1 The protective effect of WNK3 kinase on apoptosis in HEK293 cell induced by interleukin-1β

Objective: To observe the effect of high expression of WNK3 on apoptosis in HEK293 cells induced by interleukin-1β. Methods: The HEK293 cells were divided into 3 groups (Vector+NS group,Vector+IL-1β group and WNK3+IL-1β group), then tansfection was performed. Liposome 2000 was added to the 1st and 2nd group with PCMV-HA-Vector as the blank and negative control respectively; WNK3 was added to the 3rd group. After treated with 10 ng/mL IL-1β (the 2nd and 3rd group) or the same amount of normal saline (the 1st group), the CCK8 analysis was performed for the detection of cell activity after incubation for 0 h, 12 h,24 h, 36 h and 48 h at 37 respectively. Cell proteins were collected after 0 h, 18 h and 36 h incubation respectively for detection of Caspase-3, Cleaved Caspase-3, Caspase-9, Cleaved Caspase-9. In addition, cell protein were collected 0 min, 30 min and 60 min after the treatment of IL-1β for JNK detection. Results: No significant difference of cell activity was observed in the Vector+NS group within 48 h. In the Vector+IL-1β group, cell activity began to decline soon after administration. Significant difference could be found after 24 h and the cell activity reached the lowest at 48 h (P<0.01). Cell activity of WNK3+IL-1β group declined in a moderate manner and the significant difference appeared at 36 h and 48 h when comparing with that at 0 h in the same group (bothP<0.05). However, in comparison with the Vector+IL-1β group at the same time point, cell activity of WNK3+IL-1β group was still a little higher and the significant difference could be observed at 48 h (P<0.01). The expression of Caspase-9 and Caspase-3 began to decline 18 h after the administration with an increasing peak of cleaved Caspase-9 which decreased slightly at 36 h. An obvious cleaved Caspase-3 could also be observed 36 h after the administration. In the WNK3+IL-1β group, the decline of Caspase-3 was more moderate when comparing with that in the Vector+IL-1β group. A small quantity of cleaved Caspase-3 could be detected and showed an statistical significance in comparison with that of the same group at 0 h (P<0.05) with the WNK3; the expression of p-JNK declined but t-JNK did not change. Conclusion: IL-1β may decrease activity of HEK293 cells while transfection of WNK3 may partly reverse this affection of IL-1β. The over expression of WNK3 may inhibit the activation of JNK pathways induced by IL-1β, decrease the activation of Caspase pathway to reduce apoptosis, and finally protect the kidney cells under adverse conditions.

7 The effects of baicalein on promoting the osteogenesis of human periodontal ligament cells

Objective: To evaluate the effects of baicalein extracted from Scutellaria baicalensis georgi on the function of proliferation and osteogenesis of human periodontal ligament cells (hPDLCs). Methods:Baicalein at a concentration of 0.3125, 0.625, 1.25 μmol/L was added into hPDLCs primary cultured in vitro accordingly as experimental group, the one with no baicalein as blank control group. The effect of proliferation of hPDLCs was assessed through MTT assay. Alkaline phosphatase (ALP) activity assay, Alizarin red S staining,RT-qPCR and Western Blot were used to observe the ALP activity, formation of mineral nodules, mRNA and protein expression of collagenase-Ι (Col-Ι) of hPDLCs, respectively. Results: Compared to the control group,baicalein at different concentrations had no significant influence on the proliferation of hPDLCs. Baicalein was shown to enhance the ALP activity of hPDLCs. After 21 days of osteogenic induction, more mineral nodules could be observed compared to the control group, which increased in a dose-dependent manner at the concentration of 0.3125-1.25 μmol/L baicalein according to the results of quantification. The mRNA and protein level of Col-Ι increased by baicalein in different concentration, which reached maximum at 7 days, appearring in a dosedependent manner from 0.3125 to 1.25 μmol/L using Western Blot and Real-time PCR analysis. Conclusion:Baicalein can promote hPDLCs to differentiate into osteoblasts, which is of great value for further study acting as
the adjuvant drug for periodontal regenerative therapy.

