Effect of microparticles derived from bone marrow mesenchymal stem cells on cardiac fibroblasts

Objective: To study the effects of microparticles derived from bone marrow mesenchymal stem cells (MSC-MPs) on proliferation, migration and collagen synthesis of cardiac fibroblast. Methods: MSCs were obtained from SD mice and cultured under the condition of serum free in hypoxia. Then the conditioned media were collected and microparticles were then isolated from the media. MSC-MPs were used to treat cardiac fibroblast. CCK-8 assay and cell scratch method were used to detect the effect of MSC-MPs on proliferation and migration of cardiac fibroblast, seperately. The expression of collagens I and III, α-SMA, vimentin and fibronectin in cardiac fibroblast were measured by means of qRT-PCR. Results: The proliferation ability of cardiac fibroblast was enhanced after treated with MSC-MPs. The migration ability was not significantly affected. The expression of collagens I and III, vimentin and fibronectin in cardiac fibroblast were promoted. Conclusion: MSC-MPs can enhance the proliferation ability and collagen synthesis of cardiac fibroblast to improve the cardiac function.

The role of caveolin-1 and P21 in the process of roxithromycin to inhibit the proliferation of asthmatic rats ASMCs in vitro

Objective: To explore the role of caveolae/caveolin-1 and P21 in the process of roxithromycin to inhibit the proliferation of asthmatic rats ASMCs in vitro. Methods: Recovery freeze-stored ASMCs of asthmatic rats were divided into five groups [group A (blank group), group R10 (roxithromycin 10 µg/mL), group R25 (roxithromycin 25 µg/mL), group R50 (roxithromycin 50 µg/mL) and group R100 (roxithromycin 100 µg/mL)]. The proliferation of ASMCs induced by roxithromycin were tested by CCK-8. The changes of caveolae in ASMCs were observed by transmission electron microscope. The protein expressions of caveolin-1 and P21 were measured by Western blot. Results: Roxithromycin could inhibit the proliferation of asthmatic rats ASMCs in vitro and had concentration-dependent manner. With the increased of intervention concentration, the expression of caveolae increased and the protein expressions of caveolin-1, P21 increased. Besides, compared with group A or group R10, the expressions of caveolin-1 and P21 protein in group R50 or R100 showed significantly different (P<0.05). There was significantly negative correlation between the cell activity rate of asthmatic rats and the expressions of caveolin-1 and P21 protein (r=-0.881, r=-0.951, P<0.05). The expression caveolin-1 was positively correlated with P21 (r=0.957, P<0.01). Conclusion: Roxithromycin inhibits the proliferation of ASMCs from asthmatic rats in vitro may partly through promoting the expression of P21 protein which induced by increased the expression of caveolae/ caveolin-1.

Influence of spectral CT monochromatic imaging in combination with iterative reconstruction algorithm on image quality

Objective: To explore the effect of spectral CT monochromatic imaging in combination with adaptive statistical iterative reconstruction (ASiR) algorithm on the spatial resolution, density resolution and low-contrast detectability (LCD) of image in energy spectrum scanning. Methods: Catphan 500 phantoms were scanned using Gemstone Spectral CT. The images were reconstructed for 10 level of single energy from 45 keV to 135 keV, and meanwhile, the ASiR was applied to each keV from 10% to 100%. The CTP528 and CTP515 Catphan phantoms were used to measure the spatial resolution and density resolution, respectively. CTP515 cylinder phantoms with contrast ratio of 1% and diameter of 9.0 mm were chosen as research subjects to analyze low-contrast detectability (LCD) of images. The image quality was compared with multiple factors analysis of variance. Results: In a certain ASiR, as for spatial resolution, the distinguishable line pair per centimeter from 45 keV to 135 keV were 7, 7, 6, 6, 6, 6, 6, 6, 6 and 6 LP/cm, respectively. The density resolution and LCD decreased with the increasing of keV, and reached peaks at 55 and 65 keV. Different single energy was statistically significant (P<0.05) on the spatial resolution, density resolution and LCD. When keV was fixed, ASiR was statistically significant (P<0.05) on the density resolution and LCD, while there was no statistical difference on spatial resolution (P>0.05). Conclusion: Low single energy (45 keV to 65 keV) combined with 20% ASiR can achieve relatively high image quality in energy spectrum scanning with 5 mm slice thickness.

