625 The effect of curcumin analogue L6H3 on striatum monoamine neurotransmitter in the rat model of Parkinson disease

Objective: To investigate the effect of curcumin analogue L6H3 on seven monoamine neurotransmitters and metabolite in the striatum of rats with Parkinson disease (PD) by micro-dialysis sampling coupled with high performance liquid chromatography and electrochemical detection (HPLC-ECD). Methods: PD model was established by injecting 6-OHDA into the right striatum of rats, then verified by the rotational behavior after intraperitoneal injection of apomorphine solution. Rats were randomly divided to Sham group, PD group and PD+L6H3 group. The microdialysis sampling coupled HPLC-ECD was developed and applied to detect the level of monoamine neurotransmitters and metabolite in rat striatum. Results: 6 weeks (2 weeks after administration),7 weeks (1 week after stopping administration) after model establishment, the levels of DA, DOPAC, HVA, NE, E, 5-HT, 5-HTAA in PD+L6H3 group were significantly increased compared with PD group (P<0.01). Conclusion: L6H3 has the capability to up-regulate monoamine neurotransmitters and metabolite in the striatum of PD rat model at both 2 weeks after administration and 1 week after stopping administration.

630 Isolation and culture of hepatocytes and sinusoidal endothelial cells and preparation of hepatocyte scaffolds

Objective: To explore the methods of isolation and culture of rat hepatocytes and sinusoidal endothelial cells and preparation of hepatic acellular scaffolds. Methods: Rat hepatocytes isolated from liver tissues were digested by collagenase IV and purified in culture medium after washing, filtration and centrifugation. Cell immunohistochemistry was used to detect the cells after culture. Hepatic sinusoidal endothelial cells were isolated by Percoll density gradient centrifugation and purified in culture medium before they were detected by immunofluorescence. The scaffolds of rat hepatocyte acellular scaffolds were prepared by circulating perfusion and acellular treatment. The scaffolds were confirmed by scanning electron microscopy. Results: The isolated rat hepatocytes were round, full and clear under microscope. Immunohistochemical staining of cytokeratin 18 in hepatocytes revealed brown granules in the cytoplasm. Thehepatic sinusoidal endothelial cells were spindle-shaped and the nucleus was located in the center. Endothelin-1 immunofluorescence staining showed red signal on the cell surface. The scanning electron microscopy of the rat liver decellularized scaffold showed that the extracellular matrix was retained, and no nuclear and cytoplasmic components were found. Conclusion: Hepatocytes and sinusoidal endothelial cells are successfully isolated and hepatic acellular scaffolds were prepared.

634 Role of TAZ in osteogenic differentiation of periodontal ligament cells under tensile stress

Objective: To explore the role of PDZ binding transcriptional activator of transcription (TAZ) in tensile stress-induced cytoskeleton changes of human periodontal ligament cells (hPDLCs) to promote osteogenic differentiation. Methods: hPDLCs were obtained from PDL tissues in intact extracted third molars. Flexcell system was applied to construct the tensile stress loading model in vitro. Changes in TAZ and osteoblastic markers Cbfα1, collagen I (COL I), osteocalcin (OCN) and osterix (OSX) in hPDLCs were detected by Western blot and PCR. And the connection between TAZ and Cbfα1 was detected by co-immunoprecipitation. TAZ siRNA transfection was used to determine the role of TAZ in bone differentiation induced by tensile stress. Results: TAZ, Cbfα1, COL I, OSX and OCN mRNA and protein levels increased after 0.5 Hz and 12% tensile stress was applied to hPDLCs for 24 h and 48 h. There was a much more obvious connection between TAZ and Cbfα1 under tension for 24 h, compared with the control group. TAZ decreased after TAZ siRNA transfection in hPDLCs. Western blot results indicated that Cbfα1, COL I, OSX and OCN increased by tension were down-regulated after TAZ siRNA interference. Conclusion: TAZ induces osteogenic differentiation of human periodontal ligament cells by up-regulating Cbfα1 expression and transcriptional activity during tensile stress.

