781 Establishment and characterization of a Paraquat-resistant human lung adenocarcinoma cell line A549/PQ

Objective: To establish Paraquat (PQ)-resistant A549 cell line (A549/PQ) and explore its biological characteristics. Methods: The A549 cells were periodically co-cultured with PQ at gradually increasing concentrations, and its biological characteristics were observed. The morphological features were checked with light microscope. Sensitivity to PQ was detected by LDH assay. The cell growth curve and doubling time were studied by cell counting. CCK-8 assay was used to measure the median inhibitory concentration (IC50) and resistance index (RI) of A549/PQ cells and flow cytometry was applied to evaluate the cells cycle distribution. Results: Compared with A549 cells, the changes for larger size, irregular shape and much wide cell gap were found in A549/PQ cells. LDH release induced by PQ in A549/PQ cells was significantly lower than that of A549 cells (P<0.05). CCK-8 assay showed higher IC50 in A549/PQ cells than that in A549 cells (1716.33 ﹠ 215.03 μmol/L, P<0.05) and RI was 7.98 in A549/PQ cells. Longer doubling time, increased G0/G1 phase cells and decreased cells in S phase were found in A549 cells (P<0.05). Conclusion: We successfully constructed a PQ-resistant A549 cell line with significantly different biological characteristics and therefore a good cellular model for PQ toxicity research.

785 Effect of HoxB8 gene on colon cancer cells growth and migration

Objective: To construct a lentiviral vector Lenti-HoxB8-GFP that containing human HoxB8 gene and investigate its role in colon cancer cells. Methods: The HoxB8 gene was amplified and cloned into a lentiviral vector by restriction endonuclease EcoR I/SAC II digestion and T4 DNA ligase ligation to form recombinant Lenti-HoxB8-GFP. The positive clones were identified by PCR and sequencing, after transformation into E. coli cells. Lentivirus were produced by co-transfection of lentiviral vector and the three packaging vector into 293T cells. The RKO cells were transduced with Lenti-HoxB8-GFP and the expression of HoxB8 was detected by Real-time PCR and western blotting Cell proliferation and colony forming ability were detected by MTT and plate clone assay. Cell migration was measured by Transwell assay. Results: The lentiviral vector Lenti-HoxB8-GFP containing HoxB8 gene was successfully constructed. Most of the cells had strong green fluorescence after transfected with the collected virus for 72 h. Real-time PCR and western blotting verified the overexpression of HoxB8 in the transfected RKO cells. Overexpression of HoxB8 promoted RKO cell proliferation and cell colony formation. Moreover, overexpression of HoxB8 increased colorectal cancer cell migration. Conclusion: HoxB8 gene promoted proliferation, cell colony formation and cell migration in colorectal cancer RKO cells.

791 The inhibitory effect of CdCl2 on DNA mismatch repair activity in ZR75-1 cells

Objective: To study the effects of CdCl2 on the DNA mismatch repair (DNA MMR) activity in human breast cancer cells ZR75-1. Methods: MTT assay was used to detect the survival rate of ZR75-1 cells exposed 48 and 72 h to 0, 10, 20, 30, 40, 50 µmol/L CdCl2. The cells from 5, 10, 15 µmol/L CdCl2 exposure (48 h) and control group were parallelly transfected with homoduplex and heteroduplex plasmids, flow cytometry analysis was then carried out to quantitatively measure the relative EGFP expression (indirectly reflected DNA MMR activity) in ZR75-1 cells of each groups. Meanwhile, real-time fluorescent quantitative PCR technique was used to detect the mRNA expression of DNA MMR related genes (MLH1, MSH2, MSH6 and PMS1) after exposure 48 h to 5, 10, 15 µmol/L CdCl2. Results: 10, 20 µmol/L CdCl2 exhibited no influence on ZR75-1 cell survival (P>0.05), while cells exposed to 30, 40 and 50 µmol/L CdCl2 displayed significant mortality which showed obvious dose-dependent and time-dependent manner (P<0.05). Compared with control group, 5, 10 and 15 µmol/L CdCl2 exposure induced significant inhibition of DNA MMR activity in ZR75-1 cells (P<0.05). Real-time PCR results showed that after exposure to 5, 10 and 15 µmol/L CdCl2 for 48 h, the mRNA expression of detected genes (MLH1, MSH2, MSH6 and PMS1) was increased as a whole, which presented down after rising first with CdCl2 exposure concentration increase. Conclusion: Low dose of CdCl2 exposure which showed no effect on cell survival can represse DNA MMR activity in ZR75-1 cells, and up-regulate the mRNA expression of DNA MMR related genes.

