235 Effects of enhanced expression of S100A9 mediated by recombinant adenovirus on proliferation and apoptosis of squamous cervical cancer cells

Objective: To construct the recombinant adenovirus carrying S100A9 gene, and to explore its effect on the proliferation and apoptosis of squamous cervical cancer cells. Methods: The shuttle plasmid pHBAd-MCMV-RFP-S100A9 was constructed and co-transfected into HEK293 cells with backbone plasmid pHBAd-BHG to produce the recombinant adenovirus Ad-S100A9. Then the C-33A cells were infected with Ad-S100A9, cells infected with Ad-RFP as negative control group, and cells without infection as blank group. S100A9 protein expression was examined by western blot. CCK-8 and flow cytometry were carried out to examine the effects of S100A9 over-expression cell proliferation and apoptosis of C-33A cells, respectively. Results: Ad-S100A9 was successfully constructed. Ad-S100A9 infection significantly enhanced the expression of S100A9 in C-33A cells, and effectively promoted cell proliferation (P<0.05), but had no effect on cell apoptosis. Conclusion: Ad-S100A9 can mediate S100A9 over-expression and promote cell proliferation of squamous cervical cancer cells.

239 Compartmental imbalance and aberrant immune function of dendritic cells in peripheral blood from Graves’ disease

Objective: To assess the imbalance of dendritic cells (DC) in the peripheral blood from patients with Graves’ disease (GD), and the influence on the differentiation of T helpler cells (Th). Methods: In total, 36 untreated GD patients and 21 healthy individuals were enrolled in the study. The circulating DC levels, including plasmacytoid DC (pDC) and myeloid DC (mDC), were identified by FACS. In further, pDC and mDC isolated by MACS were co-culture with the naïve T cells to investigate the role on proliferation and differentiation of Th cells. Results: A significant increase in the percentage of circulating DC and pDC subset was found in GD patients compared with healthy individuals, and GD-derived pDCs were conferred the ability to induce the proliferation of T cells as well as the polarized differentiation of Th1 in an IFN-α-dependent manner. Conclusion: The elevated circulating pDCs in GD patients can play a role on the differentiation of Th1 in IFN-α-dependent manner, and contribute significantly to the pathogenesis.

244 Impact of 5-HTTLPR on rest-state brain function in children with ADHD

Objective: To explore the impact of serotonin transporter gene linked polymorphism region (5-HTTLPR) on the rest-state brain activities of amplitude of low frequency fluctuation (ALFF) in children with  attention deficit hyperactivity disorder (ADHD). Methods: Fifty-one children with ADHD and thirty-seven normal controls were checked up by resting state functional magnetic resonance scanner. The ALFF approach was used to analyse the brain function in resting state. Restriction fragment length polymorphism (RFLP) method was applied to detect 5-HTTLPR. Results: Compared with the normal controls, ADHD showed decreased ALFF in left median superior frontal lobe, right precuneus, and left precuneus. Compared with each other among gene subtypes of 5-HTTLPR, there was diferent ALFF value in left median superior frontal lobe and left precuneus. Post hoc analysis showed ALFF value of S/S genotype was higher than L/L genotype, S/L genotype higher than that of L/L genotype, and there was no statistically significant difference between S/L genotype and S/S genotype. There were no statistically significant differences in the frequencies of the genotypes and alles of 5-HTTLPR between ADHD (L/L, S/L, S/S genotypes: 3.9%, 41.2% and 54.9%; S, L allels: 40.7%, 59.3%) and normal controls (L/L, S/L, S/S genotypes: 5.4%, 37.8% and 56.8%; S, L allels: 36.4%, 54.6%). Conclusion: S allele activity is associated with altered activities in particular brain regions of ADHD children.

