Glypican3-WNT-JNK pathway mediates the molecular mechanism of local cell inflammation in lung induced by lipopolysaccharide

Objective: To investigate the important role of (glypican-3) GPC3 in lipopolysaccharide (LPS)-induced microenvironment of local inflammation in airway epithelial cells, and to further investigate the GPC3-WNT pathway molecular mechanism of LPS-induced local inflammation in the lung. Methods: The expression of GPC3 in alveolar lavage and lung tissue and its expression location were detected in an ALI mouse model simulated by LPS airway aspiration. Cultured human lung 16HBE cells were stimulated by LPS to establish acute inflammatory cells. The mRNA expressions of GPC3, TNF-α and TGF-β were detected by RT-PCR, meanwhile the expressions of GPC3, TNF-α and TGF-β proteins were assessed by Western Blot. The expression of inflammatory cytokines TNF-α and TGF-β was detected in 16HBE cells stimulated by exogenous GPC3. Cells were treated with GPC3-WNT pathway inhibitor at different concentrations, then expression levels of TNF-α and TGF-β were measured by the above-mentioned methods. Results: GPC3 expression showed a significant increase in ALI mouse model induced by LPS. In addition, increased expression of GPC3 was also observed in LPS in vitro. Meanwhile, exogenous GPC3 protein stimulated bronchial epithelial cells to produce a variety of inflammatory factors, which may indicate that GPC3 has pro-inflammatory effect. Additionally, the production of inflammatory cytokines in bronchial epithelial cell induced by GPC3 was blocked by inhibitors of the WNT-JNK pathway, suggesting that GPC3 involved in the potential signaling pathway in local lung cells during ALI/ARDS. Conclusion: There is a linear signal pathway, GPC3-WNT-JNK, in the process of LPS stimulating bronchial epithelial cells. This discovery may provide a new molecular target and molecular mechanism for the treatment of ALI/ARDS.

The relationship between expression of miR-631 and prognosis of epithelial ovarian cancer

Objective: To explore the data of epithelial ovarian carcinoma (EOC) patients in the Tumor and Cancer Gene Map (TCGA) database by analyzing the relationship between the expression of microRNA-631 (miR-631) and the clinicopathological features and prognosis of EOC patients and to investigate the effect of miR-631 on EOC cell migration and tumor volume. Methods: Clinical pathological features and corresponding microRNA expression date were extracted from TCGA database of 570 patients with EOC. The optimal cutoff value of miR-631 expression was calculated by X-tile software, and based on this optimal cutoff value the patients were grouped. The Kaplan-Meier curve and the Cox proportional hazards model was performed to assess the prognostic value of miR-631 expression in EOC patients. The miR-631 low-expressing EOC cell lines A2780, COV504 and miR-631 high-expressing cell lines ES2 and SKOV3 were selected by up-regulating the expression of miR-631 in low-expressing cell lines and down-regulating the expression in high-expressing cell lines to observe the ability of cell to migrate. The nude mouse EOC model was established and divided as the control group and miR-631 mimic group. The tumor volume of the two groups was observed. Results: Kaplan-Meier analysis showed that the overall survival (OS) of EOC patients with miR-631 low-level expression group (miR-631 expression value<4.78) was higher than that of EOC patients with miR-631 high-level expression group (miR-631 expression value≥4.78) (HR=0.65, 95%CI=0.47-0.89, P=0.008). Univariate and multivariate COX regression analysis revealed that miR-631 expression may be an independent factor influencing the prognosis of patients with EOC (HR=0.72, 95%CI=0.54-0.97, P=0.029). Up-regulation of miR-631 expression inhibits migration of EOC cells, and down-regulation of miR-631 expression promotes migration of EOC cells. Upregulation of miR-631 expression in the EOC nude mouse model reduced EOC tumor formation volume. Conclusion: Low expression of miR-631 may indicate poor prognosis in patients with EOC. Up-regulation of miR-631 can inhibit EOC migration and significantly reduce tumor volume, which may have therapeutic implications for EOC.

