547 Imagination of novel targeted molecular imaging probes in the early diagnosis of upper urinary tract epithelial carcinoma model

Objective: To investigate the characteristic and tumor targeting of up-converting phosphor (UCP) nano-probe in labeling orthotopic transplantation tumor of early upper urinary tract urothelial carcinoma in vivo. Methods: Water soluble UCP was synthesized by high temperature pyrolysis. Then, UCP was link to the pH (low) insertion peptide (pHLIP). Imaging of urothelial tumors in a tumor bearing animal model by pHLIP-UCP probe was proceeded using a small animal in vivo multispectral Maestro imaging system (CRI). Results: NaYF4:Yb3+, Er3+ upconversion nanocrystals were synthesized with good dispersibility, small particle size and high solution permeability. The upconversion luminescence signal of the UCP nanoprobe had stable luminescence and high sensitivity in the dark environment. Conclusion: The UCP nanoprobe can specifically bound to orthotopic transplanted tumor of upper urinary tract urothelial cancer in vivo, which shows the potential for in vivo imaging of early upper urinary tract epithelial carcinoma.

553 Study on circadian rhythms of olfactory response and its regulatory mechanisms in Spodoptera litura

Objective: To study circadian rhythms of olfactory response and its regulatory mechanisms in Spodoptera litura, providing new insight in controlling this pest. Methods: Electroantennogram (EAG) was used to determine the electrophysiological responses of S. litura to chemicals originated from sex pheromones and host plants at eight different time points during day, finding the circadian rhythms of EAG responses in S. litura and selecting out the compounds to which the moths strongly responded. Then the wind tunnel was used to measure the behavioral responses of S. litura to these compounds at the same five time points during the scotophase. In addition, the expression levels of genes regulating circadian rhythms and olfaction at the same time points were detected by the real-time fluorescence quantitative PCR (RT-qPCR) technique to reveal the molecular regulatory mechanism of olfactory circadian rhythms in S. litura. Results: The results showed that the strongest EAG responses of male S. litura to all tested plant odor compounds and sex pheromone components occurred after 3 hours and 9 hours in the dark, showing a bimodal pattern. A total of five compounds: Z9E12-14:OAc, Z9E11-14:OAc,
caryophyllene, linalool and phenylacetaldehyde were selected and tested in behavioral responses (wind tunnel). The behavioral responses of S. litura to the five chemicals at five time points during scotophase showed similar patterns to that obtained from the EAG tests. The expression levels of the most olfactory receptor and clock genes reached the peak after 3 and 9 hours in the dark, which is largely consistent with the circadian rhythms of olfactory responses in S. litura. Conclusion: The circadian rhythms of olfactory responses were consistent with those of expression levels of olfactory and clock genes in S. litura, suggesting that the circadian rhythms are probably essentially regulated by clock and olfactory genes.

561 The influence of different imaging systems on radiation dose to operator in percutaneous coronary intervention

Objective: To discuss the effect of different imaging systems on radiation to the first and second operator s position dose in percutaneous coronary intervention. Methods: In this study, the measurements of the first and second operator’s surface entrance dose rate in 125 cm and 155 cm height were obtained in no and have radiation protection separately through transradial approach by special and public imaging system. The paired t test was used for statistical analysis of dose rate arithmetic mean values. Results: The operator’s surface entrance dose rate of public imaging system (except for the foot and right foot position of first operator in 155 cm height; the right foot position in 125 cm height and foot and right foot position in 155 cm height of second operator) was significantly higher than that of special imaging system in no bedside protection situation. In a bedside protection situation, the operator’s surface entrance dose rates of foot, right foot, left and right position of first operator in 125 and 155cm height in both systems were not detected. The dose rates of the left foot position of public imaging system in 125 cm and 155 cm height were higher than that of special imaging system. The dose rates of head and left head position in 125 cm height and head position in 155 cm height of special imaging system were higher than that of public imaging system. For second operator, the operator’s surface entrance dose rates of public imaging system were significantly higher than that of special imaging system. Conclusion: The operator’s radiation dose of public imaging system is significantly higher than that of special imaging system. In daily work the special imaging system should be used as much as possible so as to reduce the operators’ radiation dose.