13 Remodeling of parasympathetic/sympathetic ratio in the infarcted myocardium after Schwann cell transplantation

Objective: To investigated the feasibility of remodeling nervous system regeneration after myocardial infarction and the efficacy by which it may prevent ventricular arrhythmia following Schwann cells transplantation. Methods: Myocardial infarction was induced by left anterior descending artery ligation in syngenic Lewis rats, then the rats were randomized into cell transplantation group (n=31) and control group (n=30).Schwann cell were isolated from sciatic nerves, and immunohistochemical of S100 was carried out for the identification of Schwann cells. 5×106 Schwann cell were intramyocardially injected into the border zone of infarct region. The density of sympathetic and parasympathetic nerves and growth-associated protein 43 in the infarct region were stained by immunohistochemical method and immunofluorescence at 2 weeks after Schwann cells transplantation. Meanwhile, dynamic electrocardiography and programmed electric stimulation were also performed to detect the susceptibility of ventricular arrhythmias. Results: More than 95% of cells were S-100-positive Schwann cells during the entire culture process. Immunohistochemical and immunofluorescence staining illustrated increases in sympathetic and parasympathetic nerves in both groups. However, Schwann cells transplantation significantly increased the parasympathetic/sympathetic ratio at 2 weeks after cell injection. In addition,the Schwann cells also significantly decreased the low-/high-frequency ratio and susceptibility of programmed electric stimulation-induced ventricular arrhythmia at 2 weeks after cell injection. Conclusion: Transplanted Schwann cells in the infarcted myocardium can alter the ratio of parasympathetic/sympathetic nerve density to normalize irritable myocardium.

19 Synthesis of triethylenetetramine-starch and its antitumor effects#br#
CHENG Rumei, BU Yexu.
Objective: To design and synthesize the potential antitumor drug, CTS (triethylenetetraminestarch). Its interactions with the human telomere DNA (d[g3(T2Ag3)3 series]) and cytotoxicity to oCM-1 tumor cells are investigated. As a consequence, it provides a new strategy to develop targeted antitumor drugs. Methods: The triethylenetetramine reacted with the epichlorohydrin crosslinked-starch (CS) to produce the CTS. It was characterized by various methods. The circular dichroism (CD) spectra were used to clarify the interaction mechanism between the CTS and human telomere DNA. Its cytotoxity to oCM-1 cells was determined by CCK-8 method. Results: The antiparallel g-quadruplex could be induced and stabilized by CTS. This may be the reason that CTS inhibited the growth of oCM-1 cells, and moreover it showed very low cytotoxicity to ARPE-19 human cells. Such observations indicated that CTS was a promising antitumor drug. Conclusion: The CTS can stabilize the g-quadruplex and has higher cytotoxicity to oCM-1 cells. It has insignificant effects on the growth of normal human cells.

24 The expression of IL-17 in the cervical cancer with high-risk human papillomavirus infection#br#

Objective: To investigate the expression of IL-17 in patients diagnosed as cervical squamous cell carcinoma (SCC) with high-risk human papillomavirus (HPV) infection, with the aim to explore the potential role of specific immune response mediated by IL-17 in the disease progression and carcinogenesis of cervical cancer which is closely associated with high-risk HPV infection. Methods: The study groups included 28 cervical SCC patients with high-risk HPV infection and 25 high-risk HPV infected women with normal cervical cytology. Thirty patients diagnosed as uterine myoma with normal cervical cytology and negative HPV testing were enrolled as the control group. The concentration of IL-17 in serum and supernatant of cervical tissue homogenate were examined with ELISA method. Results: The serum concentrations of IL-17 in patients with cervical SCC were significantly higher than those in the control group (P<0.05), but there was no significant difference observed between SCC group vs the high-risk HPV infection group (P>0.05) and the high-risk HPV infection
group vs the control group (P>0.05); The average concentrations of IL-17 in supernatant of cervical tissue homogenate of all groups were obviously higher than those in serum respectively (P<0.05); The concentrations of IL-17 in supernatant of cervical tissue homogenate differed significantly among the study groups and the control group (P<0.05). The concentrations in the cervical SCC group were higher than those in the high-risk HPV infected group, which were much higher than the control group (P<0.05). Conclusion: These results indicate that the immune reaction mediated by IL-17 may participate in and promote the persistent infection of high-risk HPV,which is closely associated with the disease progression and carcinogenesis of cervical cancer cervical cancer.