Long non-coding RNA MIAT in regulating cell proliferation, migration and invasion in nasopharyngeal carcinoma cells through miR-124 sponge

Objective: To investigate whether long non-coding RNA MIAT regulates cell proliferation, migration and invasion in nasopharyngeal carcinoma cells through miR-124. Methods: qRT-qPCR was performed to detect MIAT expression in nasopharyngeal carcinoma cells. The interaction between MIAT and miR-124 was evaluated by dual luciferase reporter assay. si-MIAT and miR-124 inhibitor were transfected into nasopharyngeal carcinoma cells to detect their effects on proliferation, migration and invasion of nasopharyngeal carcinoma cells. The expression of Wnt/β-catenin signaling pathway was determined by Western blot. Results: Compared with normal nasopharyngeal epithelial cells NP-69, MIAT was significantly upregulated in nasopharyngeal carcinoma cell HONE-1. The online software and luciferase activity assay confirmed the interaction between MIAT and miR-124. Further experiments indicated that si-MIAT could increase the expression of miR-124, thus inhibiting cell proliferation, migration and invasion of nasopharyngeal carcinoma. The results of Western blot showed that MIAT functioned as an oncogene in nasopharyngeal carcinoma cell by up-regulating Wnt/β-catenin signaling pathway through miR-124. Conclusion: Our results demonstrated that MIAT affected proliferation, migration and invasion of nasopharyngeal carcinoma cells by regulating the expression of Wnt/β-catenin signaling pathway via miR-124. MIAT was a potential target for the diagnosis and treatment of nasopharyngeal carcinoma.

The effect of Let-7a over-expression on proliferation and invasiveness of colonic mucinous adenocarcinoma cell line LS-174T

Objective: To study the effect of let-7a overexpression on the proliferation and invasiveness of colonic mucinous adenocarcinoma cell line LS-174T. Methods: The let-7a overexpression lentiviral vector was constructed and infected with colonic mucinous adenocarcinoma LS-174T cells. A blank control group was established: no virus transfection; B transfected empty virus group; C1 transfection group (concentration 1× 106 TU/L): transfection of hsa-let-7a [containing let-7a virus and enhanced green fluorescent protein (EGFP)]; C2 transfection group (concentration 1×107 TU/L); C3 transfection group (concentration 1×108 TU/L). The infection efficiency of each group was observed by fluorescence microscope. The expression of let-7a was detected by RT-qPCR. The proliferation of each groupcells was detected by MTT assay. The invasion and migration ability of cells were evaluated by Transwell invasion and scratch test. RT-qPCR and Western blot analysis of TGF-β1 mRNA and protein levels were expressed. Results: The expression of green fluorescence protein in each group was observed by inverted fluorescence microscope. The fluorescence expression of C3 transfection group was the strongest, with transfection efficiency over 90%, which was positively correlated with the initial virus concentration (P<0.05). The expression levels of let-7a in each group were detected by RT-qPCR. The results showed that the expression level of let-7a in the transfected group was significantly higher than that in the negative control group, and the expression level of let-7a in the transfected cells was positively correlated with the gradient of lentivirus concentration (P<0.05). The results of Western blot and RT-qPCR showed that the expression of TGF-β1 mRNA and protein decreased gradually after let-7a overexpression (P<0.05). The results of invasion and scratching showed that the overexpression of let-7a reduced the invasion and migration ability of LS-174T cells, and was negatively correlated with the initial virus concentration (P<0.05). Conclusion: Overexpression of let-7a can inhibit the invasion and metastasis of LS-174T cells by down-regulating the content of TGF-β1.

Related influencing factors of distal caries in the mandibular second molar induced by the mandibular third molar

Objective: To study the related factors of distal caries in the mandibular second molar induced by mandibular third molar, and provide theoretical foundation for the prophylactic removal of the mandibular third molar. Methods: The panoramic radiographs of 100 patients with distal caries of mandibular second molar and partially erupted mandibular third molar were reviewed at Hospital of Stomatology Wenzhou Medical University from December 2016 to December 2017. The distal caries in the mandibular second molar, age, gender, angulations, impaction degree, and the distance of the cementoenamel junction (CEJ) between the second molar and third molar were assessed. Results: In Chi-square test and univariate logistic regression analysis, age, angulation and CEJ distance were statistically significant (P<0.05). In the logistical regression analysis, age under 30 (OR=6.781, 95%CI=1.937-23.732, P=0.003) and the angulation between 40°-70° (OR=13.241, 95%CI=3.106-56.442, P=0.001) were statistically significant. Conclusion: Age under 30 and erupted mandibular third molar with an angulation between 40°-70° are risk factors for caries on the distal surface of the mandibular second molars, a prophylactic removal of lower third molars with the above-mentioned features might be advisable.

