 95 patients completed the study and were qualified for selection, among whom 26 were in the treatment group 1, 24 in the control group 1, 23 in the treatment group 2 and 22 in the control group 2. Significant differences were observed in the distribution of UA, SCr, Cys-C, eGFR, 24 hours of urinary protein quantification, ACR, NGAL and RASS at 24 weeks between CKD 3 treatment group1 and control group 1. There were significant differences in the distribution of UA, SCr, Cys-C, eGFR, ACR, NGAL and RASS between CKD4 treatment group 2 and control group 2 (P<0.05), but no significant difference in urine protein quantification (P>0.05). In treatment group 1, the UA standard rate was 95% after 24 weeks of treatment. In treatment group 2, the UA standard rate was 85% after 24 weeks of treatment. Conclusion: Febustat can improve renal function in patients with CKD3 and CKD4 stage DN with HUA and has proved to be safe.

387 The effect of calcium dobesilate on the formation of atherosclerotic plaques and its mechanism

Objective: To discuss the effect of calcium dobesilate on the formation of atherosclerotic plaques in rabbits and its mechanism. Methods: Atherosclerotic plaque model was set up by liquid nitrogen frostbite in 24 health male New Zealand rabbits. The rabbits were randomly divided as the model group (n=12) and the treatment group (n=12). The model group was given high-fat diet for 8 weeks, while the treatment group was given high-fat diet and calcium dobesilate (0.15 mg·g-1·d-1) for 8 weeks. Serum endothelin-1 (ET-1), nitric oxide (NO) and vascular endothelial growth factor (VEGF) were measured by ELISA before the experiment and after 8 weeks of treatment. After 8 weeks, the animals were killed, the right common carotid artery was removed, and the pathomorphological changes were observed by light microscope and electron microscope. The expression of matrix metalloproteinase 9 (MMP-9) and plasminogen activator inhibitor-1 (PAI-1) were detected by immunohistochemistry. Results: After 8 weeks, the vascular intima in the model group showed typical atherosclerotic plaque lesions, while the vascular intima in the treatment group showed slight atherosclerotic plaque lesions. The expression of PAI-1 and MMP-9 protein was observed locally in the carotid arteries of both groups, but the expression level in the treatment group was significantly lower than that in the model group (P<0.05). The expression of ET-1 was significantly decreased. The expression of NO and VEGF was significantly increased in both groups, but the decrease in ET-1 in the treatment group was greater than that in the model group, the increase in NO and VEGF was greater than that in the model group, with significant difference (P<0.05). Conclusion: Calcium dobesilate can improve endothelial dysfunction by reducing the expression of PAI-1 and MMP-9 proteins in local vascular walls, reducing the ET-1 content in blood circulation and increasing the levels of NO and VEGF, which play the role of anti-atherosclerotic plaque formation.

391 The relationship between vascular endothelial growth factor gene polymorphism and endometriosis

Objective: To investigate whether polymorphisms of vascular endothelial growth factor (VEGF) in promoter region (rs3025040 C/T, rs10434 G/A) are associated with hereditary susceptibility of endometriosis. Methods: A total of 150 patients with endometriosis pathologically diagnosed who were admitted to the First Affiliated Hospital of Wenzhou Medical University from March 2014 to March 2015 were selected as case group and another 150 healthy volunteers were collected as control group. 5 mL blood samples were obtained from both groups in this assay and genomic DNA was extracted from peripheral blood leukocyte using TIANamp Blood DNA Kit. Special DNA PCR primers were designed using Primer Premier 5 software. The genotypes of all sites were detected by polymorphism chain reaction and restriction fragment length polymorphism analysis (PCRRFLP). Results: The C and T allele frequencies in VEGF-rs3025040 C/T site in case group and control group were 76.67%, 23.33% and 58.00%, 42.00%, respectively. There was significant difference in allele distributions of the VEGF-rs3025040 C/T site between two groups (χ2=23.762, P<0.001, OR=1.800, 95%CI=1.410-2.298). Compared with the T allele, the C allele significantly increased the risk of developing endometriosis. The G and A allele frequency in VEGF-rs10434 G/A site in case group and control group was 60.00%, 40.00% and 81.67%, 18.33% respectively. There was significant difference in allele distributions of the VEGF-rs10434 G/A site between two groups (χ2=34.084, P<0.001, OR=1.344, 95%CI=1.207-1.497). Compared with the G allele, the A allele significantly increased the risk of developing endometriosis. Conclusion: The results suggest that VEGFrs3025040 C/T, rs10434 G/A polymorphisms are associated with endometriosis.