28 The relationship between bone density and marrow fat in postmenopausal females with type 2 diabetes#br#

Objective: To explore the relationship between vertebral bone density (BMD) and marrow fat fraction (FF) in postmenopausal females with type 2 diabetes (T2DM). Methods: Patients with T2DM (49-85 years, n=43) and healthy controls (50-77 years, n=35) underwent DXA and MR spectroscopy scanning to
obtain BMD and FF. Blood glucose (FPg), HbA1c, fasting insulin (FINS) and total cholesterol (TC), triglycerides (Tg), high-density lipoprotein cholesterol (HDL-C) and low-density lipoprotein cholesterol (LDL-C) were measured after fasting for 12 h. Homeostasis model assessment was performed to assess the status of insulin resistance (HoMA-IR). Results: Subjects with T2DM had higher vertebral BMD [(1.103±0.187)g/cm2] and FF(74.1%±16.7%) than those of healthy controls [BMD=(1.015±0.171) g/cm2, FF=63.3%±12.9%, respectively].Even controlling for years of menopause, BMI, FPg, HbA1c, HoMA-IR and blood lipids index, FF in the T2DM group remained higher than that in the control group (F=10.857, P=0.010). FF was inversely correlated to BMD In the control group (r=-0.430, P=0.014) but positively associated with BMD in the T2DM group (r=0.385,P=0.029) after adjustment for the confounders. Conclusion: Despite having higher BMD values, patients with T2DM in postmenopausal females have higher vertebral marrow fat content.

33 Effect of homocysteine on the phenotype transformation in the cultured rat vascular smooth muscle cell#br#

Objective: To verify whether homocysteine (Hcy) could induce the dedifferentiation of vascular smooth muscle cells (VSMCs). Methods: The primary culture and identification of rat VSMCs was conducted using VSMCs in passage 4-7 for the following experiments. The VSMCs were divided into 4 groups: control
group, Hcy (100 µmol/L) modulation group, Hcy (500 µmol/L) modulation group, Hcy (1 000 µmol/L) modulation group. MTT were used to investigate the proliferation of VSMCs. Transwell chambers and wound healing were employed to test the migratory ability of VSMCs. ICC were used to detect the VSMCs’ morphology structure. Western blotting was used to investigate the expressions of SM-actin, SM-MHC, Calponin, OPN in VSMCs of every group. Results: Compared with control group, the proliferation and migration ability of VSMCs were significantly increased in the Hcy modulation group (P<0.01). The expression of SM-actin had no significant difference between each group. The expression of SM-MCH and Calponin increased and OPN decreased in the Hcy group compared with control group (P<0.01). This effects of Hcy were positively correlated with its concentrations. Conclusion: Hcy can induce the dedifferentiation of VSMCs, this maybe the mechanism by which Hcy increases the proliferation and migration ability of VSMCs.

39 Pre-neoadjuvant chemotherapy 18F-FDG PET/CT examination to evaluate the postoperative radiotherapeutic value in stage II-III breast cancer #br#

Objective: To evaluate postoperative radiotherapy with 18F-FDg PET/CT before neoadjuvant chemotherapy in breast cancer patient staging II and III. Methods: We retrieved the 18F-FDg PET/CT record of 309 breast cancer patients in our hospital from July 2009 to May 2013. Distant metastases were detected in 85 patients, and 224 patients were included in this study. Conventional regional staging consisted of fine needle aspiration and/or sentinel lymph node biopsy. According to the guideline of NCCN 2013, patients were classified as low-risk group (T2N0), intermediate-risk group (T0N1, T1N1, T2N1, T3N0), or high-risk group (T3N1, T4,TxN2, TxN3). The presence and number of FDg-avid nodes were evaluated and the proportion of patients that would be upstaged by 18F-FDg PET/CT, based on detection of 4 FDg-avid axillary nodes defined as petN2(4+) or occult petN3-disease. Results: 18F-FDg PET/CT detected occult N3-disease in 11.4% (4/35) of 35 lowrisk patients. In 123 intermediate-risk patients, 18F-FDg PET/CT detected4 FDg avid nodes in 20 patients and occult N3-disease in 16 patients, thereby finally upstaging 25.2% (31/123) of the patients. Conclusion: 18F-FDg PET/CT imaging before neoadjuvant chemotherapy can accurately assess the patient’s risk of recurrence. For the high-risk group of patients, 18F-FDg PET/CT can detect occult lymph nodes and provide powerful guarantee for ascertainment of the range of radiation field.

