Expression of hsa_circ_0001322 in gastric cancer and it’s clinical significance |

Abstract Objective: To analyze the expression change and diagnostic value of a circular RNA, hsa_circ_0001322 in gastric cancer. Methods: Gastric cancer mucosa tissues and paired normal gastric mucosa tissues of 54 patients were collected in the First Affiliated Hospital of Wenzhou Medical University from October 2019 to April 2020. The expression level of hsa_circ_0001322 was examined using real-time quantitative polymerase chain reaction (RT-qPCR) and its correlation with different clinicopathological factor groups were analyzed. Results: The expression level of hsa_circ_0001322 in gastric cancer tissues was lower than that in normal gastric mucosa tissues (0.309 vs. 1.000, P<0.05). The lower expression of hsa_circ_0001322 was also significantly associated with lymph node (LN) metastasis (0.208 vs. 0.391, P<0.05). Partial correlation analysis showed that the expression level of hsa_circ_0001322 was negatively correlated with LN metastasis of gastric cancer (R=-0.587, P<0.05), after excluding the influence of age, gender, tumor size, tumor location, depth of invasion or cell differentiation. The area under the ROC curve of hsa_circ_0001322 was 0.808, and the sensitivity and specificity of prediction with threshold value at 0.254 was 90.0% and 75%, respectively. Conclusion: Down-regulation of hsa_circ_0001322 was correlated with gastric cancer tissue and lower expression of hsa_circ_0001322 was evidently associated with lymph node metastasis, which suggested that hsa_circ_0001322 may serve as a potential biomarker for diagnosis of gastric cancer and prediction of lymph node metastasis.