248 The correlative analysis between microRNA-135b and EGFR mutation status in lung adenocarcinoma of Zhoushan Archipelago

Objective: To analyze the relationship between EGFR mutations and miR-135b in lung adenocarcinoma of Zhoushan Archipelago and find new indicator for the guidance of EGFR-TKIs. Methods: MiR-135b quantification was retrospectively detected in 70 paraffin-embedded lung adenocarcinoma tissue samples and the corresponding para-carcinoma tissue using Taqman-based quantitative RT-PCR assays (qRT-PCR). The EGFR gene exons 19 and 21 of tumor tissue were amplified by PCR, followed by direct sequencing in 70 paraffin-embedded lung adenocarcinoma tissue. Results: MiR-135b was markedly up-regulated in lung adenocarcinomas in comparison to the corresponding para-carcinoma tissue (P<0.01) and miR-135b had a higher level in the group of invasive lung adenocarcinoma (IAC) compared to the group of noninvasive lung adenocarcinoma (adenocarcinoma in situ/minimally invasive adenocarcinoma, AIS/MIA) (P<0.01); EGFR mutations were detected in 24 cases of 70 (34.3%) patients with lung adenocarcinoma. The mutation rate in IAC (53.3%) was higher than that in AIS/MIA (20%). Patients harbouring EGFR mutations had higher miR-135b expression level, which indicated that miR-135b was positive correlation with EGFR mutations. Conclusion: EGFR mutations and miR-135b may be positive correlation with the invasion of lung adenocarcinoma and both of them can be used to assess the progress of lung adenocarcinoma. MiR-135b can be the biological marker for effectiveness evaluation of EGFR TKIs and is expected to become the new target for improving EGFR TKIs resistance.

254 Expression of transferrin receptor in cervical carcinoma and its clinical significance

Objective: To investigate the potential correlation between TFRC (transferrin receptor) gene expression and the development of cervical cancer. Methods: The expression of TFRC and TfR1 were detected in 96 cases of cervical cancer patients and 30 cases of normal controls with immunohistochemistry and reverse transcription-polymerase chain reaction (RT-PCR) method. At the same time, TCGA database was used to analyze the data of patients with cervical cancer and the survival was analyzed with Kaplan-Meier method. Results: The expression of TfR1 was positive in 50 cases of cervical carcinoma, and the intensity of staining increased with the increase of cervical cancer patients. Compared with normal cervical tissues, cervical carcinoma in stage II and III was significantly enhanced (P<0.001). The expression of TfR1 was related to the FIGO stage, the degree of tumor differentiation and the presence of metastasis (P<0.05). However, it has nothing to do with the age of patients with cervical cancer (P>0.05). The gene expression of TFRC was observed in both cervical cancer cases and normal cervical tissues. There was correlation between TFRC expression and histological grades (P<0.001). The expression of TFRC in phase II and phase III was significantly higher than that in early stage of cervical cancer. According to Kaplan-Meier survival analysis, the level of TFRC gene expression was associated with overall survival rate of cervical cancer patients (P<0.001). The stronger expression of TFRC gene, the lower the overall survival rate of patients with cervical cancer. Conclusion: TFRC may be involved in the process of development of cervical cancer and related with the prognosis of patients with cervical cancer.

258 Effect of progestin-primed ovarian stimulation scheme in patients with diminished ovarian reserve

Objective: To compare the embryo laboratory outcomes of different ovarian stimulation schemes in patients with diminished ovarian reserve, to explore the application value of progestin-primed ovarian stimulation scheme in patients with diminished ovarian reserve. Methods: A total of 228 cycles of patients with diminished ovarian reserve who received IVF/ICSI-ET treatment from November 2015 to July 2016 were analyzed retrospectively. They were divided into progestin-primed ovarian stimulation (Group A, 87 cycles), luteal-phase stimulation (Group B, 45 cycles) and mild stimulation (Group C, 96 cycles) according to treatment protocols. The basic situations and the embryo laboratory outcomes were compared and analyzed between Group A, Group B and Group C. Results: The basic situations in three groups were no significant differences (P>0.05). The dosage of Gn administration in Group B was significantly higher than those in Group A and Group C (P<0.05). The D3 quality embryo rate in Group A was significantly higher than Group C (P<0.05). The serum LH level of the patients in Group B on the day of HCG administration was significantly lower than that in Group A and Group C (P<0.05). The serum P level of the patients in Group B on the day of HCG administration was significantly higher than Group C (P<0.05). There were no significant differences in the retrieved oocytes number, mature oocytes number, D3 good-quality embryos number, available embryos number, the normal fertilization rate, the cleavage rate, the available embryo rate among three groups (P>0.05). Conclusion: Progestin-primed ovarian stimulation scheme can obtain higher D3 quality embryo rate and is a viable regimen for patients with diminished ovarian reserve.

