703 Characterization of specific miRNAs in acute monocytic leukemia

Objective: To characterize the molecular markers of microRNA (miRNA) with specifically high expression in acute monocytic leukemia. Methods: A batch of miRNAs was firstly screened out for their significantly higher expression in THP-1 (acute monocytic leukemia cell line) by analyzing high-throughput miRNA transcriptome sequencing data of THP-1 and K562 (chronic myeloid leukemia cell line). These miRNAs were further tested in the THP-1 and K562 cell line by quantitative real-time PCR (qRT-PCR) to obtain the specifically high expressed miRNAs in THP-1 cell line. Finally, these screened miNRAs were verified in bone marrow samples of acute monocytic leukemia patients by qRT-PCR. The finally verified miRNAs could serve the candidates for clinical diagnosis of this disease in the future. Results: Three miRNAs that are specifically highly expressed in acute monocytic leukemia including let-7d-5p, miR-221-3p and miR-222-3p were characteried. Conclusion: By integrating the bioinformatics analysis and verification in cell lines and patient bone marrow samples, we finally characterized 3 miRNAs as potential molecular diagnostic markers in acute monocytic leukemia.

709 The genetic analysis of patients with non-syndrome deafness come from twenty-two families in Yuyao, Zhejiang province

Objective: To explore the clinical, genetic and molecular characteristics of twenty-two families with non-syndrome deafness in Yuyao area. This study focused on analyzing mutations of mitochondrial gene and coding sequence of GJB2, GJB3, GJB6 gene. Methods: The samples were 33 patients with non-syndrome deafness come from 22 families in YuYao People’s hospital, and 254 cases with normal hearing. DNA were extracted out from peripheral blood of all subjects. All samples’ GJB2, GJB3 and GJB6 gene encoding region and mitochondrial gene were analyzed by direct sequencing, audiological examination, genetic testing for deafness and pedigree data. Results: The sequencing results revealed that 4 families carried GJB2 235delC mutation, 2 families carried GJB2 compound heterozygous mutations accounted for 27.3% and 3 families carried mitochondrial 12S rRNA A1555G accounted for 13.6% in the 22 families with non-syndrome deafness. All families without pathogenic mutations in the GJB3 and GJB6 encoding region. According to clinical data, 6 families with GJB2 gene mutations and 3 families with 1555A>G mutations had different levels in hearing loss, age of onset, and the rate of hearing loss. Conclusion: GJB2 and mitochondrial 12S rRNA 1555A>G gene mutation are the main genetic inheritance for the patients with non-syndrome deafness in Yuyao area, there may also be other unknown genes or environmental factors to coordinated with GJB2 gene and mitochondrial 12S rRNA gene co-modulate the variable penetrance and expressivity of deafness.

716 Applied research of iterative reconstruction technique in the low-dose chest CT scans of children

Objective: To explore the feasibility of low tube current combined with iDose4 iterative reconstruction technique in the low-dose chest CT scans of children. Methods: Eight-eight children continuously underwent routine chest CT scan were divided into two groups using conventional dose (50 mAs) and low dose (25 mAs) scan, respectively. Routine dose group underwent filtered back projection (FBP) reconstruction, record as group A, low dose group was performed FBP, iDose4-1, iDose4-3, iDose4-5 iteration reconstruction (IR), and so referred to as group B, C, D, E. Measured and calculated CT value, noise, signal to noise ratio of each group images in order to evaluate the quality of the images objectively; quality of images was assessed according to five levels rating method by two diagnostic radiology physicians of deputy chief physician title; recorded volume CT dose index (CTDIVOL) and dose length product (DLP) provided by CT scanner dose reports, and calculated the effective dose (ED). Subjective and objective images quality and radiation dose were statistically analyzed. Results: Compared with routine dose group, effective dose (ED) of low dose group decreased by 52%. Subjective and objective quality of images of group B, the worst quality of low dose groups, was less than that of routine dose group (P<0.05), subjective and objective quality of images of group C was less than that of routine dose group (P<0.05), no significant statistical difference was existed between group D and group A in subjective and objective quality of images (P>0.05), subjective and objective quality of images of group E was better than that of group A (P<0.05). Conclusion: When low-dose chest CT scans of children are taking, 25 mAs tube current combined with iDose4-3 iterative reconstruction technique can obtain the same quality of images as routine dose group, while the radiation dose is reduced by 52%.

