Abstract Objective: To optimize the preparation technology of trauma swelling elimination and painkiller ointment and to determine the content of its active components. Methods: An orthogonal experiment was designed to optimize the ethanol extraction process. The single factor method was used to optimize the preparation process. The content of emodin and polydatin was determined by HPLC. Results: The best alcohol extraction process was to extract 7 times the amount of 70% ethanol three times, 2 hours each. The viscosity of the paste was moderate when the extraction was 3 times the amount and the ratio of gelatin to Arabic gum was 1:4. The content of emodin and polydatin was 4.14 μg·g-1 and 12.71 μg·g-1. Conclusion: The optimized process is stable and feasible, and thus suitable for the preparation of the trauma swelling elimination and painkiller ointment. The index components of emodin and polydatin were stable and feasible, which can be used for quality control.