263 Relationship among air pollutants and meteorological factors and respiratory infection in children

Objective: To assess the relationship among the concentrations of different air pollutants and meteorological factors in Wenzhou city and respiratory tract infection in children. Methods: The study was conducted in Wenzhou city, by recording the live air quality data and meteorological factors from January 2015 to December 2015, and collecting the clinic visiting data from the pediatric outpatient and emergency department of the Second Affiliated Hospital & Yuying Children’s Hospital of Wenzhou Medical University. The relationship between air pollutants concentrations and meteorological factors and the clinic visit volume caused by respiratory infection were analyzed by Pearson correlation statistics and multivariant linear stepwise regression model. Results: The average concentrations of NO2, PM2.5 and PM10 were higher than the national standards of the second level of annual average concentration. The rates of exceed standard of NO2, PM2.5 and PM10 concentration were 16%, 29% and 6% respectively. Their levels were highest in winter. The numbers of days of exceeding standard were more than half a year. Regression analysis revealed that 10 μg/m3 increase of NO2 might result in 2.9%, 9.1%, 2.5% increase of clinic visits due to bronchitis, bronchiolitis and pneumonia. And a 10 μg/m3 increase of PM10 was associated with 2.9% increase of clinic visits caused by upper respiratory tract infection and 1.8% increase of clinic visit volume caused by bronchitis and 2.5% increase of clinic visits due to pneumonia. An increase of 1 ℃ of the daily minimum temperature might result in 1.8%, 2.8%, and 3.3% decrease of clinic visits due to bronchitis, bronchiolitis and pneumonia. Conclusion: In year 2015, the average concentrations of NO2, PM2.5 and PM10 stayed in high level. NO2, PM2.5, PM10 are major air pollutants in Wenzhou city. The increase in level of NO2, PM10 and the decrease of the daily minimum temperature are correlated with the increase of number of respiratory tract infection in children.

268 Application of CTA and MRA in preoperative evaluation of anterolateral thigh perforator flap transplantation

Objective: To evaluate the value of computed tomography angiography (CTA) and magnetic resonance angiography (MRA) in the anterolateral thigh perforator flap (ALTPF) transplantation. Methods: According to preoperative imaging, patients who underwent the ALTPF transplantation were divided into two groups: computed tomography angiography (CTA) Group and magnetic resonance angiography (MRA) Group. Thirty cases were in each group. The parameters of perforator size and origin location in the ALTPF were compared between preoperative and operation by paired t-test; The imaging quality of CTA and MRA and the parameters of perforator course and origin location measured by CTA and MRA were compared between preoperative and operation by Fisher exact test; The success rate and complications of operation between CTA group and MRA group were also compared. Results: There was no significant difference in imaging quality between CTA Group and MRA Group (P>0.05). Comparing preoperative measurement with operation findings, there was no significant difference between two Groups in preoperative imaging accuracy of the types of the perforators, the diameters and the origin location of perforators, and the operation success rate and complications (all P>0.05). Conclusion: Preoperative mapping using CTA and MRA is a feasible and reliable method for the flap design and preoperative evaluation in ALTPF transplantation.

