Abstract Objective: To investigate feasibility of bone marrow mesenchymal stem cells transplantation in uterine scar reparation. Methods: Animal model of uterine scar was constructed in female SD rats. MSCs marked with Ad-GFP was injected into the site of uterine scar. Muscle tissue was obtained after two weeks of cell transplantation for HE staining. The directional differentiation situation of WMCs at the incisional part was detected with VEGF, TGF-β1 immunohistochemistry, and immunofluorescence staining for α-SMA, and uterine scar bursting pressures test. Results: Animal model of uterine scar was successfully constructed. MSCs transplantation upregulate expression of VEGF, and downregulate TGF-β1 (both P<0.01). With the aid of α-SMA immunofluorescence, transplanted MSCs demonstrated characteristic of immunotype of smooth muscle cells differentiaton. MSCs transplantation increased bursting pressures significantly (P<0.05). Conclusion: These results provide the evidence that MSCs transplantation can be used to promote regenerative reparation of uterine scar, reduce the risk of uterine scar dehiscence in the following pregnancy.