796 Expression of MMPs, TIMPs and inflammatory factors in habitual abortion cases

Objective: To investigate the expression of MMPs, TIMPs, Oxidative stress and inflammatory factors in serum and endometrial tissue of habitual abortion cases and to evaluate their clinical significance. Methods: From Oct. 2012 to Oct. 2014, 32 habitual abortion pregnant women were recruited as observed group, meanwhile 35 pregnancy abortion women were enrolled as control group. The expression of MMP-2, MMP-9, TIMP-2, TIMP-1 in serum and endometrial tissue were measured by ELISA. The expression of inflammatory factors in endometrial tissue including IL-6, TNF-α, MDA, SOD and GSH-PX were also detected by ELISA. Finally, the correlations of them were analyzed. Results: Compared to control group, the expression of MMP-2 in serum of observed group decreased obviously (P<0.05), MMP-9, TIMP-2 increased significantly (P<0.05); the expression of MMP-2 and MMP-9 of observed group in endometrial tissue declined obviously (P<0.01), while TIMP-2 improved remarkably (P<0.05); the expressions of MDA, IL-6, TNF-α of observed group in endometrium tissue were upregulated (P<0.05), but SOD was downregulated (P<0.05); in endometrial tissue of observed group, there were positive correlation between MMP-2, MMP-9 with SOD, GSH-PX, and between TIMP-2 with MDA, TNF-α. Conclusion: The imbalance of MMPs/TIMPs system and abnormal expression of inflammatory factors may be associated with habitual abortion.

799 Correlative analysis between erythrocyte parameters and cognitive functions in male alcohol-dependent patients

Objective: To investigate whether male alcohol-dependent patients had cognitive impairments, and cognitive impairments were correlated with erythrocyte parameters in male alcohol-dependent patients. Methods: Sixty adults male, including 30 alcohol-dependent patients and 30 healthy controls in Wenzhou Kangning Hosipital Affiliated to Wenzhou Medical University were enrolled. Cognitive functions in all subjects were assessed by the Repeatable Battery for the Assessment of Neuropsychological Status (RBANS). Erythrocyte parameters were examined in all subjects. Analysis of variance was conducted to explore difference in both cognitive functions and erythrocyte parameters between alcohol-dependent patients and controls, and the influence of erythrocytes, mean corpuscular volume and hemoglobin on cognitive functions in alcohol-dependent group were examined using pearson correlation analysis. Results: There were significantly difference in immediate memory, visuospatial, language, attention, delayed memory and total score of RBANS between alcohol-dependent patients and controls (all, P<0.01). Alcohol-dependent group had lower erythrocytes and higher mean corpuscular volume compared to controls (both, P<0.01). There was a positive correlation between erythrocytes and delayed memory (P<0.01). Mean corpuscular volume was negatively correlation with language (P<0.05), delayed memory (P<0.01), and total score of RBANS (P<0.05). Conclusion: Male alcohol-dependent patients have cognitive impairments. Moreover, erythrocytes and mean corpuscular volume influence cognitive impairments in male alcohol-dependent patients.