Isolation, identification and cultivation of hepatocytes, hepatic stellate cells and Kupffer cells from rats

Objective: To establish a protocol for isolation, identification and cultivation of hepatocytes (HC), hepatic stellate cells (HSC) and Kupffer cells (KC) from rats. Methods: By combining in situ perfusion with collagenase, density gradient centrifugation and selective adhension, primary HC, HSC and KC were obtained. Cell yield and viability were assessed by automated cell counter. The purity of these cells were detected with immunofluorescent staining, special staining and phagocytosis assay. Results: The average yield, viability and purity of isolated HC were (21.0±8.2)×106/per gram liver, 92.1%±2.1%, and 96.5%±2.2%, respectively. The average yield, viability and purity of isolated HSC were (2.5±0.8)×106/per gram liver, 90.1%±3.8%, and 93.1%±1.5%, respectively. The average yield, viability and purity of isolated KC were (4.1±1.2)×106/per gram liver, 90.1%±3.8%, and 95.1%±1.4%, respectively. The isolated cells could be cultured in vitro for long-term. HSC underwent a gradual phenotypic transition to a myofibroblast-like phenotype with vitamin A losing in vitro culture. KC could proliferate, and had the ability of phagocytosis, and the expression of CD 68 at 14 d of culture. Conclusion: The simultaneous method is feasible to isolate and culture primary rat HC, HSC, and KC.

Effects of microRNA-212 on migration and invasion of gastric cancer cell lines SGC7901

Objective: To investigate the effect of human microRNA212 (miR-212) on the migration and invasion of gastric cancer cell line SGC7901. Methods: The i-miR-212 as the suppressor gene of miR-212 was transfected into SGC7901 cells according to different concentration (25, 50, 75, 100 nmol/L) via lipofectamin 2000. Empty vectors and unrelated fragment were also applied as controls. The expression of mature type miR-212 was detected by real-time PCR. Cell Counting Kit-8 was used to evaluate the effect of miR-212 on the proliferation of SGC-7901 cells, meanwhile Transwell assay for migration and invasion of SGC7901 cells. Results: Real-time PCR showed that cells Transfected with i-miR-212 had significantly low expression of miR-212, with the optimal concentration of miR-212 being 50 nmol/L; the expression of miR-212 was 18 times lower than empty group (P<0.01). The Cell Counting Kit-8 showed that the proliferation of cancer cells with i-miR-212 (50 nmol/L) was higher than that of the control group (P<0.01). The Transwells showed the migration and invasion of cancer cells with i-miR-212 (50 nmol/L) was higher than that of the control group (P<0.01), respectively. Conclusion: The i-miR-212 can effectively reduce miR-212 expression and promote the migration and invasion of SGC7901 cells. miR-212 may be a tumor suppressor gene in gastric cancer tumor cells.

The influence of operators for the repeatability and reproducibility of axial length measured by Lenstar

Objective: To explore the influence of different operators to the repeatability and reproducibility of Lenstar in axial length measurements. Methods: Sixty left eyes of 60 participants were enrolled. Each participant was performed two measurements which was finished by operator I and operator II, each measurement including at least three continue measure. The SD (standar deviation) and CV (coefficient of variation) of all the participants was calculated to evaluate the reproducibility of operator I and operator II. The difference between operator I and operator II in AL measurement and it’s 95%CI (confidence interval) was calculated to evaluate the reproducibility between operator I and operator II. Results: The Mean±SD of operator I and operator II were (24.05±0.02) mm and (24.05±0.02) mm. The CV of operator I and operator II were 0.07% and 0.06%. The Mean±SD of the difference of AL between operator I and operator II was (0.00±0.03) mm. The 95%CI of the difference of AL between operator I and operator II was 0.00±0.06. Conclusion: The factor of operators don’t affect the repeatability and reproducibility of Lenstar.

