Abstract Objective: To determine the changes of mmu_circ_42398 expression in the liver tissue from a murine model of liver fibrosis and cell culture models that reproduce liver disease, and make an initial exploration into its biological function. Methods: A mouse model of liver fibrosis was prepared and three murine cell culture models were reproduced: a hepatic stellate cell (JS1) activation model, a hepatocyte (AML12) oxidative stress injury model, and a macrophage inflammation (RAW264.7) model. A normal control group was also established.qPCR was used to measure the expression of mmu_circ_42398 in the liver tissue and in cells from the above cell culture models, and bioinformatic analysis was used to predict the circRNA target gene, to analyze its biological function, and to map its competing endogenous RNA (ceRNA) signaling pathway. The mmu_circ_42398 over expression vector was constructed and produced and then transfected into JS1 cells to examine the protein expression of a-SMA, ColI. Results: Compared with the control group, the expression of mmu_circ_42398was significantly decreased in liver tissue from the mouse liver fibrosis model (P<0.05), and its expression was significantly decreased in all three cell culture models. Bioinformatic analysis predicted that mmu_circ_42398could affect the activation of hepatic stellate cells by binding to the target genes miR-338-3p and miR-344-3p.The over expression of mmu_circ_42398 in JS1 cells resulted in decreased of protein expression of a-SMA, ColI.Conclusion: The expression of mmu_circ_42398 is significantly changed in the liver fibrosis model and may be
involved in the activation of hepatic stellate cells.
