Abstract Objective: To study the expression of (PBX3) and Twist mRNA and protein in thyroid papillary carcinoma (PTC), and to explore their clinical significance. Methods: The expression of PBX3 and twist in paraffin embedded tissues was detected by immunohistochemical En Vision two-step method, and the expression of PBX3 and Twist mRNA in fresh tissues was detected by real-time fluorescence quantitative PCR. The expression of PBX3 and Twist mRNA and protein in PTC and its relationship with different clinicopathological characteristics and was analyzed by statistical software. The relationship between the expression of PBX3 and Twist protein was Panalyzed by Spearman rank correlation analysis. Results: The expression level of PBX3 and Twist mRNA in PTC was higher than that in paracancerous tissue (P<0.05), and the expression rate of PBX3 and Twist protein in PTC was 73.33% (44/60) and 68.33% (41/60), respectively. The expression of PBX3 and Twist protein in cancer tissues was significantly higher than that in adjacent tissues. The expression rate of PBX3 and Twist protein in paracancerous tissue was 16.67% (5/30) and 13.33% (4/30), respectively (P<0.05). The expression of PBX3 mRNA and protein in PTC tissues with capsule invasion, lymph node metastasis and stage III-IV were higher than those in PTC tissues without capsule invasion, lymph node metastasis and stage I-II (P<0.05). The expression of Twist mRNA and protein in PTC tissues was different (P<0.05) in the presence or absence of capsule invasion,lymph node metastasis, and at stage I to II or stage III to IV. There was a significantly positive correlation between PBX3 and Twist protein expression in PTC (r=0.400, P<0.05). Conclusion: The expression of PBX3 and Twist mRNA and protein is up-regulated in PTC tissue. The abnormal high expression of PBX3 and Twist is closely related to the occurrence, invasion and metastasis of PTC. The expression of PBX3 is positively correlated to the expression of Twist, indicating that the expression of PBX3 and Twist may play a synergistic role in the development of PTC.