566 Expression of lncRNA-AL049452 in the tissue of colorectal cancer and its clinical significance

Objective: To explore the expression and clinical significance of lncRNA-AL049452 in colorectal cancer (CRC). Mothods: Eighty-four cases of CRC tissues and corresponding paracancerous tissues were collected. Quantitative real-time PCR (qRT-PCR) was used to detect the expression level of lncRNA-AL049452 in these tissue specimens. Correlation between the expression of lncRNA-AL049452 and clinicopathological features was analysed by Mann-Whitney U test. Kaplan-Meier survival curves was used to analysed overall survival time and disease-free survival time between different lncRNA-AL049452 levels. Receiver operating characteristic (ROC) curve was performed to observe the diagnostic value of lncRNA-AL049452 for colorectal cancer. Results: lncRNA-AL049452 expression level in CRC tissues was obviously higher than that in corresponding paracancerous tissues (P=0.0035). High expression of lncRNA-AL049452 indicated larger tumor size (P=0.024). The lncRNA-AL049452 high-expression group presented significantly lower disease-free survival rates than the low-expression group (log-rank=4.208, P=0.034). In addition, ROC curves analyse showed that the AUC of ROC curve was 0.723 (95%CI=0.613-0.832, P<0.001) with 86.0% sensitivity, 51.2% specificity. Conclusion: The high expression of lncRNA-AL049452 in tumor tissues may play an important role in the progression in CRC, and can serve as novel biomarker for the diagnosis and prognosis of CRC.

571 Evaluation of the inhibitory action and toxicity of elemene liposome combining ultrasound cavitation on the ectopic tumor of the nude mice model

 Objective: To evaluate the inhibitory action and toxicity of elemene liposome (EL) combining ultrasonic cavitation on the entopic tumor for the nude mice model. Methods: The inhibitory efficacy was studied using C6 cells bearing armpit tumor model. Mice were assigned to 5 groups (n=7) as follows: group 1 for saline, group 2 for EL, group 3 for EL+MB, group 4 for EL+US and group 5 for EL+MB+US. Mice were administered via tail vein at the dosage of 50 mg/kg amount to 0.1 mL/10 g (injection solution/mice weight), 1 time per 2 d, 3 times in all. Tumor volume, tumor weight change, body weight growth rate were all measued. The white blood cell count (WBC) at the end point was also tested. Results: Compared with EL, EL+MB+US showed higher tumor growth inhibition, fewer mice body weight change and white blood count change (P<0.05) in the armpit tumor model. Conclusion: With better anti-tumor activity and lower blood toxicity, EL+MB+US may be developed as a new cancer targeted delivery system.

576 Optimization of the scan protocol of single energy or dual energy and reconstruction method in hepatic arterial phase

 Objective: To Compare the image quality and radiation dose of image acquired from a dual energy CT scan with image obtained from a single energy scan in the second dual-source generation system abdominal CT combined with iterative reconstruction. To determine the optimal scan methods and reconstruction methods for clinical work. Methods: Two size arterial phase liver phantoms containing two density lesions (low and high) was scanned using a second generation dual source dual energy CT (DSCT) scanner in the single energy mode (120 kVp) and the dual energy mode (100 kVp/Sn 140 kVp). Images were reconstructed with both filtered back-projection (FBP) and iterative reconstruction (IR) algorithms. Volume CT dose index (CTDIvol) and the dose length product (DLP) recorded from all protocols were used to dose evaluation. For quantitative analyses of the phantom, the contrast-to-noise ratio (CNR) and figure of merit (FOM) were measured. Results: For small size phantom, CNR of high density lesions in the mixed images derived from a dual energy CT scan was significantly higher than that in images acquired with a single-energy scan (P<0.05), while there was no statistically significant difference existed in low density lesions (P>0.05). For big size phantom, CNR of both high and low density lesions in dual-energy mixed images showed a higher CNR in comparison to single-energy images (P<0.05). Dose of dual energy CT scan was higher than that of single energy scan. As to FOM, except in the low density lesions group IR images of small size phantom, there was no significant difference between all lesions of the dual energy scanning standard fusion image and single energy scanning image (P>0.05). Compared with FBP images, CNR of IR images was significantly better than that of FBP images (P<0.05). Conclusion: Take both the image quality and radiation dose to consider, there is no obvious difference between the dual energy scanning standard fusion image and single energy scanning image. IR images always have higher CNR than FBP images. Above all, IR image reconstruction is suggested in hepatic artery phase scanning image both in dual or single-energy images, and dual energy scan should be choosed according to whether need material-specific information

