Construction of a 14-3-3γ expression vector and its effect on proliferation and apoptosis of uterine leiomyoma cells

Objective: To construct stably and highly expressing 14-3-3γgene eukaryotic expression plasmid vector and to explore its antitumor effects on uterine leiomyoma cells in vitro. Methods: The total RNA was isolated from the uterine leiomyoma cells. After reverse transcription reaction, 14-3-3γwas amplified by RT-PCR. The PCR product was cloned into the eukaryote expression vector pCMV-N-Flag, the resulted recombinant was designated as 14-3-3γ-pCMV-N-Flag. After sequence determination, the recombinant vector was transfected into uterine leiomyoma cells by lipofectin. The expression of the interesting protein was confirmed by Western Blot analysis. Cell proliferation was assessed by cell counting kit-8 assay and the percentage of apoptotic cells detected by annexinV-FITC/PI double-staining was determined by flow cytometry. Results: ①The 14-3-3γgene eukaryotic expression vector was successfully constructed and identified by sequencing. ②The expression level of 14-3-3γprotein was increased obviously after transfection (P<0.05). ③The inhibitory effect on the rate of cell proliferation was 52.90% after transfection (P<0.05). ④After transfection, the percentage of apoptotic cells was increased obviously (P<0.05). Conclusion: The 14-3-3γ-pCMV-N-Flag gene eukaryotic expression plasmid vector is successfully constructed which could suppress proliferation and induce apoptosis of uterine leiomyoma cells.

The influence of miR-34c on growth and apoptosis of type II endometrial carcinoma cell line HEC-1-B

Objective: To investigate the effect miR-34c on the growth and apoptosis of type II endometrial carcinoma cells (HEC-1-B cell line). Methods: Up-regulated the expression of miR-34c by transfecting cells with hsa-miR-34c mimics, the transfection efficiency was detected by flow cytometry and further verified by quantitative real-time PCR (qRT-PCR). Cell proliferation assay by CCK-8 and colony formation assay applied to demonstrate that miR-34c could inhibit the growth of HEC-1-B cells. Cells apoptosis assay was analyzed by flow cytometry. Results: The transfection efficiency was 81.16%. The overexpression of miR-34c (detected by qRT-PCR) could inhibit cell proliferation, colony formation and promote cells apoptosis (P<0.05). Conclusion: MiR-34c can inhibit the growth and promote apoptosis of HEC-1-B cells significantly, and function is a potential tumor suppressor in type II endometrial carcinoma.

Expressions of SDHB and PDK1 and their clinical significance in nasopharyngeal carcinoma

Objective: To investigate the expressions of SDHB and PDK1 and their correlation with the clinicopathological factors and prognosis in nasopharyngeal carcinoma (NPC). Methods: Immunohistochemstry was employed to analyze the expressions of SDHB and PDK1 in 78 specimens of NPC and 35 specimens of chronic nasopharyngitis tissues (CNT). Retsults: SDHB expression detected in 39.7% of the cancer tissues was significantly lower than those in 62.9% of nasopharyngitis tissues (x2=5.183, P<0.05). While PDK1 expression detected in 53.8% of the cancer tissues was significantly higher than those in 20.0% of nasopharyngitis tissues (x2=11.269, P<0.05). SDHB and PDK1 expressions were significantly associated with lymph node metastasis,tumor recurrence, cranial nerve palsy. however, there was no significant association between the expression and gender, age. Kaplan-Meter survival analysis showed that the survival rates of positive SDHB expression and negative PDK1 expression were higher than that of negative SDHB expression and positive PDK1 expression.Multivariate Cox regression analysis showed that the development of NPC, lymph node metastasis and recurrence were independent prognostic factors for NPC, but the expressions of SDHB and PDK1 were not an independent factor. Conclusion: The expressions of SDHB and PDK1 play an important role in the development of NPC, lymph node metastasis and recurrence. SDHB and PDK1 expressions may have an impact on prognosis, but it was not independent factor affecting the prognosis in pafients with NPC.