The influence of proton pump inhibitor on nutritional status in patients with post-stroke dysphagia

Objective: To explore the influence of proton pump inhibitor (PPI) on nutritional status in patients with post-stroke dysphagia. Methods: A total of 155 cerebral infarction patients with dysphagia were enrolled in this study. All the patients were given enteral nutrition by a gastric tube. Then they were randomly assigned into the PPI treatment group and the control group. Pantoprazole was given the PPI group to protect the gastric mucosa. The control group didn’t give any gastric mucosa protective treatment. Then the hemoglobin, serum albumin, triceps skinfold thickness, arm muscle circumference, gastric juice pH, NIHSS score, acute gastric mucosal lesions (AGML) and mortality rate were compared between the two groups. Results: The two groups had no significant difference in hemoglobin, serum albumin, triceps skinfold thickness, arm muscle circumference, gastric juice pH and NIHSS score on admission (P>0.05). After two weeks, the hemoglobin, serum albumin, triceps skinfold thickness, arm muscle circumference in two groups both decreased (P<0.05), and the PPI group decreased more compared to the control group (P<0.05). Also the PPI group had a lower NIHSS score (P<0.05) and lower risk of AGML (P<0.05). But the two groups had no significant difference in mortality rate (P>0.05). Conclusion: The using of PPI will influence the nutritional status in patients with post-stroke dysphagia. Using PPI has a poor neurological outcome, but has no relationship with mortality rate. In the protective treatment of gastric mucosa in cerebral infarction patients with dysphagia, the clinicians should weigh the risk of dystrophy. Then, an individualized treatment to each patient is given.

The relationship between injured uterosacral ligaments and ectopic pregnancy

Objective: To compare the morphology of uterosacral ligaments and the distribution of nerve fibers in the isthmus of the fallopian tube in patients who presented with ectopic pregnancy with previous history of induced abortion and patients who presented with ectopic pregnancy with out history of prior surgical procedures. The relationship between abortion and ectopic pregnancy were also explore and investigated to offer new research ideas into the pathogenesis and management of tubal pregnancy. Methods: The patients with ectopic pregnancy were subdivided into two main groups per their history of induced abortion. The induced abortion group comprised of 20 cases with reproductive history of 0-0-N-0 and the no-abortion group comprised of 12 cases with reproductive history of 0-0-0-0. The isthmuses of the fallopian tube from these two groups were obtained and were immunohistochemically analyzed to detect the protein gene product 9.5 in comparison with results from the morphology of the uterosacral ligaments. Results: ①The uterosacral ligaments were symmetrical and smooth with complete arches connecting the right and left ligaments (10/12, P<0.05) in the no-abortion group. In the nulliparous women with prior surgical evacuations, there was evidence of injury to one, or both, uterosacral ligaments (19/20, P<0.05), and possible neovascularisation of peritoneal surfaces over the ligament and adjacent peritoneum and mesosalpinx. ②The strong positive expression of PGP 9.5 in the nerve fiber from the inner and outer muscle layer of the isthmus of fallopian tube in abortion group was significantly reduced when compared with the no-abortion group (P<0.05). ③PGP 9.5 showed a strong positive expression in the mesosalpinx of the fallopian tube among the two groups (P>0.05). Conclusion: Induced abortion may damage the nerves in the uterosacral liagments and the endometrial-muscular interface and the decrease of PGP 9.5 positive nerve fibers expression in the muscle layer of the isthmus of fallopian tube may lead to the occurrence of ectopic pregnancy.