580 Comparative study between low tube voltage cerebral CT angiography and 3D rotational angiography in diagnosis of intracranial micro-aneurysms

Objective: To investigate the diagnostic value of low tube voltage cerebral CT angiography (L-CTA) in intracranial micro-aneurysms (IMA, maximum diameter≤3 mm). Methods: The clinical and imaging data of 59 IMA patients confirmed by 3D rotational angiography (3DRA) in the First Affiliated Hospital of Wenzhou Medical University from May 2013 to December 2016 were retrospectively analyzed. All patients had underwent low tube voltage cerebral CT angiography. Comparing with 3DRA, the diagnostic sensitivity, specificity, and accuracy of L-CTA in intracranial micro-aneurysms was evaluated. The related radiation dose in L-CTA was objectively evaluated with the conventional tube voltage. Results: Based on the results of 3DRA, 65 cerebral micro-aneurysms of 70 aneurysms were detected in these 59 cases. CTA detected a total of 64 micro-aneurysms, including multiple micro-aneurysms (n=6), false positive cases (n=3) and missed cases (n=4). Comparing with the 3DRA (gold standard), the diagnostic sensitivity, specificity, and accuracy of L-CTA in IMA were 93.85%, 99.56% and 99.08%, respectively, and the measurement results of aneurysm size were in good correlation with 3DRA. Compared with conventional CTA, the effective dose (ED) of L-CTA reduced by 35.71%. Conclusion: L-CTA has high diagnostic sensitivity in IMA and can effectively reduce the radiation dose.

585 Effects of sandblasting on the flexural strength and fatigue property of the dental zirconia ceramics

Objective: To evaluate the effect of sandblasting on the flexural strength and fatigue property under cyclic loading of the dental zirconia ceramics. Methods: After polished, sixty bar-shaped specimens of HT were assigned to two groups (n=30) according to the random number table, one of which was sandblasted by alumina particles, the other was polished. The flexural strength of experimental group and control group was obtained from three-point flexural test, then examined by Weibull statistics and analyzed by the group t-test. The number of cycles to fracture was obtained from cyclic fatigue test. The fatigue crack growth rate curve based on Pairs law was used to describe the crack propagation. Results: The phase content of m-ZrO2 experimental group and control group were 11.1% and 6.24%. The flexural strength of experimental group and control group were 1 228.36±137.33 MPa and 1 200.55±114.74 MPa (P<0.05). The Weibull module of the two groups was 13.52 and 12.03. The stress corrosion cracking susceptive indexes were 35.80 and 32.87. The crack propagation occurred at the critical stress-intensity factor was as low as 51.9% and 50.9% of its fracture toughness. Conclusion: After dealing with the method described in this article, both the flexural strength and the crack growth resistance of the HT are increased.

590 The correlation between the relaxation extent of zonule and the closure of eye chamber angle in primary angle-closure glaucoma

Objective: To explore the influence of relaxation of zonule in the pathogenesis of primary angle closure glaucoma. Methods: Seventeen subjects with primary acute angle-closure glaucoma (24 eyes), five subjects with primary chronic angle-closure glaucoma (10 eyes) and six subjects with angle-closure suspect (12 eyes) were enrolled in this study. The changes of anterior chamber depth (△ACD), lens thickness (△LT), lens position (△LP), trabecular-ciliary process angle (△TCPA) after myotic and mydriatict were measured by Lenstar and ultrasound biomicroscopy. The △LP and △TCPA could reflex the relaxation of zonule. Results: The differences of △ACD, △LT, △LP between the three groups were not significant (P>0.05). The differences of △ACD, △LT, △LP between the fellow eye and the acute eye in acute angle-closure glaucoma were not significant (P>0.05). The △TCPA of primary acute angle-closure glaucoma were larger than that of another two groups (P<0.001). But the differences of △TCPA between primary chronic angle-closure glaucoma and angle-closure suspect were not significant (P>0.05). The △TCPA of the acute eye in 6 o’clock was smaller than that of the fellow eye in acute angle-closure glaucoma (P=0.044). Conclusion: The weaken zonule maybe exist in primary acute angle-closure glaucoma in a certain extent.