Effects of ERK1/2 on pneumocyte apoptosis after lung ischemia/reperfusion injury and ischemic postconditioning intervention

Objective: To investigate the role of ERK1/2 on pneumocyte apoptosis after lung ischemia/reperfusion injury and ischemic postconditioning (IPO) intervention. Methods: Forty adult male Sprague-Dawley rats were randomly divided into 5 groups based upon the intervention (n=8): control group (C), IR group (IR), IR+IPO group (IPO), IPO+solution countrol group (D), IPO+U0126 group (U). Left lung tissue was isolated after the 2 hours of reperfusion, The ultrastructure of the left lung were observed under electron transmission microscopes. Apoptosis index (AI) of lung tissue was determined by terminal deoxynuleotidyl transferase mediated dUTP nick end and labeling (TUNEL) method. The mRNA expression and protein levels of Bcl-2 and Bax were measured by RT-PCR and quantitative immunohistochemistry (IHC). Results: Compared with C group, AI and the expression of Bax of IR were significantly increased, the expression of Bcl-2 and Bcl-2/Bax were significantly decreased (P< 0.05), and ultrastructure abnormality was obviously found in lung tissue. Compared with IR group, all the indexes of IPO except for the expression of Bcl-2 and Bcl-2/Bax were obviously reduced, the expression of Bcl-2 and Bcl-2/Bax was increased (P<0.05). All the indexes between D and IPO were little or no significant (P>0.05). the expression of Bcl-2 and Bcl-2/Bax of U was significantly decreased and other indexes were increased than those of IPO (P<0.05). Conclusion: IPO may attenuate pneumocyte apoptosis in LIRI by activation of ERK1/2 MAPK, up-regulating expression of Bcl-2/Bax ratio.

The association of adenosine A1 receptor gene variants with the multidrug-resistant partial seizure epilepsy

Objective: To explore the association of the single-nucleotide polymorphisms (SNP) of adenosine A1 receptor (A1AR) gene with partial pharmacoresistance seizure in south of Zhejiang province Han population. Methods: One hundred and twenty cases of partial seizure patients were obtained for this study, multidrug-resistant group and full controlled with no seizures group (followed: named the none pharmacoresistance group) each 60 cases, and 60 healthy patients as normal control group. A polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) was used for the detection of rs903361, rs10920573, rs3766553 SNP, via the retrospective case-control study and multiple logistic regression analysis methods to explore its correlation of drug resistance epilepsy. Rusults: Both the frequencies of the allelic of all SNPs among the three group and genotypes of three SNPs between the normal control and none pharmacoresistance group had no statistical difference. The frequency of rs903361 CT genotype in pharmacoresistance epilepsy patients was obviously lower than in the none pharmacoresistance group, P=0.017. The frequency of rs10920573 CT genotypes in pharmacoresistance epilepsy patients was obviously higher than in the none pharmacoresistance group, P=0.010. According to the multiple logistic regression analysis, the CC and TT genotypes in rs903361 were both the independent risk factor for pharmcoresistance in epileptic patients and associated with a 2.73 and 2.609 times added risk for pharmacoresistance than CT genotype, P=0.043 and 0.047 respectively. In rs10920573, CT genotype was also an independent risk factor for pharmcoresistance in epileptic patients,and associated with a 2.572 times increased risk for pharmacoresistance than TT genotype, P=0.037. There was no difference among all rs3766553 genotypes.Conclusion: The CC and TT genotypes of rs903361 and the CT of rs10920573 are both the independent risk factors associated with mulltidrug-resistant partial seizure epilepsy.

Expression of miR-195 in plasma of patients with breast cancer and its clinical significance

Objective: To explore the expression of miR-195 in plasma of patients with breast cancer in pre-operative and postoperative and two weeks and it correlation with clinicopathologic features of breast cancer. Methods: By normalized to miR-16, the expressions of miR-195 in 48 invasive ductal carcinoma of the breast cancer at preoperative and postoperative 2 weeks and 35 healthy controls were examined by stem-loop real-time RT-qPCR method. The relationship between miR-195 expression and clinicopathological characteristics were further analyzed. Results: Expression of miR-195 in plasma of patients with breast cancer was obviously higher than that in the healthy controls (P<0.05). At 2 weeks postopratively, a significant decrease of plasma miR-195 was observed, reaching levels of the healthy control subjects. ROC curve showed plasma levels of miR-195 could detect individuals with breast cancer with 94.4% sensitivity and 68.8% specificity. Correlative analysis of clinical features showed that there was no significant association between plasma miR-195 expression and the size of tumor, the state of ER, PR, lymph node metastasis and Her-2 receptor of breast cancer. Conclusion: Aberrant expression of miR-195 in plasma of breast cancer patients has potential use as novel noninvasive biomarkers for breast cancer.

