Abstract:Objective: To investigate the expression of estrogen receptor-β in nucleus pulposus cells of the human intervertebral disc. Methods: Seven nucleus pulposus specimens were obtained from 7 patients during surgery. Cultured disc cells were used for immunocytochemical localization of ERβ. mRNA was isolated from cultured human disc cells, and RT-PCR amplification of ERβ was employed to document molecular expression of this receptor. Results: Expression and localization of estrogen receptor-β were detected in all 7 specimens. A goldview-stained agarose gel electrophoresis showing gene expression of ERβ as a single band at the predicted 323 bp size. Localization was seen in the cytoplasm in all cultured disc cells with the immunocytochemistry method. Conclusion: This study demonstrates the expression of estrogen receptor-β in nucleus pulposus cells of the human intervertebral disc
