469 Roles of Drp1 and Mfn2 in palmitate induced rat hepatocyte injury and the effects of curcumin derivative L6H4 on it

Objective: To explore the roles and mechanism of mitochondrial dynamin-related protein 1 (drp1) and mitochondrial fusion protein 2 (Mfn2) in palmitate (PA) induced rat hepatocyte injury in the model of nonalcoholic-fatty-liver-disease (NAFLD) in vitro, and the effects of curcumin derivative L6H4 on it. Methods: Rat BRL-3A hepatocytes were cultured with difference concentrations of PA and oleate (OA) and PA+ curcumin derivative L6H4 mixed medium for 24 hours respectively. Cell vitality was detected by MTT assay and the optimal concentration of PA for establishing non alcoholic fatty liver model in vitro and drug intervention of curcumin derivative L6H4 were screened. Cells were divided into 4 groups: control group (CON group), oleate group (OA group), palmitate group (PA group) and curcumin derivative L6H4 intervention group (L6H4 group), and were cultured with normal medium, containing oleate medium, containing palmitate medium and containing palmitate and curcumin derivative L6H4 mixed medium for 24 hours, respectively. Total SOD activity was detected by hydroxylamine method. The content of MDA was analyzsed by thiobarbituric acid method. The expression of mRNA and protein of Drp1, Mfn2, Bcl-2, Bax, Caspase-3, TNF-α of hepatocyte were detected by real-time PCR and Western blot respectively. Results: Different concentrations of PA had certain inhibitory effect on BRL-3A cells growth (P<0.05), and showed a clear dose dependence manner, 0.1 mmol/L PA was the cut-off point, while 0.05 mmol/L OA had no obvious effect. 0.05 mmol/L PA was selected as the optimal concentration to establish the model of NAFLD in vitro. The cell viability was higher than 75% when the concentration of curcumin derivative L6H4 was lower than 10 µmol/L. The cell viability dropped to below 50% (P<0.05) when the concentration of curcumin derivatives L6H4 was higher than 20 µmol/L, 10 µmol/L L6H4 was the cut-off point. Therefore, 5 µmol/L curcumin derivative L6H4 was chosen for subsequent L6H4 interfere concentration. Compared with the CON group, MDA content of cellular homogenization and the expressions of mRNA and protein of Drp1, Bax, Caspase-3, TNF-α were increased (P<0.05) significantly, T-SOD activity and the expressions of mRNA and protein of Mfn2, Bcl-2 were decreased (P<0.05) significantly in PA group. Compared with the PA group, MDA content of cellular homogenization and the expressions of mRNA and protein of Drp1, Bax, Caspase-3, TNF-α were decreased significantly (P<0.05), T-SOD activity and the expressions of mRNA and protein of Bcl-2 were increased significantly (P<0.05) in L6H4 group, but the expression of Mfn2 had no significant difference (P>0.05). Conclusion: Curcumin derivative L6H4 plays an important role in attenuation hepatocyte injury induced by PA, which is associated with its decrease the lipid peroxidation, inhibition mitochondrial fission and suppression the downstream of mitochondrial apoptotic pathway and signal transduction of inflammatory factor. It may be one of the underlying mechanism for curcumin derivative L6H4 to treat NAFLD.