Study on the relationship between maternal hemoglobin concentration and the risk of gestational diabetes mellitus

Objective: To explore the relationship between maternal hemoglobin concentration (Hb) in early pregnancy and the incidence of gestational diabetes mellitus (GDM). Methods: In a case-controlled study, the clinical date of 109 women with GDM (GDM group) and 151 healthy pregnant women (control group) were analyzed retrospectively. Results: The univariate analysis revealed that the age, body mass index (BMI) before pregnancy, high Hb concentration at early pregnancy, systolic pressure and diastolic pressure at early pregnancy, educational level, gravidity and parity in the GDM group were higher than that in the control group (P<0.05). The subjects were divided into 4 quartiles in order of increasing Hb concentration level, among which Hb concentration and the incidence of GDM were compared. With the Hb concentration increasing the risk of GDM increased. A positive correlation was found between Hb concentration and the incidence of GDM by the spearman rank correlation (r=0.201, P=0.001). Multivariate logistic regression analysis revealed that, with adjustment for the confounding factor above-mentioned, high Hb concentration (Hb≥130 g/L) at early pregnancy had an increased risk of GDM (OR=3.54, 95%CI: 1.60~7.83, P=0.002). Conclusion: High Hb concentration at early pregnancy increases the incidence of GDM, Hb examining can be effective measure of screening for GDM.

The prevalence and dangerous factors of early childhood caries(ECC) among 5-year-old children in Wenzhou

Objective: To investigate the prevalence and dangerous factors of early childhood caries (ECC) among 5-year-old children in Wenzhou, and provide the scientific foundation for preventing deciduous dental caries. Methods: The sampling method was cluster and random in this study. A total of 693 children aged 5 were selected in the survey, caries check was carried out by a dentist reference WHO caries disease diagnosis criteria, and the questionnaire were completed by the parents. The related factors of dental ECC was analyzed by chi-square test and logistic regression. Results: The prevalence of ECC was 72.72%, and the filling rate was only 14.88%. The mean dmft and dmfs were 4.35 and 10.75 respectively. The key factors for ECC included: family economic income, oral health knowledge of parents, frequency of consumption of sweets, eating before sleep, sleeping with feeding-bottle and whether parents supervised or helped to brush or not. Backwards logistic regression was performed, variables remained in the final model were: frequency of consumption of sweets, eating before sleep, parents?supervising or helping brush or not, sleeping with feeding-bottle or not and oral health knowledge of parents. Conclusion: The prevalence of ECC is high among 5 years old children in Wenzhou. To strength parents?oral health knowledge, limiting amount of eating sugar, and culturing scientific oral hygiene habits are very important for preventing ECC.

Expression and significance of ecdysoneless in gastric cancer

Objective: To probe the ecdysoneless (ECD) expression and its correlation with clinical variables, such as clinical pathologic factors and prognosis in gastric cancer. Methods: ECD mRNA expression level in gastric cancer samples and matched normal gastric tissue from 56 patients was detected using Real-time PCR method. Immunohistochemical method was adopted to deteced the protein expression level of ECD in gastric cancer tiusse which with 5 years follow up. Results: ECD expression was up-regulated in gastric cancer tiusse compared with matched normal gastric tissue. Positive ECD immunoreactivity was seen in 54.0% of gastric cancer specimens, which was correlated with Lauren typing, lymph node metastasis, vessel invasion, distant metastasis, and TNM stage (P<0.05). Compared with ECD negative patients, ECD positive patients had significantly shorter survival times (35.8±1.9 month vs 45.2±1.9 month, P<0.01) and short survival rate (24.1% vs 50.0%, P<0.01). Multiariable COX regression analysis indicated that Lauren typing and TNM stage could serve as a dependent prognsis factor of gastric cancer (P<0.01), except ECD expression (P=0.212). Conclusion: ECD expression is increased in gastric cancer tissue. The ECD expression correlates with Lauren typing, lymph node metastasis, vessel invasion, distant metastasis, and TNM stage and poor survival rate in gastric cancer.