Indirect computed tomography lymphography for diagnosis of early neck lymph node metastasis in tongue cancer

Objective: Experimental evaluation of the potential utility of indirect computed tomography lymphography (CT-LG) for detecting early cervical lymph node metastasis in rabbits VX2 tongue cancer model.Methods: Eighteen New-Zealand rabbits were divided into 3 groups randomly, one group were injected egg-yolk emulsion in the neck, one group were transplanted with VX2 tumor tissue mass suspension in the tongue, and the last remained untreated. CT-LG with submucosal injection of 0.5 mL Iohexol in the tongue was performed, followed by postmortem examination of deep cervical lymph nodes. Results: Only deep cervical lymph nodes were enhanced in CT-LG, there was no noticeable enhancement in the blood vessels. The average diameter of metastatic lymph nodes were (0.73±0.08) cm in CT, the hyperplasic group were (0.75±0.10) cm and the normal group were (0.47±0.05) cm. The contrast in metastatic nodes reduced more slowly than normal and hyperplasic nodes with significant difference. Metastatic nodes’ manifestations in CT-LG included filling defect, rugosity in line and heterogeneous while the other two groups appear to be smooth and homogenous. Conclusion: Indirect CT-LG has some advantage in finding out the early neck lymph node metastasis in tongue cancer.

Clinicopathologic features, diagnosis and treatment with extrauterine stromal sarcoma

Objective: To study the clinicopathologic and immunohistochemical features, biological behavior, diagnosis and treatment of extrauterine stromal sarcoma (ESS). Methods: A retrospective clinicopathologic analysis was made on 10 cases of ESS admitted to the Third Clinical Institute Affiliated to Wenzhou Medical University and the First Affiliated Hospital of Wenzhou Medical University from April 1998 to September 2012. Results: Among 10 patients, 9 had low-grade ESS and 1 had extrauterine undifferentiated endometrial sarcoma (EUES). 6 tumors located in the ovary, while the other 4 cases involved bilateral ovaries/uterine rectum nest, the right ovary/genital vein/inferior vena cava, the left ovary/retroperitoneal and fallopian tube respectively. The clinical manifestations were mainly abdominalgia, abdominal mass and algomenorrhea, 5 patients had endometriosis. The others were alive until the end of follow-up time. Immunohistolochemically, CD10 (8/10), Estrogen receptor (5/10), Progesterone receptor (9/10), Smooth muscle actin (5/10), Desmin (2/10), Caldesmon (1/10), All 10 patients were treated with complete surgical resection and received postoperative adjuvant therapy. The follow-up time for all patients ranged from 19~127 months. 5 patients had one or more recurrences, the time of first recurrences was within 2 years. 2 of 5 patients died of multiple recurrences including 1 EUES. Conclusion: ESS which is the most common in ovary is an extremely rare neoplasm. Most of them are associated with endometriosis. The preferred therapy is complete excision and postoperative adjuvant therapy. ESS with low-grade had one or more replases easily in short time, but the patients should be alive for a long time. CD10 as immunohistochemical marker is specific.

Good right body put impact on treatment outcomes for patients with stroke

Objective: To study the good body position is put for stroke patients with hemiplegia side limb motor function, muscle tone, and the influence of daily life (ADL) of independence. Methods: Meet the requirements for the study of stroke patients were divided into experimental group and control group, experimental group 30 cases and control group 30 cases. After admission in the two groups were treated with the same time, the strength of treatment, treatment method, the experimental group and use good body position to put all the way, the control group is affected by many take random position. Without the rating scale, ADL scale and the modified Ashworth muscle tension, the function of patients in hospital and 30 days in hospital were evaluated. Results: The patients with hemiplegia side body complications were significantly lower than the control group (P<0.05). Motor function, muscle tension and daily life activities ability improvement compared with the control group with significant difference (P<0.05). Conclusion: Patients with stroke hemiplegia limbs early to put a good body position all the way can reduce the hemiplegic side body complications, limb spasm and contracture, and promote the paralyzed limbs rehabilitation, so as to improve the life of patients with self-care ability and life quality, promote patients early return to society.