476 Effect of low-dose cyhalothrin on motor behavior in zebrafish embryos

Objective: To study the effects of low-dose cyhalothrin on motor behavior in zebrafish embryos. Methods: The dose-response effects and malformation phenotype were evaluated by observing zebrafish embryos exposed to different concentrations of 0, 10, 20, 40, 80, 100 µg/L cyhalothrin solutions. The impact of low-dose of 0, 2.5, 5, 10 µg/L cyhalothrin on several motor behaviors in zebrafish embryos was then explored, including embryonic spontaneous movement, touch response and larval swimming speed. Results: The predominant malformation phenotypes observed in surviving embryos were edema of pericardialsac, bent spine, curved tail and yolk sac edema. Compared with the control group, zebrafish embryos exposed to 20 µg/L and above concentrations showed significantly increased malformation rate (P<0.05), while 10 µg/L cyhalothrin exhibited no influence on embryonic development. Cyhalothrin concentration that led to 50% malformations (EC50) of the embryos at 120 hours post fertilization (hpf) was 38.61 µg/L. A series of motor behavior tests revealed that embryos exposed to 2.5 µg/L cyhalothrin displayed remarkably elevated spontaneous movement frequency at 19, 21 and 22 hpf (P<0.05), while this frequency was decreased in embryos treated with 10 µg/L cyhalothrin at 20-21 hpf (P<0.05); besides, 5, 10 µg/L cyhalothrin exposure induced markedly reduction of touch response movement in zebrafish embryos at both 27 and 48 hpf (P<0.05); moreover, 10 µg/L cyhalothrin reduced larval swimming movement speed at 120 hpf (P<0.01). Conclusion: Cyhalothrin causes severe embryonic toxicity in a dose-dependent manner. Low dose of cyhalothrin exposure which shows no effect on embryonic development significantly alters several motor behaviors at early life stage.

482 Heplotype gene linkage analysis for hemophilia A families

Objective: To develop variable number tandem repeat (VNTR) polymorphism, short tandem repeat (STR) polymorphism and restriction fragment length polymorphism (RFLP) heplotype gene linkage analysis for hemophilia A (HA) pedigrees in Wenzhou, further to provide evidences of gene prognosis of males and female carriers for HA genetic counseling and family planning guidance. Methods: For the HA probands and related family members, haplotype gene linkage analysis was processed. PCR was used to analyze the VNTR polymorphism in extragenic site DXS52 (St14) of clotting factor VIII (FVIII) gene, to analyze the STR polymorphism in extragenic sites DXS15 (CA)n, DXS9901 (GT)n, DXS1073 (GT)n and in intragenic sites intron 1 (GT)n, 13 (CA)n, 22 (GT)n (AG)n, 24 (GT)n of FVIII gene, and the STR polymorphism was identified by capillary electrophoresis. Also PCR/RFLP was used to analyze the RFLP in intragenic sites intron 18, 19 and 22 of FVIII gene. Results: The results of two HA pedigrees were taken as examples. In pedigree 1, the very young brother of proband was normal, the mother and grandmother of proband were carriers, the young aunt of proband was carrier and her very young son was normal. In pedigree 2, the grandmother of proband was not carrier, the X chromosome of proband was interited from his grandfather. Since the grandfather was not HA patient, supposing the most serious situation, it was deduced that a mutation of FVIII gene had happened to the X chromosome of the mother which interited from the grandfather when it was in germ cell of grandfather, thus the mother of proband was carrier. In pedigree 2, the very young sister of proband is carrier, and there will be risk for her sons, but the eldest aunt of proband and her very young daughter are not carriers. Conclusion: The VNTR-PCR, STR-PCR and PCR/RFLP haplotype gene linkage analysis of HA pedigrees of this study, especially the presymptomatic gene diagnosis of males, the gene diagnosis of female carriers without HA offspring, will have their very important clinical implications, can provide reliable evidences for genetic counseling and family planning guidance.