The diagnostic value of multi-slice helical CT and image post-progressing technique in laryngeal and hypopharyngeal carcinoma

Objective: To evaluate the diagnostic value of multi-slice helical CT and post-processing technique of two dimension MPR (multi-planar reconstruction), three-dimensional CTVE (CT virtual endoscopy) and three-dimensional transparent VRT (volume rendering technique) for the extent of regional invasion of laryngeal and hypopharyngeal carcinoma. Methods: Fifty-one patients with the laryngeal and hypopharyngeal carcinoma were prospectively examined with 16-slice helical CT, then MPR, CTVE, VRT images were reconstructed. The axial images, MPR and the combined image were respectively compared with the pathological results, and the CTVE, VRT image compared with electronic laryngoscope results. These results were studied in a blind way. Results: There were 31 patients with laryngeal carcinoma and 20 patients with hypopharyngeal carcinoma. The results were basic identical (P>0.05) when assessing the epiglottis, ventricular bands, aryepiglottic fold, vocal cord, anterior commissure, subglottic region, paralaryngeal space, sinus piriformis, cartilage laryngeal and lymph node between the axial, MPR, the combined image and pathological finding of operation. The results were also basic identical (P>0.05) when assessing the epiglottis, ventricular bands, aryepiglottic fold, vocal cord, anterior commissure, sinus piriformis between the CTVE image and electronic laryngoscope and assessing the sinus piriformis, ventricle of larynx and distortion of airway between the CTVE, VRT image and electronic laryngoscope. Conclusion: The multi-slice helical CT and post-processing technique can display the position, the shape, the size, the affected extent and metastasis of lymph node comprehensively, especially in the affected extent and metastasis of lymph node. The CTVE is simultaneously good complementary methods to conventional electronic laryngoscope.

Screening metastasis related peptides binding to the surface of ovarian tumor cells by phage display

Objective: Using 7-mer phage display peptide library in vitro to isolate metastasis related peptides on the surface of the high metastatic potential ovarian tumor cell line HO-8910PM. Methods: Phage display technology with biopanning and rapid analysis of selective interactive ligands between cell lines HO-8910 and HO-8910PM was utilized. After five rounds of biopanning, 14 phage clones were randomly selected for sequencing of DNA. Using ELISA and phage competitive binding experiment the affinity of positive phages combined with HO-8910PM cells was identified. Results: The phage clone named Z3 (peptide LRLRNTR) showed higher affinity to HO-8910PM cells. Conclusion: The peptide LRLRNTR screened by phage display technology may offer a new direction for early diagnosis, metastasis and treatment of ovarian tumor.

RhoA gene expression and clinicopathology parameters in pancreatic carcinoma

Objective: To investigate the expression of RhoA gene in tissues of pancreatic cancer and to discuss its significance during carcinogenesis and progression of tumor. Methods: Intratumor RhoA expression level was determined and compared with that in adjacent nontumorous pancreatic tissues using quantitative real time polymerase chain reaction and Western blot in 48 samples of pancreatic carcinoma. Results: The mRNA levels of RhoA were significantly higher in tumor tissues than that in the unaffected portions (t=3.669, P=0.0007). The expression of RhoA mRNA in the primary lesion was higher in patients with low and middle differentiation cancer (t=2.447, P=0.006), protal vein invasion (t=2.544, P=0.008), lymph node metastasis (t=2.351, P=0.041), and advanced pTNM stage (stage III/IV, t=2.667, P=0.025) than in those without. There was a significant difference between high RhoA protein levels in the tumor tissues and noncancerous tissues in pancreatic carcinoma patients(t=3.461, P=0.0003), the quantity of RhoA expression associated with tumor invasion and metastasis. There was a significant association between high tumor RhoA protein levels and low and middle differentiation cancer (t=2.323, P=0.016), and the presence of protal vein invasion (t=2.119, P=0.036), lymph node metastasis(t=2.117, P=0.029), and advanced pTNM stage (t=2.776, P=0.004). Conclusion: There is a significant correlation among RhoA expression, tumor stage, and metastasis. The expression of RhoA can be used as a good tumor marker for invasive and advanced pancreatic carcinoma as well as a prognosis predictor.

Study on the expression and its clinical significance of 53BP1 in cervical carcinoma