The posture correction chair’s impact on improving the sitting position ability of late developers

Objective: To study the posture correction chair’s impact on improving the sitting position ability of late developers. Methods: Forty seven cases children with developmental delays and cannot sit alone, correction were randomly divided into group of 24 cases, 23 cases of control group, 2 group adopt routine rehabilitation, correct set equipped with posture correction chair at the same time. Before and after treatment, Gesell developmental scale was used to assess the patient’s total motor development level (development quotient, DQ) and gross motor (GM) and fine motor (FM) ability were scored, score before and after treatment and the result was compared and analyzed. Results: After treatment for 2 months, 2 groups of DQ, GM, FM score was improved obviously in the correction (P<0.05) group scored was higher than the control group (P<0.05). Conclusion: Equipped with posture correction chair is more conducive to improve sports movement function of children with developmental delay.

Analysis for nosocomial infections in patients with induction period of acute leukemia

Objective: To explore the clinical features and risk factors for nosocomial infection of patients with acute leukemia in induction period, so as to provide the theoretical basis for clinical intervention. Methods: One hundred and nine patients with acute leukemia in Wenzhou Central Hospital from February 2011 to September 2015 treated by remission induction chemotherapy were retrospectively analyzed to research the distribution of infection, pathogenic bacteria and risk factors for nosocomial infection. Results: The nosocomial infection rate of patients with acute leukemia in induction period was 71.56%, and the main part was the lower respiratory tract infection accounting for 27.72%. Gram negative bacilli were the major pathogens. The univariate analysis revealed that the incidence of nosocomial infection with acute leukemia in induction period was associated with age, leukemia type, neutrophil count, agranulocytosis time, days of hospitalization and chronic basic diseases (P<0.05); The multivariate logistic regression analysis indicated that age, neutrophil count, days of hospitalization and chronic basic disease were independent risk factors. Conclusion: Age, neutrophil count, days of hospitalization and chronic basic disease are independent risk factors for patients with acute leukemia in induction period, therefore efficacious protective steps should be taken against these factors.

The clinical value of CT guidance Hook-wire positioning thoracoscopic surgery for ground-glass opacity lung nodules

Objective: To evaluate the clinical application of guidance Hook-wire positioning for ground-glass opacity before video-assisted thoracoscopic resection. Methods: Preoperative location of 25 patients with ground-glass opacity lung nodules was performed using CT-guided Hook-wire technique. Video assisted thoracic surgery (VATS) pulmonary wedge resection was adopted. According to the result of intraoperative frozen pathology, the next mode of operation was adopted. Results: GGO diameter (11.48±3.50) mm, diatance from the parietal pleura (16.52±5.98) mm. Twenty five patients were successfully located. The mean procedure time for preoperative CT-guided Hook-wire location was (15.04±3.38) min. 5 patients had micro pneumothroax after positioning. The chest tube drainage was not required. One patient had minimal needle tract parenchymal hemorrhages. No one occured migration and fall out of the hookwire. VATS wedge resection surgery successful rate was 100%. Twelve cases of pure thoracoscopy pulmonary wedge resection operation time was (21.67±3.94) min. Thirteen patients were diagnosed as infiltrating adenocarcinoma of the lung. All of them underwent lobectomy or lung resection and lymph node cleaning by VATS. The mean procedure duration for VATS was (58.84±12.01) min. Conclution: CT-guided Hook-wire fixtion is safe, reliable and easy to learn clinical techniques. It reduces the operation time, and has a low rate of a minor complictions. CT-guided Hook-wire fixtion is worthy of clinical popularization and application.