490 Study on the role of Akt signaling in FGF21-induced prevention on diabetic carodiomyopathy

Objective: To explore the role of fibroblast growth factor 21 (FGF21) on diabetic cardiomyopathy and analyze the role of Akt signaling in FGF21-induced prevention on diabetic carodiomyopathy. Methods: Male C57BL/6J mice were allowed to establish mouse model of type 2 diabetes. Diabetic mice were randomly selected for the treatment of FGF21 for 6 months. Heart tissues were obtained for detection of histopathological changes, cardiomyocyte hypertrophy, fibrosis index and apoptosis after heart functions were detected. Isolation and culture of primary myocardial cell in mice, high glucose/lipid (HG/Pal) treatment of cells to simulate the environment in type 2 diabetes mellitus, at the same time, FGF21 and Akt2-siRNA intervention were given for 39 hours to detect the change of cardiomyocyte hypertrophy, apoptosis and fibrosis. Results: It was demonstrated in vivo study that FGF21 could significantly alleviate heart structural changes, dysfunction, cardiomyocyte hypertrophy and fibrosis induced by type 2 diabetes (P<0.05). It was verified in vitro study demonstrated that FGF21 could upregulate Akt phosphorylation level, activation of Akt and downstream signaling pathways, inhibit the activity of Akt, abate FGF21 protection of myocardial injury. Conclusion: FGF21 plays a protective role in diabetes-induced myocardial injury, and Akt signaling is a key mechanism in FGF21-induced myocardial protection.

497 Effects of curcumin derivative WZ01 on asthmatic airway inflammatory mediators IL-13, histamine and glucocorticoid receptor

 Objective: To investigate the effects of curcumin derivative WZ01 on asthmatic airway inflammation, inflammatory mediators IL-13 and histamine, and glucocorticoid receptor (GR). Methods: Thirty mice were randomly divided into: control group, asthma group, low dose WZ01 group, moderate dose WZ01 group and high dose WZ01 group. Each group was 6 mice, respectively. After sensitized with OVA, the mice were challenging with 1% OVA from the 21th to 27th day. Each mouse was intraperitoneally injected with corresponding drugs or normal saline half an hour before atomizing. After the last challenging, mice were anesthetized and sacrificed within 24 hours, lung tissues were obtained for detecting inflammatory alteration with hematoxylin-eosin staining, and the BALF for cellular classification and counting and the concentrations of IL-13 and histamine by ELISA assay, the expression of GR in lung tissues was detected with immumohistochemical staining and Western blot. Results: Compared with the normal control group, HE staining showed massive inflammatory cellular infiltration and mucous secretion accumulated in the asthma group, ELISA analysis showed significantly increased number of inflammatory cells and concentrations of IL-13 and histamine in the BALF (P<0.01). Compared with the asthma group, each dose of WZ01 markedly inhibited the inflammatory cellular infiltration and mucous secretion in the airway; In each dose WZ01 group, the number of lymphocytes and eosinophils in BALF were distinctly decreased (P<0.01), accompanied by the remarkable decline of the concentrations of IL-13 and histamine (P<0.01); the number of lymphocytes had remarkably positive relationships with the concentrations of IL-13 and histamine in the BALF (r values were 0.886 and 0.886, respectively, P<0.01). The number of esoinophils had also remarkably positive relationships with the concentrations of IL-13 and histamine in the BALF (r values were 0.897 and 0.898, respectively, P<0.01). The results of immumohistochemical staining and Western blot showed that GR expression in the lung tissues of asthmatic mice was decrease. Compare with the asthma group, different doses of WZ01 could predominantly reverse and up-regulate the expression of GR in the lung tissues of asthmatic mice. Conclusion: WZ01 can significantly inhibit the inflammatory cellular infiltration and mucus secretion in asthma, which may be attributable to the decline of the inflammatory mediators IL-13 and histamine and the up-regulated expression of GR in the lung tissues of asthmatic mice. WZ01 may be a new drug of prevention and treatment for airway inflammation in asthma.