639 Sedative effect of Jiao-Tai-Wan with different concentrations and compositions on mice

Objective: To compare sedative effects of Jiao-Tai-Wan with separated prescriptions of Coptis and Cinnamon on mice. Methods: Jiao-Tai-Wan groups were intragastric administrated 7 days by Jiao-Tai-Wan respectively with high, medium and low dosage of 0.220, 0.110 and 0.055 g/mL. Coptis groups were respectively administrated 7 days by Coptis with high, medium and low dosage of 0.200, 0.100 and 0.050 g/mL. Cinnamon high dose group was administrated 7 days by Cinnamon with concentration of 0.030 g/mL. Positive control group was administrated by Zolpidem with concentration of 0.200 g/mL. Blank control group was injected with water. The locomotor activity meter was used to determine the total number of locomotor activities of mice within 15 min, 30 min, 1 h, 3 h, 12 h, 24 h after administration from day 1 to day 7. The number of locomotor activities of mice on day 8 without administration continued to be monitored. Results: On day 1 within 30 min, 1 h, 3 h, 12 h, 24 h after administration, the number of locomotor activities of Jiao-Tai-Wan group with high dose was statistically declined respectively compared with blank control group (P<0.05). These differences were observed again on day 3, day 5, day 7. These differences remained on day 8 without administration. On day 3, day 5, day 7 within 1 h, the number of locomotor activities of Jiao-Tai-Wan group with medium dose were statistically declined respectively compared with blank control group (P<0.05). On day 7 within 1 h, the number of locomotor activities of Jiao-Tai-Wan group with low dose was statistically declined comparing with blank control group (P<0.05). On day 3, day 5, day 7 within 15 min and 1 h, the frequency of locomotor activity of Coptis groups with high dosage and medium dosage were statistically declined respectively compared with blank control group (P<0.05). On day 7 within 3 h, 12 h and 24 h after drug administration, the frequency of locomotor activity of Coptis group with high dosage was statistically declined c respectively omparing with blank control group (P<0.05). Conclusion: Jiao-Tai-Wan can produce a sedative effect, in which Coptis plays a major role.

644 The effect of geniposide on mice with diabetic nephropathy

Objective: To investigate the effect of geniposide on diabetic nephropathy. Methods: Diabetic mice were established by streptozotocin and randomly divided as model group, geniposide low dose group (100 mg/kg), high dose group (200 mg/kg) and normal control group. The model group and the normal group were given normal saline, and the geniposide dose groups were given the corresponding dose intragastrically for 8 weeks. The living conditions of the mice were observed and recorded. The body weight and blood glucose were measured. The pathological changes of renal tissue were observed by HE staining, and the expression of TNF-α, IL-6 and IL-1β mRNA were detected by RT-qPCR. Results: Compared with the normal control group, the model group had poorer life quality, slower weight gain (P<0.01), increased blood glucose (P<0.01), and obviously damaged glomerular structure shown by pathological staining. A large number of interstitial inflammatory cells were infiltrated, and the expression of TNF-α, IL-6 and IL-1β in renal tissue was increased, as detected by RT-qPCR assay (P<0.01). Compared with the model group, the mice in the low and high dose groups had a better living conditions, more rapid increase of the body weight (P<0.05), significantly decreased blood glucose (P<0.01) but improved pathological morphology of renal tissue and decreased expression of TNF-α, IL-6 and IL-1β in renal tissue (P<0.01). Conclusion: Geniposide can improve the symptoms of diabetes in mice and protect the kidney tissues. Its regulatory mechanism may be related to the improvement of inflammation in diabetic mice.