395 Expression of LINC00460 in clear cell renal cell carcinoma and its clinical significance

Objective: To explore the expression and clinical significance of LINC00460 in clear cell renal cell carcinoma (ccRCC). Methods: Gene expression profiles of 611 ccRCC patients and their clinical data were downloaded from TCGA database. We studied the expression of LINC00460 in pairs of ccRCC tissues and matched non-cancerous adjacent tissues, and then analyzed the relationship between expression of LINC00460 and clinical parameters and prognosis of patients. Results: Over-expression of LINC00460 was found more in ccRCC tissues than non-cancerous adjacent tissues (P<0.05). The increased expression of LINC00460 was significantly associated with high pathological stage (I/II vs. III/IV, OR=2.82, 95%CI=1.96-4.07, P<0.001), T stage (T1/2 vs. T3/4, OR=2.65, 95%CI=1.84-3.84, P<0.001), N stage (N0 vs. N1, OR=7.80, 95%CI=2.12-50.40, P=0.007), M stage (M0 vs. M1, OR=3.22, 95%CI=1.91-5.61, P<0.001) and histological grade (G1/2 vs. G3/4, OR=2.08, 95%CI=1.46-2.95, P<0.001). Cox regression analysis showed that LINC00460 was an independent factor in predicting the overall survival of ccRCC patients (HR=1.05, 95%CI=1.01-1.10, P=0.010). Conclusion: LINC00460 have over-expression in ccRCC tissues than non-cancerous adjacent tissues. Patients of high LINC00460 expression have worse prognosis than patients of low LINC00460. Therefore, LINC00460 could be a biomarker for the prognosis of ccRCC patients.

401 Association between serum uric level and renal function in elderly women

Objective: To investigate the relationship between the renal function and the serum uric acid (SUA) level in elderly women. Methods: A total of 230 qualified female patients (71-93 years) were asked to complete a questionnaire and do medical checkups. Multiple linear regression and logistic regression were used respectively to analyze the association between SUA and renal function. Results: The prevalences of chronic kidney disease (CKD) in four SUA quartiles (UA1: 157-mmol/L, UA2: 262-μmol/L, UA3: 312-μmol/L, UA4: 362-556 μmol/L) were 31.58%, 39.66%, 53.45% and 84.21%, respectively, with significant difference (P<0.05). A significant inverse correlation was observed between the estimated glomerular filtration rate (eGFR) and SUA based on a multivariable linear regression model after adjusting age, body mass index (BMI) and total cholesterol (TC). Compared with those in UA1, higher SUA levels were significantly associated with a greater decline in eGFR (UA2: β=-4.50, 95%CI: -9.04, -0.04; UA3: β=-6.57, 95%CI: -10.67, -2.47; UA4: β=-13.48, 95%CI: -17.73, -9.23) after adjusting the above confounding factors. Meanwhile, a multiple logistic regression analysis clearly revealed that participants in the UA4 category had the highest odds to develop CKD as compared to their counterparts, in which the adjusted odds was 10.64 times of that in participants from UA1 (OR: 10.64, 95%CI: 4.15-27.34, P<0.01). Conclusion: Elevated SUA was an independent risk factor of the declined renal function and significantly associated with increased odds of CKD in old women.

406 Dissolution determination for vitamin B2 in compound vitamin B tablets and its evaluation

Objective: To establish a method to determine the dissolution of vitamin B2 in compound vitamin B tablets and evaluate the dissolution curves of different samples. Methods: The HPLC was used, and the separation was performed on a CAPCell PAK C18 (4.6 mm×250 mm, 5 μm) column with the mobile phase consisting of the solution containing 2 mmol/L sodium heptane sulfonate and 50 mmol/L ammonium dihydrogen phosphate-acetonitrile (85:15) at a flow rate of 1.0 mL/min. The column temperature was 35 ℃. The detection wavelength was 267 nm. The paddle method was used for the dissolution test of vitamin B2 with 600 mL water as the medium and the rotation rate was set at 75 r/min. Results: Under the chromatographic condition in this study, vitamin B2 (Riboflavin) revealed good linearity within the range of 0.126-5.023 μg/mL (r=0.9999). The average recovery was 103.7%, 100.0% and 99.4% respectively at low, intermediate and high concentration, with the RSD being 0.7%, 1.3%, 0.3%, respectively. The dissolution method was suitable for showing differences in dissolutions among products of different manufacturers. Conclusion: The established HPLC method is simple, which shows good specificity, precision and accuracy. The dissolution method can reflect differences among products of different manufacturers. Some manufacturers should improve the production process, strengthen the dissolution of products to improve the effectiveness of drugs.