803 The construction of SW480 cells nude model transfected by hNIS gene and radionuclide imaging

Objective: To examine the feasibity of radionuclide imaging mediated by colon cancer cells (SW480) nude model transfected with a recombinant pcDNA3.1+-hNIS plasmid. Methods: pcDNA3.1+-hNIS was confirmed by restriction enzyme digestion, the hNIS gene and protein expression were detected after transfection, the SW480 cells of stably expressing hNIS were screened, the tumors nude mice model was established, then radioactive nuclide 99mTcO4- imaging was obtained. Results: After SW480 cells was transfected with pcDNA3.1+-hNIS, the hNIS gene and prote in expression were successfully detected. Radionuclide imaging was successfully mediated by colon cancer cells nude model. Conclusion: The hNIS expression can be achieved in colon nude model by the recombinant plasmid pcDNA3.1+-hNIS, and make the radionuclide imaging and radionuclide therapy of colon cancer as possible.

807 The effect of rosuvastatin on prognosis in elderly patients with severe overlap syndrome

Objective: To investigate the effect of rosuvastatin on prognosis in elderly patients with severe overlap syndrome (OS), which includes obstructive sleep apnea hypopnea syndrome (OSAHS) and chronic obstructive pulmonary disease (COPD). Methods: According to the random number table, 40 elderly patients with severe OS were randomly divided into treatment group (n=20) and control group (n=20). Rosuvastatin tablets 10 mg/qn were taken in the treatment group, while statins and other drugs which can influence blood lipid level were not taken in control group. Respectively, monitoring the parametric variation at the 1st day, 30th day, 90th day and 180th day, the parameters included triglycerides (TG), total cholesterol (TC), low density lipoprotein cholesterin (LDL-c), Lipoproteinα (LPα), apolipoprotein A1 (apoA1), apolipoprotein B (apoB), apnea hypopnea index (AHI), the mean pulse oxygen saturation (M-SaO2), the lowest pulse oxygen saturation (L-SaO2), Epworth sleepiness scale (ESS), PaCO2, PaO2, FEV1/FVC, FEV1% and the hospitalization rate of AECOPD in 180 days. Blood glucose, creatine phosphokinase, liver function and renal function, neurological abnormalities, myalgia and gastrointestinal discomfort during therapy were monitored in tow groups every month. The parameters were underwent comparison intergroup and intragroup by SPSS18.0. Results: ApoA1, M-SaO2, L-SaO2, PO2, FEV1/FVC% and FEV1% at the 30th day, 90th day and 180th day after treatment were higher than those at the 1st day in both treatment group and control group, and the differences were statistically significant (P<0.05). ApoA1, M-SaO2, L-SaO2, PO2, FEV1/FVC% and FEV1% at the 30th day, 90th day and 180th day after treatment in treatment group were higher than those in control group, and the differences were statistically significant (P<0.05). LPα, apoB, TG, TC, LDL-C, AHI, ESS and PCO2 at the 30th day, 90th day and 180th day after treatment were lower than those at the 1st day in both treatment group and control group, and the differences were statistically significant (P<0.05). LPα, apoB, TG, TC, LDL-C, AHI, ESS and PCO2 at the 30th day, 90th day and 180th day after treatment in treatment group were lower than those in control group, and the differences were statistically significant (P<0.05). The hospitalization rate of AECOPD in 180 days in treatment group were less than that in control group, and the differences were statistically significant (P<0.05). There were no abnormalities in blood glucose, creatine phosphokinase, liver function and renal function, neurological, myalgia and gastrointestinal discomfort in two groups during therapy. Conclusion: Dyslipidemia in elderly patients with severe OS can be corrected by taking rosuvastatin, which shows that rosuvastatin can improve the prognosis in elderly patients with severe OS.