595 Applicated studies of 23 items revise of review criteria in Sysmex Hematology sampler transport HST-N302

Objective: To evaluate the XE-2100 hematology review criteria suggested by internal consensus group with Sysmex Hematology sampler transport HST-N302. And to perfect review criteria of our clinical laboratory. Methods: The total 25 097 patients’ blood samples were respectively detected and stained seminal smear randomly with the methods of Automated Hematology Analysis on Sysmex HST-N302 base on hematology review criteria suggested by internal consensus group (review criteria 1 in short), hematology review criteria suggested by internal consensus group and LIS history record and scatter diagram (review criteria 2 in short), at the same time, differential leukocyte on the smear included positive and negative sample were counted with the microscope by two docimaster. Calculated true positive (TP), false positive (FP), true negative (TN), false negative (FN) and the ratio of review quantity, then studied out optimal program. Results: According to review criteria 1: TN, FN, TP, FP and review rate respectively were 70.10%, 1.33%, 4.15%, 24.42%, 28.57%. According to review criteria 2: The TN, FN, TP, FP and review rate respectively were 89.89%, 1.22%, 2.32%, 6.58%, 8.90%. Conclusion: It can enhance work efficiency and reduce unnecessary labor work to utilize new review criteria.

599 Analysis of hearing characteristics on children with cytomegalovirus infection and clinical follow-up

Objective: To explore the difference of auditory brainstem response (ABR) after human cytomegalovirus (HCMV) infection, then follow-up the children with hearing loss, so as to provide theory basis for clinical diagnosis and treatment. Methods: One hundred fifty-eight hospitalized cases of children with HCMV infection were collected from 2013 to 2014 in the Second Affiliated Hospital & Yuying Children’s Hospital of Wenzhou Medical University. All of the patients were performed by ABR testing. The children with hearing loss were followed up in out-patient, and underwent ABR testing once again about 3-6 months later. Results: In the first ABR tested 158 cases, there were 120 cases with normal hearing , and 38 cases (55 ears) with hearing loss, among whom 42 ears (76%) were mild abnormal, 7 ears (12.7%) were moderate abnormal, 3 ears (5.5%) were severe loss and 3 ears (5.5%) were profound loss. All of the children were divided into three groups according to the months of age, respectively for≤3 months, 3 to 6 months, ≥6 months comparison difference in three groups
of children with hearing impairment was statistically significant (χ2=7.830, P=0.020). Compared two among three groups, the hearing impairment of 3 to 6 months group was higher than that of≤3 months group, and the difference was significant (χ2=5.725, P=0.017). The incubation period in normal hearing ear I, III, V wave pointed compared with normal children, the difference was statistically significant (P<0.05). 38 cases (55 ears) of children who had hearing loss were followed up, and performed ABR testing again. Five cases (7 ears) of children had persistent anomalies on hearing loss, among whom 3 ears were sensorineural hearing loss. 2 cases of children, who had bilateral hearing loss, were symptomatic HCMV infection. Three cases of children, who had unilateral hearing impairment, were asymptomatic infection. Conclusion: HCMV infection may resulted in hearing impairment, they usually are mild injury, sometimes are sensorineural hearing loss. The incidence rate of hearing impairment among children with different months is different. Hearing impairment has the characteristics of reversibility, regardless of first hearing testing is normal or not, long-term follow-up and hearing testing are suggested.

604 The correlation and changes of serum level of cytokines between intra-amniotic infection and feto-maternal hemorrhage

Objective: To explore the correlation and changes of serum level of cytokines between intra-amniotic infection (IAI) and feto-maternal hemorrhage (FMH). Methods: Eighty cases of premature rupture of membranes were selected as research object. According to pathological examination of placenta and umbilical cord after childbirth, all cases were assigned into observation group (40 cases) with IAI and control group (40 cases) without IAI respectively. All cases were performed Kleihaue-Betke test and used ELISA to detect serum levels of IL-6, TNF-α and IL-10 antepartum. Incidence of maternal FMH and changes of cytokines in patients with IAI were analyzed. Results: Compared with the total number of pregnant women in our hospital, the incidence of IAI was 0.54% (40/7 638), the incidence of FMH was 0.14% (11/7 638). However, the incidence of FMH in patients with IAI was 22.5% (9/40). The finding showed that 9 cases with Kleihaue-Betke test≥0.6% in observation group and 2 cases with Kleihaue-Betke test≥0.6% in control group. The incidence of FMH was statistically significant (P<0.05). The levels of IL-6 and TNF-α in serum in observation group and FMH group were higher than those in control group and group without FMH (P<0.05). The plasma concentrations of IL-10 was lower (P<0.05). Conclusion: In Patients with IAI, the incidence of FMH is increased. We can find the elevated levels of IL-6, TNF-α in serum and the reduced level of IL-10 in serum in patients with IAI complicating FMH. Changes of cytokines keep consistent with those change induced by IAI.