A comparative study of clear corneal incision versus superior scleral tunnel incision in secondary intraocular lens implantation

Objective: To evaluate and compare the efficacy between clear corneal small incision and Superior Scleral Tunnel Incision in patients undergoing secondary intraocular lens implantation with suture fixation. Methods: Retrospectively analyzed, 40 aphakic patients performed clear corneal small incision treatment (Group A; 40 eyes) and 41 aphakic patients who underwent traditional corneoscleral incision surgery (Group B; 41 eyes) in Lishui Central Hosptial from 2009 January to 2015 June. Analyzed postoperative complications including visual acuity, surgically-induced astigmatism, hyphema, corneal edema and intraocular lens deflection at 1 day, 1 week, 1 month and 3 months, after surgery. Results: Camparted A and B groups, the difference of visual acuity and surgically-induced astigmatism, in different time after surgery was statistically significant (P<0.05 ), The incidence rate of hyphema, corneal edema and intraocular lens deflection from Group-A was less than Group B, and the difference of incidence rate of corneal edema was statistically significant (P<0.05). Conclusion: The secondary intraocular lens suture through tunnel incision on the temporal clear cornea plays a positive role to enhance the postoperative uncorrected visual acuity and decrease complications for patients of aphakia, which is important supplement of posterior chamber intraocular lens.

The influence of zoledronic acid on the osteoporotic patients after total knee arthroplasty

Objective: To analyze the impact of zoledronic acid on patients suffering from osteoporosis after total knee arthroplasty (TKA). Methods: A retrospective study was conducted to 38 cases selected from patients in need of knee arthroplasty admitted from September 2009 to June 2013. Among these, 18 cases (study group) were given intravenous drip of zoledronic acid to 5 mg after knee arthroplasty; 20 cases (control group) were given the same amount of placebo, and closely observed for the subsequent reaction in infusion process. Their bone density was tested in preoperative and postoperative 3 months; meanwhile, new fractures in other parts or around prosthesis appearing during follow-up visits were recorded, as well as HSS function after surgery for TKA. Results: Bone density at the lumbar (L2-4), distal femur and proximal tibia (g/cm2) of study group are (0.642±0.059), (0.621±0.059) and (0.601±0.051), respectively; these of the control group are (0.654±0.062), (0.623±0.061) and (0.602±0.057), respectively. Three months later, these data of study group increased to (0.785±0.078), (0.797±0.064), (0.757±0.063), respectively; these data of control group respectively become (0.652±0.067), (0.621±0.059), (0.601±0.059), respectively. Comparison shows that preoperative and postoperative bone mineral density of the two groups has obvious statistical significance (P<0.05). Moreover, fracture incidence in other parts of study group and HSS score were 5.56% and 82.2 (points), respectively; those of the control group were 20% and 76.6 (points). Comparison of the two groups had significant statistical significance (P<0.05). Conclusion: The application of zoledronic acid to patients with osteoporosis after total knee arthroplasty is safe and effective, not only reducing incidence of fractures around prosthesis and in other parts, but also improving bone density around knee joint, lumbar spine and increasing HSS score of knee joint.

Clinical characteristics of aspiration pneumonia in elderly patients with nasogastric feeding

Objective: To investigate the clinical characteristics, pathogenic bacterial characteristics, predisposing factors of elderly patients with nasogastric feeding and to explore the prevention and therapeutic strategies. Methods: Retrospective analysis on clinical characteristics, and the bacterium of cause of disease of sputum culture structure of 60 cases of aspiration pneumonia in patients with nasogastric feeding from Wenzhou Hospital of Traditional Chinese Medicine between January 2014 to January 2015. The age, basic diseases, etiology characteristic, symptoms and signs in aspiration pneumonia in elderly patients with nasal feeding were investisated. Results: Aspiration pneumonia in patients with nasogastric feeding often merged a variety of basic diseases, age, difficulty swallowing, disturbance of consciousness obviously increased the proportion. Gram-negative bacteria was the main pathogen bacteria in this patients. Conclusion: aspiration pneumonia in patients with nasogastric feeding causes more susceptible factors. However, because of complicated history and untypical symptoms that treatment should be depend on the characteristics of etiology and high effective antibiotics.