The role of four inflammatory factors in the relationship between periodontitis and coronary heart disease

Objective: To study the possible role of inflammatory factors (including IL-1β, IL-6, TNF-α, MCP-1) in the association between periodontal diseases and CHD. Methods: A total of 129 cases were selected from the Department of Stomatology of Wenzhou Chinese Medicine Hospital and the Cardiovascular Department of Wenzhou People’s Hospital from July 2015 to May 2016, including 39 healthy swarm (HC group), 35 cases of chronic moderate and severe forms of periodontitis (PD group), 31 cases of coronary heart disease alone (CHD group) and 24 cases of coronary artery disease along with periodontitis (PD+CHD group). The IL-1β, IL-6, TNF-α and MCP-1 in the serum of each recipient were detected by ELISA method. Results: There was statistical difference in IL-1β, IL-6, TNF-α, MCP-1 among enrolled groups (P<0.05) .The levels of IL-1β, IL-6, TNF-α and MCP-1 were highest in PD+CHD group, while the levels of IL-1β, IL-6 and TNF-α in PD group and CHD group were higher than those in HC group. Conclusion: The elevated serum levels of IL-1β, IL-6 and TNF-α in patients with moderate to severe periodontitis may promote the progression of CHD.

Effects of curcumol on proliferation, migration, invasion and apoptosis of human ovarian cancer SKOV3 cells

Objective: To investigate the effects of curcumol on proliferation, migration, invasion and apoptosis of human ovarian cancer SKOV3 cells. Methods: The CCK-8 method was used to detect the inhibition rate of SKOV3 cells at different concentrations (0, 5, 10, 25, 50, 100 μmol/L) for 24 h and 48 h, and the IC50 value was calculated. The scratch test and Transwell chamber method were used to detect the cells. Migration and invasion ability, apoptosis rate was detected by flow cytometry, Western blot was used to detect the effects of curcumol and PI3K/AKT pathway blocker LY294002 on the expression of PI3K, p-PI3K, AKT and p-AKT proteins. Results: Curcumol significantly inhibited the proliferation of SKOV3 cells in a concentration-dependent but time-independent manner, with an IC50 value of 48 μmol/L. Meanwhile, curcumol significantly inhibited the migration and invasion of SKOV3 cells and promoted apoptosis (P<0.01). The expression of total protein PI3K and AKT was decreased (P<0.05), and the expression of phosphorylated forms p-PI3K and p-AKT was significantly decreased (P<0.01). The combination of curcumol and LY294002 showed a good synergistic effect, which was more obvious than that of the curcumol alone in the down-regulation of p-PI3K, p-AKT (P<0.05). Conclusion: Curcumol can inhibit the proliferation, migration and invasion of SKOV3 cells and induce apoptosis, which is related to the inhibition of PI3K/AKT pathway.

Attenuation of lung ischemia-reperfusion injury by Alda-1 via activating mitochondrial acetaldehyde dehydrogenase in rats

Objective: To investigate the protective effect of Alda-1 activated mitochondrial acetaldehyde dehydrogenase 2 (ALDH2) on lung ischemia-reperfusion injury (LIRI) in rats. Methods: Twenty-four SD male rats were randomly divided as sham operation group (Sham group), ischemia-reperfusion group (I/R group) and Alda-1+ischemia-reperfusion group (Alda-1 group). The lung tissue and arterial blood samples were taken at the end of the experiment. The wet-to-dry weight ratio (W/D) of the lung tissue was measured; the alveolar structure was observed by HE staining; the malondialdehyde (MDA) and 4-hydroxynonenal (4-HNE) in plasma and lung tissue were detected; the content of ALDH2 was determined by Western blot and the activity of ALDH2 by ELISA. Results: Compared with the Sham group, the I/R group showed that the W/D value was significantly increased (P<0.05), the alveolar structure markedly destroyed, MDA and 4-HNE in the plasma and lung tissues remarkably improved (P<0.05), the activity of ALDH2 signally decreased (P<0.05). However, in the Alda-1 group, the W/D value was significantly decreased (P<0.05), the alveolar structure markedly improved, the MDA and 4-HNE of plasma and lung tissues remarkably decreased (P<0.05), and the activity of ALDH2 signally improved (P<0.05), compared with the I/R group. Conclusion: Alda-1 can accelerate the metabolism of aldehydes by activating ALDH2 to improve lung ischemia-reperfusion injury.