720 Experimental study of flavonoids from Coreopsis tinctoria of lipid-lowering effect on liver cells

Objective: To study the lipid-lowering effect of flavonoids from Coreopsis tinctoria on the liver cells with fatty degeneration. Methods: The medical fat emulsion injection was used as fat resource to make human liver fat cell model on normal liver cell line L-02. With different concentrations (20, 50, and 100 μmol/L
flavonoids from Coreopsis tinctoria were co-cultured with hepatocytes with fatty degeneration and incubated for different times (3, 12, 24, 48 h). The oil red staining was used to mark the intracellular lipid droplets and morphological changes. The cell culture supernatants were collected to test triglyceride content. Results: L-02 high fat cell model was feasible. High concentration of flavonoids (100 μmol/L) showed the significant decrease of lipid droplets and lower triglyceride levels. The triglyceride decreased with prolonged incubation time. Conclusion: Flavonoids from Coreopsis tinctoria can reduce triglyceride in the liver cells with fat degeneration. There is no significant liver cell enzyme damage. The effect depends on the concentration and incubation time.

724 Association between ERCC1, XPD, XPC polymorphisms and chemosensitivity of platinum-based neoadjuvant chemotherapy in patients with locally advanced cervical cancer

Objective: To investigate whether single-nucleotide polymorphisms of ERCC1, XPD, XPC affected response in locally advanced cervical cancer (LACC) receiving platinum-based neoadjuvant chemotherapy (NACT). Methods: A total of 114 patients who were pathologically diagnosed as cervical carcer treated with platinum-based neoadjuvant chemotherapy were examined for genotyping of ERCC1, XPD and XPC in peripheral blood lymphocytes with the method of polymerase chain reaction-restriction fragment length polymorphism (PCR-RPLF), and than the correlation between different genotypes with NACT curative effect was analized. Results: Variant genotypes of ERCC1 Asn118Asn were significantly associated with /C genetype LACC chemosensitivity (P<0.05). To compare ERCC1 Asn118Asn C/T genetype with ERCC1 Asn118Asn C/C genetype, chemosensitivity increased 4.48 times (odds ratio 4.48; 95%CI: 1.436-13.976; P<0.05). While no significant difference was found between chemotherapy response and SNPs of XPD condon 751 and XPC condon 939 (P>0.05). Conclusion: Our study provides evidence for the predictive role of ERCC1 Asn118Asn on  chemotherapy response in LACC patients treated with.

730 Morphologic characteristics of the lateral ankle fragment: A 3-D computer tomography based study

Objective: To evaluate the morphological characteristics of the lateral malleolar fracture line and associated fragment geometry for the design of novel internal fixation. Methods: A retrospective analysis of 3-D computed tomography data of lateral ankle fracture cases from February 2012 to April 2016 was conducted. The anterior tip distance (ATD), crest tip distance (CTD), posterior tip distance (PTD), the proximal cross angle (p-CA), and distal cross angle (d-CA) were measured as morphologic assessments in the Picture Archiving and Communication System (PACS). Results: Based on the recognition of the lateral ankle fracture by CT reconstruction, at a confidence interval of 95%, the fragment was described with ATD (2.97±0.03) cm, CTD (4.52±0.07) cm, PTD (3.17±0.11) cm, p-CA (103.20±1.94)°, d-CA (78.50±1.78)°. The coefficient of variation (CV) of p-CA was calculated 9.86, and the CV of d-CA arrived at 11.90. Conclusion: The anterior initiation site of lateral fracture line is relatively invariable in comparison with proximal and posterior frontier. The double facet cross angle of the fragment appears decrescent along axis of distal fibula while distal cross angle is found slightly variable, compared with proximal data. The discovery obtained from this series could be helpful in the modeling of prospective biomechanical researches and developing novel fixation.