649 Intervention of zinc on the male reproductive toxicity caused by phthalates exposure

Objective: To study the protective effect of zinc on reproductive toxicity caused by phthalate exposure in mice. Methods: Thirty-six male ICR mice were randomly divided into the control group, MIXPs (the equipotent toxic mixture of butyl benzyl phthalate (BBP), di (n-butyl) phthalate (DBP) and di (2-ethyhexyl) phthalate (DEHP) group, and MIXPs+ZnSO4 group, with 12 mice in each group. Mice in MIXPs group and MIXPs+ZnSO4 group were treated with 160 mg·kg-1·d-1 MIXPs by gavage with or without 50 mg·kg-1·d-1 ZnSO4. Mice in control group were treated with isodose 0.5% CMC-Na. After 90 days’ intervention, orbital vein blood was collected from each anesthetic mouse. Then dissect the mouse and take male reproductive organs such as the testicles and epididymis. Sperm morphology was observed with electron microscope. The levels of testosterone, SOD, MDA and T-AOC were measured with the commercial kits. Results: Compared with the control, the bodyweight, anogenital distance, serum testosterone, SOD and MDA in testicular tissue in MIXPs group were significantly reduced, while the rate of sperm deformation was significantly increased (P<0.05 or P<0.01), but there were no significant changes in MIXPs+ZnSO4 group. Conclusion: Zinc can inhibit the male reproductive toxicity induced by MIXPs exposure, which may be related to the improvement of the antioxidant level of testicular tissue and the increase of serum testosterone level.

654 An analysis of multi-modality CT imaging and high-risk clinical factors of chronic cerebral hypoperfusion

 Objective: To evaluate the microcirculation and hemodynamic changes in patients with chronic cerebral hypoperfusion (CCH) via one-stop low-dose multi-modality CT including CT perfusion (CTP) and CT angiography (CTA), and to analyze the clinical risk factors. Methods: Whole-brain CTP imaging was performed on 266 patients with suspected diagnosis of CCH using 640 multi-slice CT. Following the post-processing, cerebral blood flow (CBF), cerebral blood volume (CBV), time to peak (TTP), delay time (DT) and Four-dimensional CTA images were acquired. The relative perfusion parameters (rCBF, rCBV, rTTP and rDT) were calculated and the stenosis degree of brain arteries was evaluated. The perfusion parameter differences between two hemispheres were analyzed by comparative t-test; the perfusion parameter differences between hypoperfusion group and normal perfusion group were analyzed by independent t-test; the difference on stenosis degree of supplying blood vessel between hypoperfusion group and normal perfusion group were analyzed by the rank sum test and the clinical risk factors were analyzed by Chi-square test and logistic regression. Results: ①In the hypoperfusion group, 184 patients demonstrated perfusion asymmetry on both hemispheres. CBF and CBV of the affected side were lower than that of contralateral side, while TTP and DT were prolonged, the differences of all perfusion parameters between two sides had statistical significance (P<0.01). Among these cases, 98 cases had anterior circulation insufficient, 61 cases had posterior circulation insufficient, and 25 cases had multi-region circulation insufficient. ②rCBF and rCBV of the hypoperfusion group were lower than that of the normal perfusion group, while rTTP and rDT were higher than the latter. The perfusion parameters of CCH group were 0.75±0.44, 0.76± 0.56, 1.16±0.22, and 1.56±0.34; the perfusion parameters of normal perfusion group were 0.93±0.51, 0.92±0.09, 1.03±0.07, and 1.06±0.17. The difference between two groups had statistical significance (t=-3.943, -3.862, 2.790, 4.558, P<0.01). ③In 184 cases of hypoperfusion, 24 cases were mild stenosis, 47 cases moderate stenosis, 65 cases severe stenosis and 48 cases occlusion on CTA. Among 82 normal perfusion cases, 54 cases were mild stenosis, 21 cases moderate stenosis, 5 cases severe stenosis and 2 cases occlusion on CTA. The differences in artery stenosis between two groups had statistical significance (P<0.01). ④The incidence of diabetes, hyperlipidemia, and smoke was statistically different between hypoperfusion group and NCCH group (P<0.05). Confirmed by multi-factor logistic regression analysis, smoke and hyperlipidemia were high risk factors of hypoperfusion. Conclusion: Multi-modality CT imaging can not only demonstrate the degree of cerebral artery stenosis, but also directly and quantitatively reflect the microcirculation perfusion of brain tissue in CCH patients, and provide an objective imaging basis for the diagnosis and prevention of CCH.