503 The follow-up analysis of MR imaging in MELAS syndrome

Objective: To analyze the MRI features of dynamic change of mitochondrial encephalomyopathy, lactic acidosis and stroke-like episodes (MELAS). Methods: The clinical data and imaging findings of 15 cases of MELAS patients were retrospectively analyzed. All of patients underwent MRI examinations (including T1WI, T2WI, T2FLAIR and DWI) in acute attack stage (within 30 days). Forteen cases underwent MRI scanning after the acute attack stage (between 30 days~60 days), of which 8 cases were examined on MRS. Ten cases were repeated the MRI examinations in remission stage (>60 days). The location, range, signal intensity and dynamic change of the lesions were intensively analyzed. Results: In acute attack stage (within 30 days), 46.7% (7/15) unilateral cortical or subcortical cortex were affected, 53.3% (8/15) on bilateral cortical or subcortical cortex, 4 cases on bilateral basal ganglia and 1 case on brainstem. The gray matter of all 15 cases was hypointense on T1WI, hyperintense on T2WI and isointense or hyperintense on DWI. After the acute attack stage (between 30 days and 60 days) of the 14 cases, the lesions expanded in 64.3% cases (9/14), shrinked in 71.4% cases (10/14), and new foci emerged in 42.9% cases (6/14). Lactate peak increased in the fresh lesions of 8 cases, and the Lac peak increased in the contralateral area in 2 cases, while the nitrogen-acetyl aspartic acid (NAA) peaks decreased in 6 cases. In remission stage (>60 days), the atrophy was found in 40% cases (4/10) and encephalomalacia foci were found in 40% cases. Conclusion: Transient lesions are easily referred to the cerebral cortex and subcortex in MELAS patients. MRS shows prominent increasing peak. Brain atrophy in the MELAS patients develops gradually and stroke-1ike lesions shrink with progression of the disease. Combined with clinical data, the accuracy of early diagnosis of MELAS syndrome can be improved.

507 Increasing CXCR7 expression in endothelial progenitor cells can enhances its angiogenic capability

Objective: To investigate whether increasing CXCR7 expression in endothelial progenitor cells (EPCs) can improve its angiogenic capability. Methods: CXCR7high-EPCs were constructed by transfecting with adenovirus carring human CXCR7 gene. The differences between CXCR7high-EPCs and wt-EPC in cell survival under serum deprivation condition, adhesion, SDF-1 induced trans-endothelial migration, tube formation and nitric oxide (NO) production were compared. Results: Compared with ctrl-EPCs, CXCR7high-EPCs has superiority in cell survival under serum-free condition, cell adhension to activated endothelial cells, trans-endothelial migration, tube formation and NO production, which could be further enhanced by SDF-1. Conclusion: Increasing expression of CXCR7 can enhance the angiogenic capacity of EPCs. The performance of present research may provide a new idea for the treatment of vascular diseases.

513 Stress induces the perception of complex tinnitus

Objective: To explore stress, referred to as a critical factor, induces the perception of complex tinnitus. Methods: One hundred and eighty-three participants were categorized into 3 groups, namely simple tinnitus (n=45) group, complex tinnitus (n=88) group and normal group (n=50). All subjects were required to use the social readjustment rating scale (SRRS) to evaluate stressful life events (stress) over the preceding 12 months. Tinnitus Handicap Inventory (THI) was used to assess the severity of tinnitus. Results: The mean of SRRS score of the complex group was obviously higher than that of the simple group and the normal group (P<0.001), meanwhile, the Grade 2 of SRRS score (150-299) was independently associated with the onset of complex tinnitus (OR: 11.921, 95%CI: 3.613~39.333). The score of SRRS was positively correlated with THI score. Conclusion: Stress, as a critical factor, induces the perception of complex tinnitus.

517 Safety and short-term efficacy of direct percutaneous lauromacrogol foam hardening agent injection for the treatment of superficial arteriovenous malformations