273 The early clinical curative effect of percutaneous transforaminal approach spinal endoscopy for treatment of degenerative lumbar spinal stenosis in elderly patients

Objective: To evaluate the early clinical efficacy of minimal invasive transforaminal endoscopic spine system for treatment of single segment degenerative lumbar spinal in elderly patients. Methods: From January 2013 to January 2014, eighty single segment degenerative lumbar spinal elderly patients were randomly divided into two groups according to the random number table (n=40). The research group was undergone percutaneous transforaminal endoscopic discectomy and spine canal enlarged as well as relaxing the nerve. The control group were treated by opening lamina resection decompression as well as spine canal enlarged, interbody fusion and internal fixation. The Mac Nab’s criteria, ODI score, visual analogue scale (VAS), Lehmann low-back rating scale, the Japanese Orthopaedic Association Scores (JOA), the hospital stay time, the intraoperatve blood loss, the surgical complications in the two groups were reviewed and compared between the preoperative and postoperative. Results: The research group was superior than the control group in the Mac Nab’s criteria, ODI score, VAS score, JOA score, the hospital stay time, the intraoperative blood loss, the surgical complications. Their differences were significant (P<0.05). Conclusion: The minimal invasive transforaminal endoscopic spine system for treatment of single segment degenerative lumbar spinal in elderly patients has the advantages of small trauma, earlier rehabilitation and good effects.

277 Etiology and risk factor analysis of oral candidiasis from in-patients of dermatological department

Objective: To determinie the distribution of species among in-patients of dermatological department, investigate the risk factors associated with oral candidiasis and evaluate antifungal susceptibility profile of oral candida. To provide a theoretical basis of candida infection and prevention. Methods: Cluster sampling method was adopted in our study. Mucosal swab samplings were taken from the objects. The strains were identified by CHROMagar Candida agar medium, and API 20C AUX system for yeasts. Antifungal susceptibility was tested in vitro by the broth microdilution method. The risk factors was investigated by analysis of clinical data. Results: Oral candida carriage rate was 40.4% in in-patients of dermatological department. C. albicans accounted for 83.1% of the candida species. Infectious skin diseases, connective tissue disease, blister disease and use of antibiotics were risk factors associated with oral candidiasis. The percentage of resistant to itraconazole, fluconazole and voriconazole were 8.5%, 4.4% and 1.2%. All of the isolates were susceptible to amphotericin B. Conclusion: The oral candida colonization rate in in-patients of dermatological department is high. The percentage of resistant to C. albicans is low, but high azole-resistance rate is found among non-C. albicans fungal species. In vitro antifungal susceptibility is necessary before antifungal therapy.

282 Early identification of neutrophil-to-lymphocyte ratio for severe coronary artery stenosis in patients with chest pain

Objective: To investigate the early identification of NLR for severe coronary artery stenosis in patients with chest pain. Methods: Clinical data of 484 patients who had undergone coronary angiography owing to pectoris symptom were retrospectively analyzed. Patients were divided into severe stenosis group and mild stenosis group based on Gensini scores. Logistic regression and receiver operator characteristic curve were used to analyze early identification of NLR for coronary artery stenosis. Results: Using multiple logistic regression analysis, advanced ages (OR=1.033, 95%CI: 1.013-1.054, P=0.001), male (OR=1.750, 95%CI: 1.062-2.883, P=0.028), LDL-C (OR=1.723, 95%CI: 1.232-2.411, P=0.001), and NLR (OR=1.345, 95%CI: 1.205-1.500, P<0.001) emerged as independent identification for the severe coronary artery stenosis. In receiver operating characteristics curve analysis, NLR>2.32 had 78.5% sensitivity and 62.6% specificity in identify the severity of coronary artery stenosis. Conclusion: High NLR is a powerful identification for the sverity of coronary artery stenosis in patients with chest pain.