661 Mechanism of liraglutide polarized M2 macrophage in improving nonalcoholic fatty liver disease

Objective: To investigate the potential mechanism of liraglutide in improving nonalcoholic fatty liver disease (NAFLD) associated with type 2 diabetes. Methods: A mouse model of type 2 diabetes with NAFLD was constructed by high-sugar and high-fat diet with a low dose streptozoticin. The mice were randomly divided into 3 groups, normal group, model group and liraglutide group, with 15 mice in each group. The mice in liraglutide group were injected subcutaneously with liraglutide (100 μg/kg) once a day, lasting for 4 weeks. Body weight of each mouse and their liver index, ALT, AST and TG were measured. Pathological changes of liver tissue were observed by HE staining and oil red “O” staining. The levels of interleukin-4 (IL-4) and IL-10 in serum were detected by ELISA. The proportion of M2 macrophages was detected by flow cytometry. The expressions of IL-4, IL-10, CD206, CD301, MGL-1, MGL-2 and arginine-1 mRNA were examined using RT-PCR. Results: Compared with the model group, the body weight, liver index, TG, AST and ALT of the mice in liraglutide group were decreased significantly, the fatty degeneration in the liver tissue was down-regulated, while the proportion of M2 macrophages had an obvious rise (P<0.001), and the expressions of IL-4 and IL-10 were increased notably (P<0.05). Additionally, the expressions of IL-4, IL-10, CD206, CD301, MGL-1, MGL-2 and Arg-1 mRNA were significantly up-regulated in liraglutide group (P<0.01). Conclusion: Liraglutide has protective effect on NAFLD in type 2 diabetes, possibly by promoting the expression of IL-4 and IL-10 to polarize M2 macrophages so that the mechanism of NAFLD get improved.

667 Quantitative evaluation of benign and malignant nodules of breast by diffusion-weighted imaging combined with T2WI texture analysis

Objective: To investigate the value of apparent diffusion coefficient (ADC) and T2-weighted imaging (T2WI) texture parameters in quantitative evaluation of benign and malignant breast nodules. Methods: The retrospective study was based on breast ADC and T2WI images. A total of 82 patients with histopathologically confirmed malignant or benign breast nodules from Lishui Municipal Central Hospital were enrolled in this study. Texture features from region of interest (ROI) of breast nodules were extracted from ADC map and T2WI images. Several first-order and second-order texture parameters were generated automatically by post-processing software. The difference of texture parameters between the two groups were compared by Student’s t-test or Mann-Whitney U test. The receiver operating characteristic curves (ROCs) were generated based on the significant variables identified from the univariate analysis, and the area under the curve (AUC), sensitivity, and specificity for nodules differentiation were reported. Z test was used for multiple comparisons of the AUCs with statistical differences between malignant and benign nodules groups. Results: ADCmean, ADCmedian and ADCmin values in breast benign nodule group were significantly higher than those in malignant nodule group (all P<0.05). The skewness and entropy of breast malignant nodule group were significantly higher than benign nodule group (all P<0.05). ROC showed that the thresholds of ADCmean, ADCmedian, ADCmin, skewness and entropy were 1.34×10-3 mm2/s, 1.41×10-3 mm2/s, 0.97×10-3 mm2/s, -0.075 and 3.762, respectively; and AUCs were 0.90 (95%CI= 0.82-0.95), 0.89 (95%CI=0.80-0.95), 0.91(95%CI=0.83-0.96), 0.74 (95%CI=0.64-0.83), 0.95 (95%CI=0.87-0.98), respectively. Comparison of ROCs showed that entropy had the best diagnostic efficiency, and the sensitivity and specificity were 80.55%, 94.74%. AUCs of entropy from ADC, T2WI and both combination showed that the ADC combined with T2WI had the largest entropy AUC (AUC=0.97). Conclusion: Texture parameters based on ADC combined with T2WI images can provide useful proof for differentiating benign breast nodules from malignant, especially for the second-order texture parameters entropy which is expected to provide more diagnostic value for tumor imaging in clinical practice.