44 Epidemiological investigations of perioperative risk factors for early death in newborns and infants

Objective: To analyze clinical epidemiological characteristics with perioperative death associated risk factors of children under 1 year of age in the early period (less than 3 days). Methods: The clinical data from 76 cases of neonates and infants dead during perioperation from January 2011 to December 2013 were investigated in the Second Affiliated Hospital of Wenzhou Medical University. Retrospective analysis were conducted on their gender, gestational age, onset age, birth weight, preoperative weight, preoperative complications (such as cardiovascular disease, respiratory diseases, blood disorders, liver and kidney disease), preoperative conscious state, electrolyte imbalance or not, the malnutrition or not, ASA classification, site of surgery, type of surgery, duration of surgery and operation classification. Results: Spearman rank correlation analysis was performed respectively among birth weight, preoperative respiratory disease, ASA classification grade and preoperative state of consciousness with death in early time. P value was lower than 0.05, according to P<0.05 statistical standards, it had statistical significance. All the four risk fators above-mentioned entered into Logistic stepwise regression analysis, and finally only two risk factors (preoperative respiratory diseases and ASA classification)were reserved. From the area under the RoC curve analysis that neonates and infants during perioperative period of early (less than 3 days) time of death risk factors were highly correlated in preoperative respiratory disease(0.643), ASA classification (0.694). Conclusion: Preoperative respiratory disease and ASA classification are the effective predictor of perioperative early death in neonates and infants, and predictive value of ASA classification was stronger.

48 Application of transorally inserted anvil delivery system (OrVil) for totally laparoscopic radical gastrectomy

Objective: To evaluate the technical feasibility, safety, and clinical efficacy of the technique of transorally inserted anvil delivery system (orVil) for totally laparoscopic gastrectomy. Methods: From January 2014 to September 2014, 45 gastric cancer patients underwent D2 lymph node dissection. of these, 20 patients underwent transoral orVil EEA stapler for totally laparoscopic gastrectomy (orVil group), while 25 patients underwent conventional open gastrectomy (og group). The recovery and complication rate were compared between two groups. Results: Transoral orVil EEA stapler for totally laparoscopic gastrectomy was successfully carried out in all the patients except for 1 case who was convened to open surgery. The operation time, the mean number of removal lymph node and number of patients needed blood transfusion were not significantly between two groups. However, compared to og group, the mean blood loss were less, and first flatus time, time to resume soft diet and postoperative hospital stay were shorter in OrVil group (all P<0.05). The postoperative morbidity in orVil group and og group were 10% and 20%, respectively (P>0.05). Conclusion: OrVil is a safe and feasible procedure for totally laparoscopic Radical gastrectomy and provides satisfactory short-term efficacy.

53 Changes of plasma neuroglobin and S-100β protein concentration in patients undergone coronary artery bypass grafting surgery under cardiopulmonary bypass

Objective: To investigate the changes of plasma neuroglobin and S-100β protein concentrations in patients undergone coronary artery bypass grafting surgery under cardiopulmonary bypass (CPB). Methods:Forty patients, American Association of Anesthesiologists (ASA) and New york Heart Association (NyHA) status II or III, the Mini-Mental State Examination (MMSE, range 030) score26, undergoing coronary artery bypass grafting surgery with CPB were enrolled. Blood samples were collected immediately at anesthesia induction (T0), CPB re-warming to 36 (T1) and 1 h (T2), 6 h (T3) and 24 h (T4) after CPB, plasmas were obtained for further determination of neuroglobin and S-100β protein concentration. MMSE were measured preoperatively
and three days after operation. Results: Compared with values in T0, the plasma concentrations of neuroglobin and S-100β protein was significantly increased at T2-T4 (P<0.05), with peaking at T3. MMSE scoring at 3 d after operation was lower than preoperative baseline (P<0.05). Conclusion: Cognitive function is impaired and the plasma neuroglobin and S-100β protein levels are increased in patients undergone coronary artery bypass grafting surgery under CPB, these two can be considered as indicators of cerebral injury after cardiopulmonary bypass.