811 Determination of DEHP in white spirit by ultra performance liquid chromatography

Objective: To establish a method to determine the concentration of di (2-ethylhexyl) phte (DEHP) in white spirit and study the different concentration between the same brand and the different brands. Methods: DEHP was detected in a solid phase extraction-ultra performance liquid chromatography (SPE-UPLC) method. DEHP was extracted from white spirit, using solid phase extraction column, then detected with ultra high performance liquid chromatograph. The mobile phase was acetonitrile-water and the flow rate was 0.4 mL/min. The UV detection wavelength was 224 nm. Results: The linear relation was good in this method, the RSD was less than 15%, and the recovery was between 90.08% and 103.32%. The low limit of the detection was 1 μg/mL. The samples with five different bland were all contain DEHP. Conclusion: The method is compeletely suitable for determination of DEHP in white spirit.

814 Expression and clinical significance of chemokine CXCL14 in nasopharyngeal carcinoma

Objective: To investigate the expression of chemokine CXCLl4 and its clinical significance in nasopharyngeal cancer (NPC). Methods: Thirty patients with NPC were selected as the disease group and another 30 cases with nasopharyngitis as control group. Biopsy with nasopharyngeal tissue was performed. The CXCLl4 expression was detected with real-time fluorescence quantitative PCR and immunohistochemistry. Results: The CXCLl4 expression with NPC tissues in mRNA and protein levels declined  compared with the control group (P<0.001). Correlative analysis showed the dow-nregulation of CXCLl4 expression in protein levels with NPC tissues was associated with T classification of tumor (P=0.001), lymph node metastasis (P=0.028) and the clinical stage (P=0.001). Conclusion: It is possible that CXCLl4 plays a role in the development and progression of NPC.

819 Study the influencing factors of the lower uterine segment thickness after cesarean delivery during the third trimester of pregnancy

Objective: To explore the influencing factors of the lower uterine segment thickness after cesarean delivery during the third trimester of pregnancy. Methods: 74 pregnant women undergone magnetic resonance imaging (MRI) examination at third-trimester in our hostipal from June 2011 to December 2013 were include, 34 with a history of cesarean delivery and 40 control subjects with a history of vaginal delivery, the lower uterine segment thickness was measured on MRI. The maternal age, gestational age, previous cesarean, numbers of cesareans, interval time after cesarean influence on the lower uterine segment thickness were emphatically analyzed with a linear regression model. Results: The lower uterine segment was significantly thinner in women with a previous cesarean delivery compared with control subjects (3.34±0.71 versus 3.97±1.08 mm, P<0.05). In a linear regression model, the only variable retaining significance in the prediction of uterine wall thickness was previous cesarean delivery (P<0.05). Maternal age, gestational age, number of cesareans, and interval time after cesarean did not attain significance in the model (P>0.05). Conclusion: The previous cesarean delivery is the main factor influencing the lower uterine segment thickness after cesarean delivery during the third trimester of pregnancy.

823 Ultrasound-microbubble mediated heparin-poloxamer thrombolysis study in vitro

Objective: To explore and discuss the mechanism of theapeutical ultrasound combined with heparin-poloxamer (HP) on thrombus dissolution in vitro. Methods: Blood clots were prepared from rabbit ear blood. Main factors on the thrombolysis effects in vitro were investigated, including blood clot age, ultrasonic intensity (pure intensity seted in the range of 2~8 W/cm2), ultrasound time and material concentration. Five experimental groups were as follows: ①US; ②US+MB; ③HP; ④US+HP; ⑤US+MB+HP. Through the results of the experiment, thrombolysis effect in each group was determined and the mechanism was discussed. Results: The group (US+MB+HP) showed the best result in the thrombolysis with less ultrasound time, though the other groups showed certain thrombosis effects in vitro. Conclusion: Heparin-poloxamer combined with ultrasound microbubble has an improved effect in thrombolysis, which may be a potential technique for clinical treatment of thrombotic diseases.