673 The effect of electroacupuncture on synaptic plasticity protein in hippocampus of chronic stress depression model rats

Objective: To investigate the effects of electroacupuncture on the expression of synaptic protein in hippocampus of chronic stress depression rats, and to clarify its therapeutic mechanism. Methods: Forty-eight SD rats were randomly divided into 4 groups: blank control group, model group, electroacupuncture group and fluoxetine group. In addition to the control group, chronic unpredictable mild stress (CUMS) was used to establish a model of depression. The electroacupuncture group was treated with electroacupuncture once a day at the two points of “Baihui” and “Yintang” before daily stress; the fluoxetine group was given a positive drug fluoxetine solution before the modeling, with the dose of 10 mg/kg for 21 d. On the 7th, 14th and 21st day of stress modeling, the depression-like behaviors of rats were evaluated by open field test and forced swimming test. The pathological features of hippocampal synaptic structure were observed by Golgi staining. The hippocampal synaptic plasticity protein SYN, PSD-95, CREB and BDNF was detected by Western-blotting test. Results: Compared with the control group, the forced swimming time of the model group was significantly increased at 7 d (P<0.05), and the spontaneous activity of the open field was significantly decreased at 14 d (P<0.05), and the depression-like behavior was obvious. The structural damage, synaptic plasticity protein SYN, PSD-95, CREB, BDNF expression were significantly down-regulated (P<0.01); after electroacupuncture treatment, the depression-like behavior of rats was significantly improved (P<0.01), dendritic spine morphology The number of SYN, PSD-95, CREB and BDNF increased significantly (P<0.01). Conclusion: Electroacupuncture can significantly improve the depression-like behavior of chronic stress-depressive rats, which is related to the improvement of synaptic structure and up-regulation of synaptic plasticity protein expression.

677 Establishment of the reference interval of platelet parameters in pregnant women

Objective: To explore the variation of platelet count (PLT), immature platelet fraction (IPF), absolute value of immature fraction (IPF#), high-fluorescence intensity of immature platelet fraction (H-IPF), and mean side fluorescence intensity of platelet (PLT-X) in the pregnant women of different gestation, then to establish the reference intervals of these parameters. Methods: 1 272 normal pregnant women (20 to 30 years old) from October 2016 to October 2018 were selected, and they were divided into three groups: 348 cases of early trimester group (≤12 weeks), 369 cases of second trimester group (13 to 27 weeks) and 555 cases of third trimester group (≥28 weeks), second and third trimester group included 924 cases. Also, 200 cases of non-pregnant women of 20 to 30 years old were included as control group. At the same time, 88 cases of pregnant women with pre-eclampsia and 100 cases of gestational diabetes were collected. The PLT, IPF, IPF#, H-IPF and PLT-X of peripheral vein blood were determined by Sysmex XE-5000 and the reference intervals were established by using 95% confidence interval. Results: The PLT of the pregnant women was lower than the control group (P<0.05), but the parameters of IPF, IPF#, H-IPF and PLT-X were higher than the latter. The IPF, IPF#, H-IPF and PLT-X increased with the increase of gestational weeks in the early, second and third trimester groups (P<0.05). There was no statistically significant difference in PLT and H-IPF between early and second trimeter groups (P>0.05), but there were significant differences in PLT and H-IPF between early and second trimeter groups with third trimeter one (P<0.05). The PLTs of the pregnant women with pre-eclampsia and gestational diabetes were lower than the second and third trimester group, but the parameters of IPF, IPF#, H-IPF and PLT-X were higher than the latter. Conclusion: Platelet related parameters varied with the development of pregnant women during pregnancy, and the platelet parameters reference intervals during different trimeters of pregnancy were the basis for assessing the pathological state of pregnancy.