411 Clinical significance of monitoring serum NO and AGEs in patients with asymptomatic type 2 diabetic peripheral neuropathy

 Objective: To study the clinical significance of monitoring nitric oxide (NO) and advanced glycation end-products (AGEs) in asymptomatic type 2 diabetes mellitus (T2DM) peripheral neuropathy (DPN). Methods: From December 2015 to December 2018, a total of 80 inpatients with T2DM from the Third Affiliated Hospital of Zhejiang University of Traditional Chinese Medicine were selected. 60 patients were diagnosed as DPN by measuring vibration perception threshold (VPT) and nerve conduction velocity (NCV), and the remaining 20 patients were in simple T2DM group. And then 60 DPN patients were further divided into symptomatic DPN group (n=29) and asymptomatic DPN group (n=31) by the presence or absence of limb numbness, burning sensation, pain and other symptoms. The levels of fasting plasma glucose (FPG), 2-hour postprandial plasma glucose (PPG), glycosylated hemoglobin (HbA1c), triglyceride (TG), total cholesterol (TC), low-density lipoprotein cholesterol (LDL-c), high-density lipoprotein cholesterol (HDL-c), apolipoprotein a (APOa) and apolipoprotein b (APOb) were compared among the three groups, and the results of NO and AGEs were also compared among the three groups. Results: The levels of FPG, PPG and HbA1c in symptomatic DPN group were significantly higher than those in asymptomatic DPN group and simple T2DM group (P<0.01). The levels of FPG, PPG, and HbA1c in asymptomatic DPN group were higher than those in simple T2DM group (P<0.05). There were no significant differences in the levels of TG, TC, LDL-c, HDL-c, APOa and APOb among the three groups (P>0.05). Compared with simple T2DM group, in two DPN groups the levels of serum NO were significantly decreased and the levels of serum AGEs were significantly increased (P<0.01); the levels of serum NO and AGEs were significantly different between the two DPN groups (P<0.01). Conclusion: The incidence of early asymptomatic DPN is high in clinical practice. NO and AGEs are important biological signals and effector molecules. Early detection of them will help to understand the degree of neuropathy. Controlling blood glucose is of great clinical importance in reducing the incidence of DPN.

Objective: To analyze the relationship between the amplitude of low-frequency fluctuation (ALFF) of resting functional magnetic resonance imaging under resting state and the severity degree of Tourette syndrome in children. Methods: 20 first-episode children with Tourette syndrome, admitted by the First People’s Hospital of Taizhou from February 2017 to April 2018, were recruited in the study, and all the patients were examined by fMRI. The ALFF of brain was respectively calculated in different frequency bands, and the relationship between the severity of symptoms and the ALFF was discussed that. Results: It was shown that a positive correlation between the score of symptom severity in orbital part of left superior forehead gyrus, right-to-middle frontalis orbital, left middle frontalis orbital region and the ALFF (P<0.05), and a negative correlation between the score of symptom severity in left distal sulcus, right distal sulcus, peripheral cortex and the ALFF (P<0.05). Conclusion: There is a significant correlation between the severity of symptoms in children with Tourette syndrome in resting state and abnormal brain function at regions.

419 The preposition of distal ileum under the abdominal wall in laparoscopic anterior resection of anterior cancer

Objective: To analyze the preposition of distal ileum under the abdominal wall (without ileostomy) in laparoscopic anterior resection of rectal cancer. Methods: From December 2013 to May 2018, a total of 196 cases of rectal cancer patients in our hospital were collected as subjects, who were randomly divided as preset group and ileostomy group with 98 cases in each group. The preset group received undergoing laparoscopic anterior resection for low rectal cancer+distal ileum preposition under the abdominal wall, while the ileostomy group undergoing laparoscopic anterior resection for low rectal cancer+terminal ileum protective ileostomy. Two groups were compared in the hospital expenses (including return ileum), operation time (including return ileum), length of hospital stay (including  return ileum), postoperative anastomotic fistula, intestinal obstruction, the incidence of ileostomy and return ileum related complications. Results: Compared with the ileostomy group, the preset group had lower hospitalization cost, shorter operation time and shorter length of hospital stay, all showing statistical significance (P<0.05). Postoperative anastomotic fistula occurred in 5 patients in the preset group and 6 patients in the ileostomy group, respectively. There was no statistical difference in the incidence of postoperative anastomotic fistula between the preset group and the ileostomy group (P=0.611). There were 10 cases of intestinal obstruction in the preset group and the ileostomy group, respectively. There was no statistical difference in the incidence of postoperative intestinal obstruction between the preset group and the ileostomy group (P=0.824). There were 1 case and 21 cases in the preset group and the ileostomy group, respectively, showing ileostomy and return ileum related complications., and the incidence rate of ileostomy and return ileum related complications in the preset group was significantly lower than that of the ileostomy group (P=0.008). The difference was statistically significant. Conclusion: The application of distal ileum preposition under the abdominal wall can reduce complications related to laparoscopic rectal resection of rectal cancer without increasing the risk of anastomotic leakage.
