141 Association analysis of CDKALl rs7756992 polymorphism with the clinical characteristics and susceptibility to type 2 diabetes meilitus in Han Chinese

Objective:To investigate the association of CDKALI rs7756992 with type 2 diabetes mellitus rT2DM)in Chinese Han population and investigate correlation of the clinical traits related with one certain genotype for further screening and clinic genotyping.Methods:534 T2DM patients and 453 healthy controls from Wenzhou,Zhejiang province were involved in this study.High—resolution dissolving the curve(HRM)and DNA sequencing were used to identify the alleles and genotypes of CDKALI rs7756992,The frequencies of allele and genotype distribution in both groups were analyzed with unconditional logistic regression method.Plus,the clinical traits and complication in different genotypes were described and compared.Results:There was a significant difference in GG/AA gene model(P=0.048)and the frequency of rs7756992(G)(P=0.045)between T2DM and controls.It showed that G was the risk allele for T2DM.It was found that the GG genotype showed lower fasting glucose concentration compared to either AA or AG genotype,even with sex and age adjusted(P<0.01).No significant difference found in HbAlc value.In addition,there was no significant difference statistically between allele and genotype frequencies and the chronic complications of diabetes(P>0.05).Conclusion:The present study reveals that CDKALl rs7756992 is associated with T2DM in Chinese Han population.In addition,it still should be noted that the role of this gene may regulate the blood glucose.

147 Experimental study of microporous acellular dermal matrix and mesenchymal stem cells composite grafttor remodeling the cutaneous defect of nude mice

Objective:To investigate the healing effect and the remodeling practicability of skin function of the composite graft consisting of laser micropore porcine acellular dermal matrix(LPADM)and mesenchymal stem cells(MSCs)on full—thickness skin wounds.Methods:The composite grafts were prepared by seeding the MSCs on the LPADM(MSCs—LPADM)or the normal porcine acellular dermal matrix(MSCs—PADM).0ne fullthickness wounds(2 cm x 2 cm)were created on each back of l 8 nude mice and were randomly divided into 3groups(n=6).Then the MSCs—LPADM were transplanted synchronously with the split—thickness autograft on
the wounds(group A).The mice treated with LPADM(group B)or MSCs—PADM(groups C)were considered as control.The gross wound healing status was examined.Tissue samples were collected at day 3,7 and 14 post—operation for histological examination and transmission electron microscopy(TEM)obeservation.Results:H&E staining showed the appearance of the cavity—like vascular structure in group A and B at day 3 post—operation,but not in group C.At day 7,the grafts of A and B groups survived well and new vessels can be seen evidently in the micropores.However,grafts necrosis occurred in group C with pallor or black tissue.At day 14,all the grafts survived in A and B groups and abundant blood vessels in the micropores of the matrixs were observed as well,while in group C,no blood vessel appeared and all the grafts went through necrosis and exfoliation.Through TEM observation,we found newly generated unmyelinated nerve—like structure in group A with visible sebaceous gland—like cells in the superficial layer and single sebaceous gland—like cells in the deep layer of the demal matrix,which can not be seen in group B and C.Conclusion:The composite graft(MSCs—LPADM)has better healing effect and provides a good microenvironment for the exogenous hMSCs to proliferation and differentiation,thus may have agreat potential for use in wound reconstruction in the future.

161 Antidepressant.1ike effect of trans.resveratrol on behavioral changes in ovariectomized mice and the possible mechanism

Objective:To explore the antidepressant--like effects of trans··resveratrol on behavioral changes in ovariectomized mice and its possible mechanism.Methods:Sstablished were Perimenopausal model by ovariectomy and depression model by chronic unpredictable stress were established after three weeks’administration.Use forced swimming test(FST)and open field test to observe the behavioral changes in mice,the expression of brain—derived neurotrophic factor(BDNF)and B—cell lymphoma protein 2(Bci-2) protein in hippocampus were determined with immunohistochemistry,the levels of superoxide dismutase (SOD),glutathione(GSH)and malondiaIdehyde(MDA)were detected.Results:Compared with OVX group,the immobility time in FST and MDA level were increased,activities in open field test were decreased,BDNF and Bcl一2 expression,the activity of SOD and GSH level were reduced in OVX+Stress group.Transresveratrol( 30 mg。kg一1’d。1 or 60 mg’kg一1‘d一1)could reverse these effects mentioned above.Conclusion:Trans—resveratrol can ameliorate depressive—like behaviors in ovariectomized mice,and the mechanism may be related to its neuroprotective and antioxidant effects.

