Abstract:The extracellular adenosine metabolism system, which is involved in numerous and varied physiological and pathological processes, has drawn wide interest as targets for new drug development.However, the research progress of drug development targeting ecto-nucleotide pyrophosphatase/phosphodiesterase-1 (ENPP1) is rare. Recently, the roles of ENPP1 have received progressive attention after the breakthrough finding as the dominant 2’3’-cyclic GMP-AMP dinucleotides-hydrolase. Here, having briefly introduced the structure of ENPP1, we focus on its physiological and pathological functions in biomineralization and tumor, as well as the research and development of enzyme replacement therapy and small molecule drugs targeting ENPP1.
