Abstract The Hippo pathway, comprising MST, SAV1, MOB and LATS kinases, is commonly disrupted in aggressive cancers. Dysregulation of Hippo pathway activation drives tumorigenesis, chemoresistance, and metastasis. Post-translational modifications (PTMs) such as ubiquitylation, SUMOylation, acetylation, play a critical role in regulating the activation, deactivation, and subcellular localization of signaling proteins. PTMs also facilitate signal initiation, amplification, and transduction. Emerging evidence indicates that several key signaling molecules in the Hippo pathway are tightly regulated by PTMs. This review summarizes the current understanding of PTMs’ effects on Hippo signaling and discusses potential therapeutic interventions based on PTMs and the Hippo pathway.