608 The diagnostic value of hypotonic water enema spiral CT enhanced scanning on the clinical staging of colon cancer

Objective: To explore the diagnostic values of hypotonic water enema spiral CT enhanced scanning on clinical staging of colon cancer. Methods: From September 2011 to February 2016, 78 colonoscopy and biopsy confirmed colon cancer patients in our hospital were selected, the result of the hypotonic water enema spiral CT enhanced scanning, characteristics and TNM stage were analyzed. Results: Hypotonic water enema spiral CT scan were completed in all patients, the lesion was filling good, the plain CT value was (35.42±11.63) HU, the arterial phase enhanced scan value was (76.33±12.72) HU, and the portal phase, the equilibrium phase enhanced scan value were (79.43±9.54) HU and (80.14±8.11) HU. All the patients were detected by the hypotonic water enema spiral CT enhanced scan; After determination, the accuracy of CT for colon cancer T staging was 84.6%, the accuracy of N staging was 82.1%, the accuracy of M staging was 97.4%. Conclusion: The application of hypotonic water enema spiral CT enhanced scanning in colon cancer has good feasibility and effectiveness, and it can improve the accuracy of preoperative TNM staging, and it has good application values.

612 Analysis of clinical efficacy of a novel incision and drainage device for epiglottitis abscess under the guidance electronic laryngoscope

Objective: To investigate the efficacy of the new type epiglottis abscess incision operation under the guidance of electronic laryngoscopy. Methods: Thirty-four cases of acute epiglottis with abscess inpatients in department of otolaryngology of the Third Affiliated Hospital of Wenzhou Medical University, dating from Sep. 2013 to Dec. 2015 were enrolled in this study. Eighteen cases received direct laryngoscopy surgery, and 16 cases underwent the new type epiglottis abscess incision operation under the guidance of electronic laryngoscopy. The differences in operation duration, postoperative average hospitalization days, and pain scores between the two groups were compared. Results: There was no difference in postoperative average hospitalization days between the two groups, while the new incision group lengthen operative duration. The pain scores in post group were significantly lower than that in the direct laryngoscopy group. Conclusion: The new type epiglottis abscess incision operation under the guidance of electronic laryngoscopy can be easily performed with the advantages of clear view, less pain, and desirable effect.

615 The clinical observation of the local directly punctures to improve the arteriovenous internal fistula stenosis in the blood dialysis

 Objective: To investigate the effects of local puncture in treatment of arteriovenous internal fistula stenosis in hemodialysis patients. Methods: Thirty nine arteriovenous fistula stenosis cases from January 2012 to January 2016 in the First Affiliated Hospital of Wenzhou Medical University were included. In order to expand blood vessels, 16 g needles were used to puncture in the narrowest site of the fistula and separate blood vessels within the organization, with thrombolysis being used on thrombosis patients. Every time before puncture, the fistula was evaluated and touched. Whenever hemodialysis was conducted, the above puncture procedures would be repeated and internal fistula stenosis was observed. Results: Twenty-seven of 39 cases were effective treatment, and 12 cases were invalid. The treatment was more effective with patients whose narrow fistula sites were less than two and those whose stenosis length was shorter than 1 cm (χ2=14.745, 8.269, P<0.01). The blood flow increased and venous pressure decreased in patients of effective treatment after puncture (t=-9.251, 8.628, P<0.01). That the time of puncture on those patients was 8.6±3.8. Nine of the 27 effective cases showed vascular expansion and disappearance of stenosis after the puncture; another 13 cases with two narrow sites showed increased blood flow and deereading of the venous pressure that 5 cases with three narrow sites, including one thrombosis patient whose stenosis almost disappeared after thrombolysis and repeated punctures, showed dropping of the venous pressure. Nine of the 27 cases were seen noticeable shrinking aneurysm; 5 cases were reported disappearance of local sites pains and 3 cases reported pain relief. Twelve patients with invalid treatment turned to fistula rebuild or balloon expansion. Conclusion: Local puncture in short-term fistula within narrow place can expand local blood vessels, prevent and treat arteriovenous fistula in vascular stenosis, reduce the patients pain and economic burden, be convenient and feasible.