681 Diagnostic value of combining of prostate imaging reporting and data system and PSA density in clinically significant prostate cancer: Meta-analysis

Objective: To systematically review the diagnostic value of prostate imaging reporting and data system (PI-RADS ) plus PSA density (PSAD) in clinically significant prostate cancer (CSPCA) by meta-analysis so as to provide evidence-based information for clinical. Methods: We electronically searched databases from inception to Sep 30, 2018, including PubMed, Embase, EBSCO, Cochrane Library, CNKI, CBM, and VIP to collect Chinese and English diagnostic articles on combined of PI-RADS and PSAD for CSPCA published in China and abroad. Two reviewers independently screened literature and extracted data. The included articles were assessed in term of methodological quality by QUADAS tool and analyzed with meta software in Stata10.0, then the relevant parameters were extracted. Results: A total of 6 articles met the inclusion criteria, 1 228 patients were included. The fixed effect model was used for lack of heterogeneity confirmed by Q and I2 test. The pooled sensitivity of PI-RADS for CSPCA was 81.1% [95%CI: 0.639-0.912], and the specialty, DOR, positive likelihood ratio (+LR), negative likelihood ratio (-LR) and Lambda value were 82.1% [95%CI: 0.694-0.903], 19.711 [95%CI: 6.953-55.927], 4.533 [95%CI: 2.552-8.054], 0.229 [95%CI: 0.113-0.468] and 3.011 [95%CI: 1.967-4.054] respectively. The specialty, DOR, positive likelihood ratio (+LR), negative likelihood ratio (-LR) and Lambda value were 87.9% [95%CI: 0.826-0.917], 81.8% [95%CI: 0.590-0.872], 22.688 [95%CI: 7.403-69.538], 3.632 [95%CI: 1.941-6.800], 0.160 [95%CI: 0.095-0.270] and 3.778 [95%CI: 2.537-5.020] for PI-RADS plus PSAD. The Lambda and sensitivity of PI-RADS plus PSAD were significantly higher than those of PI-RADS (P<0.05). Conclusion: PI-RADS plus PSAD could effectively improve the diagnosis efficiency for CSPCA.

Objective: To investigate the relationship between the aspect ratio (AR) of the anterior communicating artery (AComA) aneurysm and aneurysm rupture. Methods: A retrospective analysis of 650 consecutive patients with single AcoA aneurysms between October 2010 and January 2016 in the First Affiliated Hospital of Wenzhou Medical University was conducted. There were 578 patients with ruptured AcomA aneurysms and 72 with un-ruptured AcomA aneurysms. The diameter of perpendicular height and aneurysm neck was re-measured using CT angiography images and the relationship between the AR and aneurysm rupture status was analyzed. Results: In ruptured aneurysm group, the perpendicular height was (3.57±1.96) mm, the neck size was (3.07±1.17) mm, and the AR was 1.24±0.67. In un-ruptured aneurysm group, the perpendicular height was (2.59±1.85) mm, the neck size was (2.71±1.15) mm, and the AR was 0.97±0.60. The perpendicular height, neck size and AR of ruptured aneurysms were larger than the unruptured, with significant difference (P<0.05) between them. The AR of AComA aneurysms was analyzed by ROC curve. The area under the curve was 0.64, and the optimal cut-off value was 0.935. Conclusion: The AR size was significantly related to the rupture of AComA aneurysms. In clinical practice, the AR could be considered a reference factor that may aid in risk evaluation of these patients.

692 The effect of Venturi mask as a wound protection for boys with circumcision

Objective: To evaluate the effect of Venturi mask as a wound protection for boys with circumcision. Methods: Eighty-six boys undergoing elective circumcision were randomly divided into two groups. After circumcision, boys in Group A (control group) received traditional medical care and boys in Group B (Venturi mask group) received an additional Venturi mask for wound protection. The following indexes were recorded and compared between the two groups: 1) the comfort values of the time-points when sent back to the ward, 12 hours and 24 hours after surgery; 2) the status of wound infection; 3) the length of hospital stay; and 4) the status of complications. Results: The comfort values were nearly the same in the two groups when they were sent back to the ward after surgery (P>0.05). However, boys got higher comfort values in 12 hours and 24 hours after surgery in the Group B than the Group A (both P<0.05). No postoperative infection was found in either group. Children stayed in hospital longer in the Group A than the Group B (P<0.05). No complication was observed in either group. Conclusion: The application of Venturi mask for wound protection with boys undergoing circumcision could improve the postoperative comfort value and shorten the hospital stay.