56 Expression of CD147 and Survivin in renal cell carcinoma and their correlation with clinicopathologic parameters

Objective: To investigate the expression of CD147 and Survivin in human renal cell carcinoma(RCC) and their correlation with clinicopathologic parameters. Methods: The RCC tissue microarrays containing 85 renal cell carcinoma specimens and 35 normal kidney tissue specimens were used to detect CD147 and Survivin expression by immunohistochemistry. Results: The expression rates of CD147 and Survivin were 65.7%(23/35) and 71.4% (25/35) in RCC group, respectively, significantly higher than that in control group (P<0.01).The expression of CD147 in RCC increased with the size of tumor and pathological grade (P<0.05). And the expression of Survivin in RCC increased with the pathological grade and clinical stage (P<0.05). The expression of CD147 and Survivin in RCC specimens showed weak consistency (Kappa=0.388, P<0.05). Multivariate logistic regression analysis showed that the positive expression of Survivin was an independent risk factor of RCC (oR=16.256, 95%CI: 1.132~ 233.515, P<0.05). Conclusion: CD147 and Survivin are over-expressed in
RCC specimens. CD147 and Survivin may play a crucial stage in development of human RCC. Detection of the expression of CD147 and Survivin can be used in the evaluation of malignant degree and prognosis of RCC.

60 Contrastive analysis of the foxic and adverse reaction of DOF vs DOX in the treatment of advanced unresectable gastric cancer

Objective: To contrastive study the toxic and adverse events of DoF and DoX scheme in the treatment of advanced unresectable gastric cancer. Methods: Clinical data of 69 advanced gastric cancer, including 31 treated in DoF and 38 in DoX, were analyzed retrospectively in order to compare the adverse event rates and severity level in both groups. Results: Both groups underwent 3 to 8 cycles, there were 189 cycles in DoF and 233 cycles in DoX respectively. The total adverse event rates were undifferentiated at 80.0% and 81.5% when statistical analyzed by cycles (P>0.05). But the total hematologic toxicity in DoF were higher than those in DoX, especially neutropenia and thrombocytopenia which respectively were 48.7% vs 33.5% and 33.3% vs 18.0% (P<0.05). Though the total non-hematologic toxicity were similar in both groups, the stomatitis and peripheral phlebitis were higher in the former, while opposite to the hand-foot syndrome and nail toxicity, the rates respectively were 41.8% vs 15.0%, 16.4% vs 3.4%, 6.9% vs 50.6% and 4.8% vs 36.5% (P<0.05). We concluded the same results that the total hematologic toxicity and its 3/4 grade in DoF were higher than those in DoX (P<0.05), however the total adverse event rates and the total non-hematologic toxicity were similar in both groups (P>0.05), while the data were statistical analyzed by patients. Conclusion: Though the total adverse event rates are undifferentiated, the hematologic toxicity, stomatitis and peripheral phlebitis are more frequently occurred in DoF, and the hand-foot syndrome and nail toxicity are higher in DoX.

65 The value of the pseudo deta wave (PdW) in the diagnosis of premature ventricular contractions

Objective: To assess the value of the pseudo deta wave (PdW) in the diagnosis of different positions of premature ventricular contractions (PVCs). Methods: The radiofrequency ablation were performed.Then the length of the PdW in different positions of PVCs in totally 97 patients with PVCs was measured. Results: The originating of PVCs: In 13 cases it was originating from the endocardium of the left ventricle. In 52 cases it was originating from the endocardium of the right ventricle. In 27 cases it was originating from the aortic sinus. In 5 cases it was from the great cardiac vein. The length of the PdW in the 4 groups was different (P<0.05). The length of the PdW from the sinus of valsalva and great cardia vein was different (P<0.05).The length of the PdW from the endocardium and epicardium according to the position of the originating site was different (P<0.05). Conclusion: The length of the PdW can be used in indentifying PVCs not only arising from the endocardium and epicardium, but also arising from the sinus of valsalva and great cardia vein.