Mutation of a suspected Meckel-Gruber Pedigree: a multiple genetics analysis

Objective: To make a genetic analysis of a case of fetal tissue induced by clinically suspected Meckel syndrome so as to provide the family with genetic counseling and re-fertility suggestions. Methods: The comparative genomic hybridization (CGH) and whole exon sequencing (WES) technology was used for detecting the genetic mutation of the fetal tissues, and Sanger sequencing was used to verify the pathogenic sites of fetal parents and sisters. Results: Chromosomal microarray showed no obvious abnormality. WES showed the fetus carried the CC2D2A gene c.3964C>T and c.4567T>C compound heterozygous variation. Conclusion: CC2D2A gene c.3964C>T and c.4567T>C compound heterozygous mutations are the cause of Meckel syndrome in this family. Combination of different techniques can be applied to find disease-causing gene mutations and give a sufficient evidence for genetic consultation, prenatal diagnosis or pre-implantation genetic diagnosis.

The effects of leucine on liver regeneration after partial hepatectomy in rats

Objective: To investigate the effect of amino acid solutions with different leucine concentrations on liver regeneration after partial hepatectomy in rats. Methods: After 70% hepatectomy, eighteen SD rats were fed via gastric tube with one of the following amino acid solutions (1 g/kg): balanced amino acid (BAA group), branched chain amino acid (BCAA group) and leucine (Leu group). They were different in leucine concentrations. Liver regeneration was determined by residual liver/body weight ratio, ki-67 and PCNA positive counts 7 days later. Results: Compared with BAA group, the residual liver/body weight ratio, ki-67 and PCNA positive counts in Leu group were significantly increased (P<0.05), and were higher than those in BCAA group. Ki-67 and PCNA positive counts in BCAA group were significantly increased too when compared to BAA group, but residual liver/body weight ratio showed no significant difference between the two groups. Conclusion: Leucine can promote liver regeneration after partial hepatectomy in rats, and is more effective than branched chain amino acid.

Cytogenetic analysis of amniotic fluid on 4 943 high-risk pregnant women

Objective: To explore how different prenatal diagnostic indications are related to the frequency and type of abnormal karyotype of amniotic fluid cells in high-risk pregnant women. Methods: A retrospective analysis was made on the clinical data of 4943 cases receiving prenatal diagnosis because of various risk factors in our hospital. Results: A total of 182 cases were diagnosed as chromosomal abnormalities, with the detection rate being 3.68%. The highest abnormality rate was 90.47% in the non-invasive DNA high-risk group, and in the other 12 cases the abnormality rate was 75% after amniocentesis due to non-invasive DNA indicated sex chromosomal abnormalities. The abnormal rate was 7.61% in the ultrasound abnormal group, 3.43% in the elderly group, 3.09% in the down-screening 21-trisomy high-risk group, 2.28% in the down-screening 18-trisomy high-risk group, and 0.37% in the adverse pregnancy history group. The non-invasive DNA high-risk group had the highest chromosomal abnormality rate, followed by the ultrasound abnormality group. Conclusion: The non-invasive DNA could not completely replace the amniotic fluid cell karyotype analysis, which was considered the gold standard for detecting the fetal chromosomal abnormality. The fetal chromosomes in elderly pregnant women were prone to mutations. It is recommended that prenatal diagnosis be performed directly, and meanwhile, extra gene chips are recommended for those with ultrasound abnormalities or the elderly pregnant women.

