Abstract:Objective: To investigate the effect and mechanism of cryptotanshinone on the chemosensitivity of QBC939 cells to gemcitabine. Methods: The human cholangiocarcinoma QBC939 cells were divided into four groups: control group, cryptotanshinone group, gemcitabine group and cryptotanshinone+gemcitabine group.
CCK-8 method was used to detect the inhibition rate of cell growth. Apoptosis was detected by flow cytometry.Western blot was used to detect the expression of apoptosis related protein. Results: After 24 h treatment, when the final concentration of cryptotanshinone was 10, 30 and 50 μmol/L, the inhibition rate of cell growth was
22.30%±2.15%, 42.53%±3.62% and 51.85%±4.81% respectively, which was lower than that of the control group(99.22%±3.52%), with statistical difference (P<0.05). After 24 h treatment, the apoptosis rate of cryptotanshinone+gemcitabine combination group was 35.40%±1.57%, which was higher than the cryptotanshinone group
(19.80%±0.92%), the gemcitabine group (24.20%±0.56%) and the control group (8.84%±0.58%). The difference was statistically significant (P<0.05). Western blot showed that the expression of active caspase-3 and Bax protein in the combination of cryptotanshinone and gemcitabine group was higher than that in gemcitabine group,cryptotanshinone group and control group. Conclusion: Cryptotanshinone can increase the chemosensitivity of gemcitabine in cholangiocarcinoma cells.