Objective: To explore the expression of P53-binding protein 1 (53BP1) in the tissue of cervical squamous carcinoma and relationship among their clinical stages, the degree of differentiation, lesion of lymph node metastasis and size. Methods: Fifty-two cases of cervical squamous carcinoma, 30 cases of cervical intraepithelial neoplasia (CIN) and 20 cases of normal cervical tissue were selected. Then the real-time fluorescent quantitative PCR was adopted for the detection of 53BP1 mRNA expression, and the statistical analysis of its expression and its clinical pathological characteristics were complemented as well. Results: 53BP1 mRNA in cervical squamous carcinoma relative expression levels were significantly lower than that in normal tissue, and the difference was statistically significant (P<0.05). 53BP1 mRNA expression in the differentiation of cervical squamous carcinoma in higher group level was significantly higher than that in the group of poorly differentiated carcinoma of the cervix, and the difference was statistically significant (P<0.05). 53BP1 mRNA in cervical lymph node metastasis-free group relative expression levels were significantly higher than that in the cervical lymph node metastasis group, and the differences were statistically significant (P<0.05). Compared the expression levels in FIGO I group of cervical cancer and cervical carcinoma FIGO II group, and focus-group and small lesions 53BP1 mRNA, the difference was not statistically significant (P>0.05). Conclusion: 53BP1 expression in cervical squamous cancer reduction is associated with the degree of differentiation, lymphatic metastasis, and lesion size, while it is not significantly related in clinical stages.

Anterior approach preperitoneal groin hernioplasty in inguinal hernias (A report of 256 cases)

Objective: To evaluate the effect of open preperitoneal groin hernioplasty in inguinal hernia. Methods: A retrospcetive study was made to analyze the experience of performing anterior approach preperitoneal groin hernioplasty for 256 patients between January 2007 to June 2012. Results: There were 148 typeII,139 typeIII,15 type IV hernias in the series. The average operation time was 58 min. 189 patients were followed up for 1~37months, the rate of follow-up was 73.8%. Hydrocele was fond in 26 patients, there were 12 patients of chronic pain in the inguinal area, and 2 patients of recurrence. None of infection, atrophy of testis occurred. Conclusion: The Anterior approach preperitoneal tension-free hernioplasty is advantageous in low tension, short operation time, less postoperative pain, fast recovery and few complications. It is an ideal procedure for patients with inguinal hernia.

Clinical and pathological analysis of malignant struma ovarii

Objective: To analyze the clinical pathological features of malignant struma ovarii (MSO). Methods: The clinical data, histological and immumohistochemical finding of 5 cases of MSO was researched with review of literature. Results: Pelvic mass was showed in all the 5 cases, without any clinical evidence of malignancy or metastasis. Morphologically, MSO was found to arrise from teratoma, sometime with transition from benign to malignant, immunohistochemistry showed TTF-1 and TG positive. Conclusion: As a rare tumor, MSO, whose histopathological diagnostic criteria is similar to that of the thyroid primary malignant lesion, is lack of specific clinical symptoms and may be constituent of teratoma. It is divided into papillary and follicular types in histology. It has a good prognosis.

Evaluation of coronary artery and left ventricular function with 64-slice computed tomography

Objective: To evaluate the feasibility and accuracy of coronary angiography and left ventricular function analysis evaluated by 64-slice computed tomography simultaneously. Methods: Fifty consecutive patients suspected or diagnosed with coronary disease were examined by retrospective electrocardiogating(ECG) cardiac CT scan for coronary artery angiography and left ventricular functional analysis. Coronary artery was divided into 16 segments according to the American Heart Association. The image quality was graded with 4-points scale. The LVEF (left ventricular eject function) was calculated from cardiac CT date and compared with ultrasonic cardiogram by pearson correlation. Results: Forty-three patients accomplished CT examination successfully, in whom, thirty-five cases finished ultrasonic cardiogram also. In 508 evaluated coronary segments, 91.93% could be diagnosed. The LVEF correlation of CT and ultrasonic cardiogram was high (r=0.64). Conclusion: The 64-slice CT can be used in coronary angiography and left ventricular function analysis simultaneously with high imaging quality of coronary artery and reliable analysis of left ventricular function.

Preparation and quality evaluation of rapamycin suppository

Objective: To prepare rapamycin suppository, and to evaluate its quality. Methods: Rapamycin was used as active ingredient, polyethylene glycol 2000 and polyethylene glycol 4000 were used as main substrates. The suppositories were made using hot melt process method, and the drug content, weight difference, melting time, drug release with time were evaluated. Results: Polyethylene glycol suppository with rapamycin appeared white and translucent. Good hardness could be maintained at room temperature. The drug content, weight variation and melting time limit matched the requirements for the suppository. More than 75% of the loaded drug in the suppositories was released at about 25 min in the medium. Conclusion: This preparation of rapamycin polyethylene glycol (PEG) suppository can reach the quality requirements of Chinese pharmacopoeia, and may be developed as a new formulation of rapamycin.