Objective: To study the feasibility of direct percutaneous lauromacrogol foam hardening agent injection for the treatment of superficial arteriovenous malformations, and evaluate it’s short-term efficacy and safety. Methods: From March 2015 to November 2015, 21 superficial arteriovenous malformations patients were treated with lauromacrogol foam hardening agent. Among the patients, 12 were male and 9 were female. Their ages were ranged from 1 week to 56 years old, average age was 37.1 years. Seven patients of them were in the quiescence stage, other 14 patients were in the expansion stage. The treatments were divided into several times. All patients were clinically followed-up in preoperation and postoperation by the related auxiliary examination. The effect was respectively evaluated depending on the basis of the improvement of clinical symptoms and imaging review of lesions fading condition. Results: There were 71 times in the treatment of direct percutaneous lauromacrogol foam hardening agent injection for 21 patients in total, 2-5 times per patient, average 3.4 times per patient. Two cases were cured, 18 cases got better, 1 case was invalid, and the total effective rate was 95.2%. One case got postoperative pain, relieved after 1 month by himself, 21 patients had no serious postoperative complications. Conclusion: Direct percutaneous lauromacrogol foamhardening agent injection treatment is expected to be one of the most important treatments for early stage superficial arteriovenous malformations.

521 Clinical features and prognostic factors of community-acquired pneumonia with “the white lung” in the CT scan

Objective: To investigate the clinical features and the prognostic factors of community-acquired pneumonia (CAP) with “the white lung” in the CT scan. Methods: A retrospective analysis were used. Adult patients with CAP who fulfilled “the white lung”in the CT scan from Jan 2011 to Aug 2014 in the First Affiliated Hospital of Wenzhou Medical University were enrolled and detailed data were recorded, receiver operating characteristic (ROC) curve was obtained. Results: 123 patients were enrolled, 68 cases in the death group and 55 cases were survived. The mortality was 55.3%. Fever and dyspnea were most common symptoms. 34 cases detected pathogenic bacteria, the total detection rate of 27.6%. Compared the death group with the survived group: age (62.86±14.58 vs 56.21±13.90) years, PaO2/FiO2 (191.46±125.25 vs 240.37±104.55) mmHg, LDH value (715.40±414.94 vs 552.68±283.95) U/L, D-dimer value (7.02±6.94 vs 3.60±3.50) mg/L PSI scores (116.96± 37.27 vs 89.18±45.73) and CURB-65 scores (1.76±0.91 vs 1.17±1.01) were statistically significant (P<0.05). Logistic regression indicated that PSI score, the level of DD and LDH are independent valuable factors for predicting prognosis. AUCs of PSI+DD+LDH was 0.759, more higher than one factor. The sensitivity of PSI+DD+LDH was 77.1%, specificity was 65.9%. Combination of PSI score and DD and LDH improved the prognosis prediction. Conclusion: PSI score, the level of DD and LDH are independent valuable factors for predicting prognosis. Combination of PSI score and DD and LDH improved the prognosis prediction.

526 The development and evaluation of glycated albumin enzymatic determination reagent

Objective: To develop and evaluate the serum glycated albumin enzymatic determination reagent. Methods: The analysis was based on the fact that the glycated albumin glycated albumin was hydrolyzed to glycated amino acids by special proteinase digestion, and glycated amino acids were oxidized to produce hydrogen peroxide, which was quantitatively measured through the Trinder’s chromogenic reaction, compared with the same treatment of the standard solution. Results: The within-run and between-run CVs of this reagent were 0.84%~1.88% and 1.14%~2.35%, respectively; the method was liner in the range of 1.3~1 200 μmol•L-1, percent deviation of the measured results of diluted serum samples diluted by 2 to 16 times compared to the results of the original samples ranged from 99.3%~108.6%, the average rate of recovery was 101.8%; No significant interference (bias%≤±5%) was found in the test of 550 μmol•L-1 GA analysis when bilirubin <35 mg•dL-1, triglyceride<750 mg•dL-1, hemoglobin <200 mg•dL-1, bile acid <8 mg•dL-1, uric acid <33 mg•dL-1, glucose <1800 mg•dL-1, Vitamin C <15 mg•dL-1, EDTA•K2 <2.5 mg•mL-1, Heparin Sodium <80 U•L-1, Sodium citrate <8.0 mg•mL-1. Comparing this methods (Y) with the ASAHI method (X), the regression equations is Y=0.9991X+0.0809, r=0.9958. The reagent was stable at least 30 days when it was open cover stored and at least 12 months when it was sealed stored at 4 ℃. Conclusion: This reagent shows high precision, wide linear range, and the dilution accuracy can meet the clinical needs. It also shows high recovery, good anti-interference capability, good correlation with imported reagent, and suitable for clinical application.