734 Correlation between abdominal adipose tissue thickness and lumbar disc degeneration: A study based on magnetic resonance imaging

Objective: To investigate the correlation between abdominal adipose tissue thickness and lumbar disc degeneration. Methods: A total of 1 108 inpatients were enrolled from the Second Affiliated Hospital of Wenzhou Medical University between October 2008 and September 2013. Parameters of abdominal adipose tissue thickness based on MRI included abdominal diameter (AD), sagittal diameter (SAD), ventral subcutaneous thickness (VST), and dorsal subcutaneous thickness (DST). Improved Pfirrmann grading system was adopted to evaluate the degree of lumbar disc degeneration. Results: Spearman correlation analysis revealed that SAD and AD had significant associations with disc degeneration score sum (r=0.20, P<0.001; r=0.19, P<0.001) in females. There was negative correlation between VST and disc degeneration score sum (r=-0.10, P=0.01) in males. Ordinal logistic regression showed that SAD (OR=1.14, 95%CI: 1.08~1.20, P<0.001) and AD (OR=1.15, 95%CI: 1.08~1.23, P<0.001) were risk factors of disc degeneration in females; VST (OR=0.70, 95%CI: 0.55~0.89, P=0.003) decreased the risk of disc degeneration in males. Conclusion: AD, SAD and VST are associated with lumbar disc degeneration, and parameters of abdominal adipose tissue thickness based on MRI may be early markers for screening lumbar disc degeneration.

738 The value of imaging characteristics on CT in the diagnosis of preinvasive lesions of lung adenocarcinoma

Objective: To analyze the computed tomography appearances of preinvasive leisions of lung adenocarcinoma and to improve the diagnosis of atypical adenomatous hyperplasia (AAH) and adenocarcinoma in situ (AIS). Methods: Twenty-eight cases of AAH and 57 cases of AIS confirmed pathologically were retrospectively recruited. Comparison of CT features of lung nodules in the two groups, including location, shape, margin, diameter, solid component, internal characteristics and mean CT attenuation. Results: The blood vessels through of focal ground glass nodule (fGGN), diameter and the mean CT attenuation were significantly different between the two diseases (P<0.05 for all), while no difference was found in the pattern of fGGN, location, shape, boundary, lobulation speculation and vacuole sign between AAH and AIS (P>0.05 for all). The average of diameters was (7.9±3.0) mm for AAH, which was smaller than (12.3±3.9) mm for AIS. The optimal cut-off value from the receiver characteristic (ROC) curve was 8.5 mm, with sensitivity of 82.5% and specificity of 71.4%. The mean CT attenuation was (-644±71) HU for AAH, which was smaller than (-492±87) HU for AIS. The optimal cut-off value from ROC was -549 HU, with sensitivity of 77.2% and specificity of 96.4%. Concluion: Comprehensive analysis of the pattern, size, density, shape and internal characteristics of fGGN is helpful for differential diagnosis of AAH and AIS.

743 Safety evaluation of MRI examination for the intraocular Ex-PRESS drainage device

Objective: To evaluate the safety of the intraocular Ex-PRESS glaucoma drainage device during magnetic resonance imaging (MRI). Methods: The Ex-PRESS drainage device was implanted in the anterior chamber or fixed through the corneal limbus respectively in pig eyes. Then they were exposed to magnetic field of standard 1.5T and 3.0T MRI scanning protocols. Testing included measurements of translational and rotational motion of the device induced by static magnetic field. In addition, the surrounding tissue of the device was also observed due to possible radiofrequency heating. Seven patients with Ex-PRESS drainage device implanted at the limbus were also studied. For each subject, 1.5T and 3.0T MRI scan were obtained. The safety and imaging quality were then assessed. Results: Rotation and displacement did not occur under 1.5T and 3.0T conditions in both tests. There was no obvious burn sign in the surrounding tissues of the device. For the patients, the position of the Ex-PRESS device kept stable after MRI scanning protocols. The presence of Ex-PRESS device had mild impact on visualization of the surrounding tissue. Only minimal artifact was present at the implant site. Conclusion: Routine MRI examination is safe for intraocular Ex-PRESS glaucoma drainage device implantation.