166 The role of caveolin.1 and p421p44MAPK in the airway remodeling of asthmatic rats and the influence of roxithromycin on its expression

objective:To explore the role of caveolin一1 and p42/p44MAPK in the airway remodeling of asthmatic rats and the intervention of Roxithromycin.Methods:Thirty—two healthy.male Sprague—Dawley rats were randomly divided into four groups averagely:control group(group C),asthmatic group(group A),Dexamethasone group(group D)and Roxithromycin group(group R).Asthmatic rats model was duplicated by the sensitization and provocation with ovalbumin.The total bronchial wall thickness(Wat/Pbm)and the bronchial smooth muscle thickness(Wam/Pbm)were measured with image—pro plus 6.0.The protein expressions of caveolin一1 and p-p421 p44MAPK in the airway smooth muscle were measured with Western blot.Results:Wat/Pbm,Wam/Pbm of group A were significantly higher than those ofgroup C(P<0.01),those ofgroup D and group R were lower than those of group A(P<0.01),nevertheless,higher than those of group C(P<O.05).The expression of caveolin一1 in group A was significantly lower than that of group C(P<O.01),that of group D and group R were significantly higher than that of group A(P<0.01),but decreased compared with group C(P<0.05);the expression of p-p42/p44MAPK in groupA significantly increased compared with group C(P<0.01),that of group D and group R were significantly decreased than that of group A(P<0.01),but still increased compared with group C(P<0.01).There were significantly negative correlation between Wam/Pbm and the protein expression of caveolin—l(r=一0.893,P<0.01).There were significantly positive correlation between Wam/Pbm and the expression of P—D42/I:144MAPK measured with Western blot(r=-0.918.P<0.01).There was significantly negative correlation between the expressions ofcaveolin一1 and P—p42/p44MAPK measured with Western blot(r=.0.873,P<O.0 1).Conclusions:There may be some connections between caveolin一1 and 042/044MAPK signaling in airway remodeling of asthmatic rats.and Roxithromycin maybe inhibits airway remodeling partly through caveolin一1 and p42/p44 MAPK signaling pathway.

175 Effects of IFN-Y on the expression of transcription factors GATA3 in rat pulmonary fibrosis

Objective:To explore the effects of IFN—v on the expression of transcription factors GATA3 in rat pulmonary fibrosis.Methods:Male SD rats were randomly divided into three groups:the control group(C),bloemycin groups(group BLM)andIFN—Y treated group(group BLM—in).The pulmonary fibrosis model was established by intraperitoneal injection with bloemycin(15 mrdkg X 10 day).Rats in BLM and BLM—IFN group were randomly sacrificed on 7山day(group BLMl and BLM—IFNl),14山day(group BLM2 and BLM-IFN2)and 280 day(group BLMd and BLM—IFN4),respectively。Semi—quantitative analysis of the histopathologic changes stained with Haematoxylin-eosin(H-E)was performed,and the mRNA expression of transcription factors GATA3 in each group was measured with RT-PCR.Results:①alveolitis was the primarily pathologic change of lung tissue in BLMl and BLM·IFNl,while alveolitis and fibrosis in BLM2 and fibrosis in BLM4.Obviously alleviated fibrosis was seen in both BLM—IFN,and BLM—IFN4 group.②As compared with group C,the expression of GATA3 gene was significantly higher in each bleomycin group,(P<0.0 l respectively).After treatment with IFN-V,the expression of GATA3 gene in BLM—INFl,BLM-INF2 and BLM·INF4 was lower than that in counterpart group of BLMs.(P<O.01 respectively),even lower than that in group C.Conclusion:IFN—v treatment can reduce the degree of rat pulmonary fibrosis induced by bloemycin.The mechanism may be that the IFN—v can inhibit the expression of GATA3 gene,and reverse the imbalance of Th l/Th2.

187 Clinical and pathological analyse for renal injury of IgA nephropathy with thrombotic microangiopathy secondary to malignant hypertension

0bjective:To evaluate the characteristic of clinical and pathological of IgA nephropathy with thrombotic microangiopathy(TMA)secondary to malignant hypertension patients.Methods:From Feb.2004 to Dec.20 1 1,a case control retrospective study was carried out in 5 cases of IgAN with TMA which secondary to malignant hypertension and 6 cases of TMA secondary to malignant hypertension as contr01.Results:Clinical manifestation of two groups showed that both groups had malignant hypertension,hematuria,proteinuria and renal injury.The IgAN group had significantly higher proteinuria and hematuria(P<0.05,VS un-IgAN group);kidney pathology of 1 l cases showed that the IgA group possessed severer glomerular pathological changes and the degree of pathological changes of glomerular artery was inferior than un—IgAN group.Conclusion:IgA nephropathy with TMA secondary to malignant hypertension is different from that without IgAN in clinical andpathological manifestation.When it is difficult to diagnosis,renal biopsy is an important method to identify these two diseases