530 Comparison of high frequency ultrasound and upper gastrointestinal barium examination in diagnosis of upper gastrointestinal obstruction in neonates

Objective: To explore the value of high frequency ultrasound and upper gastrointestinal barium examination for the diagnosis of upper gastrointestinal obstruction in neonates. Methods: A retrospective analysis was performed to review high frequency ultrasound and upper gastrointestinal barium examination of 245  neonates with upper gastrointestinal obstruction were confirmed by post-operative pathology in Neonatal Ward of the Second Affiliated Hospital of Wenzhou Medical University during November 2008 to July 2015. The two methods were compared in the location and the etiological factor with upper gastrointestinal obstruction respectively. Results: Between high frequency ultrasound and upper gastrointestinal barium examination, there were no significant difference in the location diagnostic accuracy with upper gastrointestinal obstruction (95.1% vs 97.1%, P>0.05), but there were significant difference in the etiological factor with upper gastrointestinal obstruction, and high frequency ultrasound were better (P<0.05), especially showed in pylorus level (including congenital hypertrophy or stegnosis of pylorus, atresia or stegnosis of pyloric canal). Otherwise in the location and the etiological factor of esophagus level obstruction, upper gastrointestinal barium examination were better than high frequency ultrasound (P<0.05). Conclusion: In the diagnosis of upper gastrointestinal obstruction, we can firstly consider high frequency ultrasound and combination of the two examination when vomiting of unknown cause in high frequency ultrasound.

533 Association between plasma Hcy and bone mineral density in elderly male patients with type 2 diabetes mellitus

Objective: To investigate the correlation between plasma Hcy and bone mineral density (BMD) in elderly male patients with type 2 diabetes mellitus (T2DM). Methods: Forty-five elderly male patients with T2DM were divided into osteopenia group (group A, 24 patients) and normal group (group B, 21 patients) according to the level of BMD. Correlations between Hcy, body mass index (BMI), blood calcium, blood phosphorus, total cholesterol (TC), triglyceride (TG), high density lipoprotein cholesterol (HDL-C) and low density lipoprotein cholesterol (LDL-C) were analyzed in group A and groupB. Results: BMD and BMI in group A were significantly lower than those in group B (P<0.05). Hcy in group A was significantly higher than that in group B (P<0.05). Blood calcium, blood phosphorus, TC, TG, HDL-C and LDL-C had no statistically significant difference between two groups (P>0.05). Pearson correlation analysis showed that BMD was positively correlated with BMI (r=0.26, P<0.05), and negatively correlated with Hcy (r=-0.579, P<0.05). Conclusion: The decline of BMI and incline of plasma Hcy may associate with decreasement of BMD in elderly male patients with T2DM.

539 Therapeutic effect of acupoint injection combined with alendronate sodium in the treatment of low back pain in osteoporosis

Objective: To observe the therapeutic effect of acupoint injection combined with alendronate sodium in the treatment of low back pain in osteoporosis. Methods: One hundred cases were randomly divided into the experimental group and the control group, and were treated with acupoint injection combined with alendronate sodium and only alendronate sodium treatment, respectively. The low back pain, physical function, bone mineral density (BMD), and bone conversion biochemical markers changes were detected in 1 month and 3 months after treatment. Results: The low back pain and physical function displayed significant difference between the experimental group and the control group (P<0.05) in 1 month after treatment. But, BMD and bone conversion biochemical markers changes had no significant difference between the two groups (P>0.05) in 3 months after treatment. Conclusion: The treatment of acupoint injection combined with alendronate sodium has an excellent short-term curative effect on osteoporotic patients.