747 Extraction process of oxymatrine from sophora alopecuroides L. using ultrasonic-microwave synergistic extraction

Objective: To study the ultrasonic-microwave assisted extraction of oxymatrine from sophora alopecuroides L. Methods: The extraction process of oxymatrine was investigated with ethanol concentration, solid-solvent ratio, microwave power and irradiation time. Results: Optimized process was obtained by using orthogonal rotation design method with higher extraction efficiency than that using conventional and individual extraction technologies. The results showed that the optimum condition was as following: ethanol concentration 70%, extraction time 40 min, microwave power 500 W, and the solid-solvent ratio 1:15 (g/mL). Conclusion: The ultrasonic-microwave synergistic extraction technology has advantages of reliability, extracting efficient, and is suitable for industrial application.

752 Association between serum triglyceride levels and cognitive function in depressive disorder

Objective: To investigate whether there are the characteristics of cognitive impairments in depressive patients, and to further examine whether serum triglyceride levels are associated with cognitive impairments in depressive patients. Methods: Eighty-three depressive patients and 87 healthy controls were enrolled in a case-control study. Cognitive function was assessed by RBANS in all subjects. Serum triglyceride levels were measured in both two groups. Results: Multivariate analysis of covariance showed that there were significantly lower cognitive function on the RBANS total score, the language index score, and the delayed memory index score in depressive patients than healthy controls (P<0.01). For the patients, serum triglyceride levels were negatively associated with the RBANS total score (P<0.01), the language index score (P<0.05), the attention index score (P<0.05), and the delayed memory index score (P<0.01). Further multivariate regression analysis showed that serum triglyceride levels were associated with the RBANS total score (P<0.05), the attention index score (P<0.05), and the delayed memory index score (P<0.01). Conclusion: Depressive patients have poorer cognitive function, especially in language and delayed memory. Moreover, the higher levels of serum triglyceride can cause cognitive impairments including attention and delayed memory in depressive patients.

756 Effects of tropisetron on postoperative delirium in elderly patients undergoing hip replacement operation

Objective: To observe the effect of tropisetron on the incidence and development of postoperative delirium in elderly patients undergoing hip replacement operation. Methods: According to drug application, 212 elderly patients (age≥60 years) undergoing hip replacement operation were randomly divided into tropisetron group (108 cases) and control group (104 cases). General anesthesia were applied in both groups. In tropisetron group, 5 mg tropisetron was administered intravenously each day for 5 postoperative days. Control group was given saline solution instead of tropisetron. The rate of postoperative delirium in each group was evaluated by confusion assessment method (CAM). Routine nursing intervention and drug treatment was given to delirium patients in each group, and the curative effect was observed in three days. Results: Incidence of delirium in tropisetron group was 5.6% (6 cases), significantly less than control group 14.4% (15 cases). After routine nursing intervention and drug treatment, 5 (83.3%) of the 6 delirium patients in tropisetron group were cured, but only 4 (14.4%) of the 15 delirium patients in control group were cured. Conclusion: Tropisetron can apparently reduce the incidence rate of postoperative delirium in elderly patients undergoing hip replacement operation and improve the curative effect of conventional treatment of postoperative delirium.

759 Clinical efficacy of 308 nm monochromatic excimer light for the treatment of alopecia areata in children and teenagers

Objective: To investigate the clinical efficacy and safety of 308 nm monochromatic excimer light for the treatment of alopecia areata in children and teenagers. Methods: A total of 26 children and teenagers (age≤19 years) with 49 recalcitrant patches alopecia areata were enrolled in a self-control clinical trail. Lesion was treated twice a week by the 308 nm monochromatic excimer light for 15 treatments. Every patient selected one untreated alopecia areata patch that was used as a control site. Results: While no improvement of all the untreated alopecia areata patches were observed, the clinical efficacy of treated patients was as follow: effective group 30 cases, and total effective rate of improvement was 61.22%. Conclusion: The 308 nm monochromatic excimer light is safe and effective for treatment of alopecia areata in children and teenagers.

