Abstract Objective: To analyze lncRNA expression profiles in lung squamous cell carcinoma and prelimiary validate. Methods: LncRNAs and mRNAs expression profiles in lung squamous cell carcinoma samples compared with NT samples were studied by high-throughput microarray. The interesting candidate lncRNAs were verified by real-time quantitative PCR. Results: Using abundant and varied probes, 30 586 lncRNAs and 26 109 coding transcripts were able to assess in our microarray. From the data we found there were thousands of lncRNAs and mRNAs that differentially expressed ($2 fold-change) and 1 741 lncRNAs and 1 716 mRNAs differentially expressed in lung squamous cell carcinoma samples compared with NT. P53, cell cycle, streptococcal infection, cell adhesion molecules signaling pathways may be involved in the pathogenesis of the lung squamous cell carcinoma. We also found, via quantitative PCR, that 10 lncRNAs were aberrantly expressed in lung squamous cell carcinoma compared with matched histologically normal lung tissues. Among these, KRT16P2 and BC016831 were the most aberrantly expressed lncRNAs, as estimated by quantitative PCR in 40 pairs of lung squamous cell carcinoma and NT samples. Conclusion: The results reveal lncRNAs expression patterns in lung squamous cell carcinoma by microarray. These lncRNAs and some pathways may play a key role in tumor development and occurrence.