827 Investigation for distribution of the patient suffered with cognitive disorder and the compliance of those patients at memory clinic of neurology

Objective: To observe the distribution of the outpatients suffered with cognitive disorder at memory clinic of neurology in our hospital. To find the factors which effected on the compliance of those patients. Methods: Consecutive cases of outpatients complaining with cognitive disorder at neurology clinic were collected from Nov. 2012 to Jan. 2013, the different causes for cognitive disorder of patients were definited and the degree of severity was described.2. Prospective following-up: all the cases were followed up to 56±7 days after registration, the factors with effected on the compliance of the patients were collected, non-conditional logistic regressions analysis was adopted as statistical method. Results: Total 93 cases complaining with memory disorder were screened, 60 patients with cognitive disorder were chosen, among them there were 40 cases of Alzheimer’s disease (AD) type, with the percent of 66.67%, 7 cases with cognitive impairment of vascular type, 2 cases of frontotemporal dementia (FTD), and 11 cases of other causes. Account to the degree of the severity, there were 11 cases with mild cognitive impairment (MCI), the percent was 18.33% and the cases of middle-severe patient were 34, the  percent was 57%. By the study of the 8 possible factors, it showed that the degree of education was the independent factor (P=0.01, OR=2.3, 95%CI: 1.19~4.45) for the compliance of those patients. Conclusion: Cognitive impairment of AD type is the first cause of the patients with cognitive disorder at our memory clinic, middle-severe cognitive disorder patients take a high percent, the degree of education is the independent factor for compliance of the patients.

831 Correlation between sleep disorders and the level of adipose cytokines in maintenance hemodialysis patients

Objective: Look for the potential relationship between the level of adipose cytokines and sleep disorders in maintenance hemodialysis (MHD) patients, so as to explore the mechanism of sleep disorders affecting cardiovascular disease. Methods: Eighty-two cases of MHD patients with dialysis more than 3 months were chosen in our hemodialysis center from January 2013 to August 2014 as the study group, and were divided into two groups according to PSQI score (PSQI≤5 and PSQI>5). Urea, creatinine, albumin, CRP, leptin, interleukin-6 specimens were measured in two groups before hemodialysis. Statistical analysis was performed on two groups of patients with these indicators. Results: Sex and age in two groups had no significant difference (P>0.05). But CRP and leptin in sleep disorder groups were higher than the control group, there was statistical difference (P<0.05). Conclusion: Sleep disorder correlated to the level of adipose cytokines in hemodialysis patients.

834 The nosocomial septicemia in children: A 10-year analysis of etiology and morbidity changes

Objective: Investigate the clinical features and morbidity of nosocomial septicemia in children. Anlyse pathogenic bacteria and antibiotic resistance of these infections to improve the control and treatment. Method: A total of 68 patients of nosocomial septicemia in Yuying Children’s Hospital from January 2005 to December 2014 were enrolled in the retrospective study. Results: The infection rate of nosocomial septicemia (non-neonates) was 0.054% in Yuying Children’s Hospital during 10 years. Which was totally 0.091% durling 2005-2009, statistically significant higher than 0.035% durling 2010-2014 (x2=16.530, P=0.000). 70 strains of pathogens were isolated from blood in 10 years. In which gram-positive bacteria accounted for the highest proportion (35 strains, 50%), gram-negative bacterial was 42.9% (30 strains), pathogenic fungi was 7.1% (5 strains). The predominant three pathogens of G+ bacteria were coagulase negative staphylococci, oral streptococci and staphylococcus aureus. The predominant four pathogens of G- bacteria were escherichia coli, klebsiella pneumoniae, pseudomonas aeruginosa and Salmonella. Escherichia coli, klebsiella pneumoniae and pseudomonas aeruginosa were found to have multidrug resistance. Conclusion: The infection rate of nosocomial septicemia (non–neonates) was 0.054% in Yuying Children’s Hospital which Affiliated to Wenzhou Medical College during 2005-2014. This infection rate has a decrease trend during 2005-2014 (x2=16.530, P=0.000). Gram-positive bacteria were the major pathogens in nosocomial septicemia in children (non-neonates). There was no significant difference in pathogenic bacteria proportion between 2005-2009 and 2010-2014.

