Abstract:Objective: To investigate the therapeutic effect of miR-143/145 on endometriosis cysts in SD rat models. Methods: The model of endometriosis in SD rats was established by autologous transplantation, and was identified at the histopathological level. Eighteen SD rats were randomly divided into blank control group (n=6), negative control group (n=6) and miR-143/145 mimic group (n=6), were given the corresponding treatment, respectively. Finally laparotomy, and the size of ectopic cysts were measured by vernier caliper, and the volume of ectopic cysts were compared among the three groups. The histopathological changes of the three groups were also compared. Results: The successful rate of modeling of endometriosis in SD rats was 84%. After treatment, the difference in the volume of ectopic cysts in each group: the volume of ectopic cysts in control group was (16.38±4.19) mm3, negative control group was (21.14±0.51) mm3, miR-143/145 mimic group was (5.91±1.91) mm3. There was no significant difference between the blank control group and the negative control group (P>0.05), however, the volume of ectopic cysts in the miR-143/145 mimic group was significantly reduced compared with negative control group (P<0.01). The histopathological changes: the histopathological morphology of ectopic cyst in blank control group and negative control group was similar, but in miR-143/145 mimic group the number of glands was decreased, the glandular atrophy, and the arrangement of stromal cell were messy compared with the mimic NC group. Conclusion: MiR-143/145 can atrophy ectopic cysts of endometriosis and achieve therapeutic effect.