286 Correlative study of neutrophil-to-lymphocyte ratio and lower extremity arterial disease in patients with type 2 diabetes

Objective: To investigate the relationship between neutrophil-to-lymphocyte ratio (NLR) and lower extremity arterial disease (LEAD) in T2DM patients. Methods: Six hundred and twenty-seven inpatients with T2DM were recruited in this study. The numbers of the white blood cell, neutrophils (N) and lymphocytes (L) as well as the NLR values of peripheral blood were detected. Based on the quartile of NLR, all patients were divided into four groups: Q1≤1.30 (n=159), Q2: 1.31-1.81 (n=154), Q3: 1.82-2.50 (n=156) and Q4≥2.51 (n=158). The relationship between NLR and LEAD in T2DM patients was discussed. Results: Logistic regression analysis showed the prevalence of LEAD was significant higher in Q3 (OR: 1.62, 95%CI: 1.03-2.55, P=0.037) and Q4 (OR: 1.84, 95%CI: 1.17-2.90, P=0.008) than that in Q1. After adjusting clinical characters and biochemical, the prevalence of LEAD was still higher in Q4 (OR: 1.75, 95%CI: 1.04-2.92, P=0.034). Conclusion: The prevalence of LEAD in T2DM patients increases with the rise of NLR.

290 Feasibility study on clinical application of the new labor standards in Wenzhou

Objective: To explore the feasibility of application of the new labor standards in Wenzhou. Methods: A retrospective analysis was performed in the Second Affiliated Hospital of Wenzhou Medical University. Objects included were primiparas who had singleton full-term primipara, cephalic presentation and spontaneous delivery. The study was divided into new and old labor group. According to whether labor analgesia implemented, the two groups were compared respectively in the Cesarean delivery rate (CDR), rate of artificial rupture, rate of forceps delivery, duration of labor, rate of intrapartum fever, postpartum hemorrhage in 2 hours and incidence of neonatal asphyxia. Results: The CDR and the rate of artificial rupture of the new labor group was significantly decrease compared with the old labor group (P<0.01); The main Cesarean section indications of the new labor group was fetal distress, while that under the condition of the old labor group was protracted and arrested active phase; With the implementation of labor analgesia, rate of oxytocin usage, rate of intrapartum fever, span of the first and the second stage of labor, postpartum hemorrhage in 2 hours and incidence of neonatal asphyxia in the new labor group were significantly higher than that in the old labor group (P<0.05). Conclusion: Although the new labor standards increased the rate of oxytocin usage, the rate of intrapartum fever and the incidence of neonatal asphyxia, it reduced the initial CDR and the rate of artificial rupture. In general, the new labor standard is worth to popularize.

293 Retrospective study on 57 patients with acute lower limb ischemia

Objective: To summarize the clinical characteristics and prognosis of the patients diagnosed as acute lower limb ischemia (ALLI). Methods: Retrospectively analyze the clinical data of 57 patients who were assigned according to gender, clinical classifications, prognosis, etc,. to see whether there were significant differences. Results: The total curative ratio of surgery treatment was superior to that of conservative treatment (P<0.01). The male patients usually have worse onset condition than female patients and have worse prognosis. Conclusion: The curative ratio of surgical treatment group of the ALLI patients is higher than that of conservative therapy group. If the patients’ conditions are allowed, surgery will be the better choice. The male patients usually have worse onset condition than female patients and have worse prognosis.

296 Functional study of insulin-like growth factor binding protein 7 in human gastric cancer SGC-7901

Objective: To investigate the effect of insulin-like growth factor binding protein 7 on gastric cancer SGC-7901 cells and its mechanisms. Methods: The recombinant expressing plasmid pCMV-XL4-IGFBP7 was constructed and transfected into SGC-7901 cells. Empty plasmid transfection was regarded the control. Cell proliferation was analyzed by MTS assay; cell apoptosis was detected by flow cytometer. Results: Compared with control group, the rate of SGC-7901 cell of pCMV-XL4-IGFBP7 group on G1 level was higher; while the rate of SGC-7901 cell of pCMV-XL4-IGFBP7 group on G2/M was lower than the control group. The proliferation of SGC-7901 cells had no significant change compared with the control group. Conclusion: IGFBP7 may inhibit gastric cancer SGC-7901 cells through influencing the cell cycle.