762 Endovascular treatment of arterial injuries: a clinical analysis of 8 cases

Objective: To explore endovascular treatment of arterial injuries. Methods: Clinical records of 8 patients with arterial injuries who underwent endovascular treatment between September 2012 and December 2015 at the First Affiliated Hospital of Wenzhou Medical University were retrospectively analyzed. Results: Eight patients presented with 8 arterial injuries. The causes included impact injuries (n=4), drug injection (n=1), iatrogenic injuries (n=3). Among them, 2 cases of aortic dissection, 2 cases of carotid arterial injury, 1 case of subclavian arterial injury and 1 case of femoral profound artery pseudoaneurysm were treated with stent grafting. During follow up ranged from 4 to 36 months, all stent grafts were patent without any migration or endoleak. One case of superior gluteal artery pseudoaneurysm and 1 case of posterior tibial artery pseudoaneurysm were treated with arterial embolization. During follow up from 3 to 15 months, false aneurysm sac shrank without visible blood flow. Conclusion: Endovascular treatment plays an irreplaceable role in the treatment of arterial injuries.

765 Distribution characteristics and clinical  features of intracranial mirrorimage aneurysms

Objective: To investigate distribution characteristics and clinical features of intracranial mirrorimage aneurysms. Methods: The clinical history and images of 48 patients with intracranial mirrorimage aneurysms were analyzed, and 88 patients with ordinary multiple intracraninal aneurysms (MIA) were taken as a comparison group. All patients underwent computed tomography angiography (CTA). Two independent radiologists assessed all the images of CTA and digital subtraction angiography (DSA). Distribution characteristics of intracranial mirrorimage aneurysms were analyzed. The male-to-female ratio, age, rupture proportions, constituent ratio of patients with intracranial micro-aneurysms (aneurysm≤3 mm in maximal diameter) and large aneurysms (≥10 mm in maximal diameter) were statistically compared between patients of intracranial mirrorimage aneurysms and ordinary MIA group, respectively. Results: A total of 48 (35.3%) patients with intracranial mirrorimage aneurysms in 136 patients with MIA were detected. Twenty two cases were seen in posterior communicating artery (PCoA), 12 cases in M1 part of middle cerebral artery, and 14 cases in another location. The male-to-female ratios and rupture proportions were statistically no significant difference between the two groups (P>0.05). 56.3% of patients were under age 60 in patients with intracranial mirrorimage aneurysms and 35.2% in ordinary MIA group. There were statistically significant difference between two groups (x2=5.612, P<0.05). Patients with ≥3 aneurysms were 31.3% in intracranial mirrorimage aneurysms and 15.2% in ordinary MIA. There was statistically significant difference (x2=7.062, P<0.01). Intracranial micro-aneurysms were detected in 54.2% patients in intracranial mirrorimage aneurysms and 73.9% in ordinary MIA, there was statistically significant difference between the two group (x2=5.443, P<0.05). Conclusion: Intracranial mirrorimage aneurysms has characteristic clinical and imaging features. Raising awareness of this diseases provide help for the clinical treatment.

769 The corrleation between the new lung adenocarcinoma classification and the EGFR mutation

Objective: To evaluate the correlation of EGFR mutation and histological subtypes of lung adenocarcinoma based on the IASLC/ATS/ERS classification 2011. Methods: 240 resected lung adenocarcinoma specimens were analyzed by ARMS, then the differences between histological subtypes and EGFR mutation were compared by the x2 test. Results: EGFR mutation was detected in 240 specmens, most of which were acinar and papillary predominant adenocarcinoma, 89% and 56% respectively. EGFR mutation rate of the specimens with acinar, lepidic, papillary, or micropapillary component was higher than that of without these components (P<0.05), and with solid or mucinous component was lower than that without the component (P<0.05). Conclusion: There is heterogeneity of EGFR mutation in lung adenocarcinoma. The presence of well-differentiated components in lung adenocarcinoma, such as acinar, micropapillary, papillary and lepidic, indicates a higher EGFR mutation rate, while the solid and mucinous component indicates a lower EGFR mutation rate.