838 Clinical aspects of 40 cases of children with ovarian cysts

Objective: To analyze the diagnosis and treatment of ovarian cysts in children. Methods: A retrospective study of 40 children diagnosed with ovarian cysts from January 2012 to December 2014 was undertaken. All the patients are from The Second Affiliated Hospital & Yuying Children’s Hospital of Wenzhou Medical University. Results: The ages of the girls are varied from 26 days to 14 years old, with a mean age of 9 years old. 22 cases were on the left, 17 on the right and 1 on both sides. 38 cases accepted surgical intervention (20 cases were done by laparoscopy and 18 cases were done by laparotomy). One case without operation treatment for the cyst disappeared during hospitalization, another case underwent conservative treatment for the cyst was small and asymptomatic. 6 cases underwent adnexectomy for ovary necrosis, and another 1 case underwent this operation method for the fast pathologic diagnosis was dysgerminoma. Thirty-one cases underwent oophorocystectomy retenting of ovarian cyst. The pathological report mature cystic teratoma, serous cystadenoma, mucinous cystadenoma, mixed cystadenoma, endometrioid cyst. Conclusion: Clinical symptoms of ovarian cyst in children are not special, and its diagnosis and differential diagnosis rely on image examination. AFP and β-HCG detection in blood have great significance in evaluating the Ovarian cyst’s differentiation of benign and malignant. Whether the ovary can retain or not after pedicle torsion of ovarian cyst should depends on the early diagnosis, timely operation and the nature of the tumor. Laparoscopic surgery has its advantages in the treatment of benign ovarian cyst, but we should be careful to choose the operating method when it comes to a vicious one.

842 The effects of Power-lift Stirrups in radical resection of rectal carcinoma surgery

Objective: To obserre the effects of Power-lift Stirrups in radical resection of rectal carcinoma surgery. Methods: A total of 106 patients undergone radical resection of rectal surgery with continuous epidural anesthesia were randomly assigned to either control group or Power-lift Stirrups group. Patients in control group were placed in lithotomy position, while patients in Power-lift Stirrups group were positioned by the use of stirrups under the calf which nearly horizontal. The assessment of position setup time, surgeon satisfaction and incidence of postoperative complications were collected for comparison. Results: The results showed significant shorter position setup time in Power-lift Stirrups group (P<0.05), and surgeon satisfaction is 98%, which is significant better than that in control group as 66% (P<0.05). Moreover, significant effects for postoperative complications, including varicose veins of lower extremity (3 versus 0, P<0.05) and swelling of lower extremity (4 versus 0, P<0.05), were seen in control group and Power-lift Stirrups group, respectively. Conclusion: The use of Power-lift Stirrups in radical resection of rectal carcinoma surgery may shorten position setup time, improve the surgeon satisfaction and reduce the incidence of postoperative complications.

845 The phantom study of the potential of radiation dose reduction using adaptive iterative dose reduction algorithm

Objective: To study the ability of the reconstruction algorithm of adaptive iterative dose reduction 3D to reduce the radiation dose of CT scanning, under the prernise in image quality undamaged. Methods: A 320-detector CT with volume mode was used to scan the low contrast module CTP515 and the high resolution module CTP528 of the phantom Catphan 500, while seven different tube currents 50, 80, 150, 250, 350, 450, and 550 mA were adopted respectively. CT images were reconstructed using the reconstruction algorithm of Filtered Back Projection and AIDR 3D in three degrees: mild, standard and strong. The noise, contrast-noise ratio, signal-noise ratio and spatial resolution of the images were used to evaluate the quality of the output image yielded by different tube-current/reconstruction-algorithm choices. Results: Using the algorithm of AIDR 3D to reconstruct CT image can significantly reduce image noise and increase contrast-noise ratio and signal-noise ratio with the spatial resolution undamaged. In the best case, the proportion of the radiation could be reduced by 78.41%. Conclusion: The reconstruction algorithm of AIDR 3D has remarkable ability to reduce the radiation dose of CT scanning. Different iteration degrees resultes in different level of reduction.