Clinical characteristics and prognosis of different molecular classification bladder cancer

Objective: To analyze the clinical characteristics and prognosis of patients with different molecular classification bladder cancer by taking for example the classification of bladder cancer cell division cycle related factor 3 (CDCA3). Methods: Based on the American Cancer Genome Atlas (TCGA) database, 147 copies of bladder cancer CDCA3 gene expression profiles and related cases were downloaded. The CDCA3 expression levels were ranked from low to high to find the median. The samples were divided as CDCA3 low expression group (74 cases before the median) and CDCA3 high expression group (73 cases after the median). The clinical features and prognosis were compared between different CDCA3 genotype bladder cancer patients, and the mechanism of CDCA3 in the development and progression of bladder cancer was analyzed. Results: The proportion of patients with myometrial invasion, the stage of T stage and N stage, and the proportion of tumor metastasis in CDCA3 high expression group were higher than those in CDCA3 low expression group, and the tumor differentiation degree was lower than that in CDCA3 low expression group with significant difference (P<0.05). The recurrence rate of CDCA3 high expression group was higher than that of CDCA3 low expression group at 1 year and 3 years, with significant difference (P<0.05). The 1-year and 3-year survival rates of the CDCA3 high expression group were lower than those of the CDCA3 low expression group with significant difference (P<0.05). GSEA found that over-expressed CDCA3 bladder cancer cells were enriched with G2M checkpoints, glycolysis, E2F signaling pathway, MYC signaling pathway, MTOR signaling pathway, PTK pathway, PI3K-AKT-mTOR signaling pathway, mitotic spindle, unfolded protein Gene sets such as reaction, cholesterol protein, and spermatogenesis. Conclusion: The lower the CDCA3 gene expression level, the lower the tumor stage, the smaller the risk of tumor metastasis, the higher the degree of differentiation, the better the prognosis, and the lower the recurrence rate and mortality. The role of CDCA3 gene is achieved through the regulation of multiple gene pathways. CDCA3 gene may be a potential biomarker for bladder cancer, which can be evaluated by an analysis of its expression level and prognosis.

Clinical study on the treatment of mixed hemorrhoids by automatic hemorrhoid puncture combined with cod liver oil sodium injection

Objective: To study the efficacy of automatic hemorrhoid ligation (RPH) combined with cod liver oil sodium injection in the treatment of mixed hemorrhoids. Methods: Select our hospital in March 2016-March 2017 admitted to the hospital, which is mainly composed of hemorrhoids hemorrhoids or mixed hemorrhoid patients of 200 cases were randomly divided into treatment group (automatic hemorrhoids ligation+ cod liver oil acid sodium injection) and control group (automatic hemorrhoids ligation), 100 cases in each group, observe two groups of postoperative pain, aprons early fall off, infection, hemorrhage, anal fall swelling and tenesmus, recurrence, etc. Results: Compared with control group, treatment group aprons early fall off, bleeding, postoperative recurrence is better than that in control group, two groups compare the difference was statistically significant (P<0.05), the treatment group patients in postoperative pain, anal fall swelling and tenesmus, urinary retention, the respect such as blood clots compared with control group, there was no statistically significant difference (P>0.05), two groups of patients were not seen in postoperative serious complications such as infection, anal stenosis, anal incontinence. Conclusion: Automatic hemorrhoid ligation combined with cod liver oil sodium injection for the treatment of mixed hemorrhoid can reduce apron early fall off, bleeding, postoperative recurreuce and improve clinical efficacy.

Application of beard combined with hair transplantation in the treatment of severe androgenic alopecia

Objective: For the treatment of androgenetic alopecia patients above grade V with severely deficient occipital donor area, the therapeutic effect of beard combined with hair transplantation was investigated. Methods: A total of eighteen patients with androgenetic alopecia above grade V from the First People’s Hospital of Hangzhou from July 2016 to July 2018 were enrolled. The beard and occipital hair from donor area were obtained by follicular unit extraction (FUE) and transplanted into the alopecia area. Results: All the eighteen patients were followed up for the next 12 months. The survival rate of transplanted beard was as high as 90%. Seventeen patients were very satisfied after operation, and most of the bald areas were covered. Conclusion: For patients suffering from androgenic alopecia above grade V with severe deficiency of donor area, beard combined with hair transplantation has achieved satisfactory results, so that beard can be used as a new alternative to supplement donor area.