68 The feasibility analysis of luteal-phase ovarian stimulation after the dominant follicle ovulated in unsynchronized multiple folliclar development patients

Objective: To explore the feasibility of luteal-phase ovarian stimulation after the dominant follicle ovulated in unsynchronized multiple folliclar development patients undergoing routine controlled ovarian hyperstimulation (CoH). Methods: From May 2012 to February 2014, 35 infertile patients undergoing in-vitro
fertilization (IVF) in Reproductive Medicine Center, the First Affiliated Hospital of Wenzhou Medical University were enrolled in the study. When there comes unsynchronized multiple folliclar development during the routine CoH, continue using gn until the follcle matured after the dominant follicle ovulated and serum progesterone exceeded 8 nmol/L. The highest quality embryos were extracted and cryopreserved for the next frozen embryo transfer (FET) cycles. Analyze the number of oocytes retrieved, implantation rate, clinical pregnancy rate, ongoing pregnancy rate. Results: all patients succeeded in producing oocytes except one, a total of 412 ocytes were retrieved, the average number of oocytes retrieved was 12.12±11.09, producing 167 highest quality embryos,
fertilization rate was 75.40%, cleavage rate was71.12%. A total of 158 ocytes were cryopreserved for later transfer. one patient quitted cycles because of embryos’ thawing failure. A total of 34 women underwent a total of 36 FET cycles, including 33 patients who completee one FET, 3 who finished two FETs, and a total of 74 embryos were transferred.In FETs, the implantation rate and cumulative pregnancy rate were 37.83%, 52.78%, respectively. one woman was reported to have miscarried in the first trimester (5.26%). of the 19 pregnancies in the study, 10 resulted in live births and 8 were ongoing. Delivery follow-up showed 3 twin births and 7 single births,no malformations were reported in the newborns. Conclusion: Luteal-phase ovarian stimulation is feasible for producing competent oocytes/embryos in women undergoing routine controlled ovarian hyperstimulation (CoH)after the dominant follicle ovulated in unsynchronized multiple folliclar development, with optimal pregnancy outcomes in FET cycles. It’s a safety and exciting remedial measure.

72 Expression and correlation of EGFR and FSCN1 in cervical cancer

Objective: To explore the expression of EgFR and FSCN1 protein in cervical cancer and its correlation. Methods: The expression of EgFR and FSCN1 in 120 cases of cervical cancer was evaluated with immunohistochemical technique, EnVision method. Results: EgFR protein expression rates in cervical cancer
was 89.2% (107/120). FSCN1 protein positive rate in cervical cancer was 99.2% (119/120), in which strong positive rate was 88.3% (106/120). EgFR positive expression was significantly positive correlation with FSCN1 strong positive expression (P<0.01). Conclusion: EgFR may participate in positive regulation FSCN1 expression in cervical cancer.

75 Epidemic characteristics of HFMD in Pinghu City from 2010-2014

Objective: To provide scientific evidences for developing control measures and strategies, according to the epidemic characteristics of Hand foot mouth disease (HFMD) in Pinghu City from 2010-2014.Methods:Descriptive method was adopted to analyze the epidemiological characteristics of HFMD. Results: A total of 6 466 HFMD cases were reported in Pinghu City from 2010-2014, and the average incidence rate was 189.96/100 000, the lowest (80.86/100 000) and highest (351.05/100 000) incidence rate appeared in 2011 and 2014. There was a seasonal tendency in HFMD prevalence with the main peak in spring from summer (from April-July) and the second one in autumn (from September-November). Most cases were children aged 5 years or below (92.51%). Etiology results showed CoxA6 was the main pathogen from 2010 to 2012, but other enteroviruses become main pathogen between 2013 and 2014. Conclusion: The epidemic situation of HFMD is serious in Pinghu City, and therefore, prevention and control of HFMD should focus on the key population and important places to control the epidemic.
